IL-21 Regulates the Differentiation of a Human γδ T Cell Subset Equipped with B Cell Helper Activity
2012
Vγ9Vδ2 T lymphocytes recognize nonpeptidic antigens without presentation by MHC molecules and display pleiotropic features. Here we report that coculture of Vγ9Vδ2 cells with phosphoantigen and IL-21 leads to selective expression of the transcription repressor Bcl-6 and polarization toward a lymphocyte subset displaying features of follicular B-helper T (T(FH)) cells. T(FH) like Vγ9Vδ2 cells have a predominant central memory (CD27(+)CD45RA(-)) phenotype and express ICOS, CD40L and CXCR5. Upon antigen activation, they secrete IL-4, IL-10 and CXCL13, and provide B-cell help for antibody production in vitro. Our findings delineate a subset of human Vγ9Vδ2 lymphocytes, which, upon interaction w…
Optimising experimental research in respiratory diseases: an ERS statement
2018
Experimental models are critical for the understanding of lung health and disease and are indispensable for drug development. However, the pathogenetic and clinical relevance of the models is often unclear. Further, the use of animals in biomedical research is controversial from an ethical perspective. The objective of this task force was to issue a statement with research recommendations about lung disease models by facilitating in-depth discussions between respiratory scientists, and to provide an overview of the literature on the available models. Focus was put on their specific benefits and limitations. This will result in more efficient use of resources and greater reduction in the numb…
Protection by extra virgin olive oil against oxidative stress in vitro and in vivo. Chemical and biological studies on the health benefits due to a m…
2017
Abstract We report the results of in vivo studies in Caenorhabditis elegans nematodes in which addition of extra virgin olive oil (EVOO) to their diet significantly increased their life span with respect to the control group. Furthermore, when nematodes were exposed to the pesticide paraquat, they started to die after two days, but after the addition of EVOO to their diet, both survival percentage and lifespans of paraquat-exposed nematodes increased. Since paraquat is associated with superoxide radical production, a test for scavenging this radical was performed using cyclovoltammetry and the EVOO efficiently scavenged the superoxide. Thus, a linear correlation (y = -0.0838x +19.73, regres…
Diffusion Acceleration in Randomly Switching Sawtooth Potential
2005
We investigate an overdamped Brownian motion in symmetric sawtooth periodic potential switched by Markovian dichotomous noise between two configurations. The two configurations differ each other by a translation of half of period. The calculation of the effective diffusion coefficient is reduced to the mean first‐passage time problem, and we obtain the exact expression valid for arbitrary mean rate of switchings and arbitrary intensity of white Gaussian noise. We find the area at parameters plane where acceleration of diffusion in comparison with the free diffusion case takes place.
Antibacterial and white spot lesions preventive effect of an orthodontic resin modified with silver-nanoparticles
2021
Background To evaluate the antibacterial property of a modified orthodontic resin with different concentrations of silver-nanoparticles (AgNPs), and quantify its preventive effect on the formation of white spot lesions (WSLs). Material and Methods An orthodontic resin (Transbond XT) was modified with four concentrations of AgNPs (1%, 0.5%, 0.1%, and 0.05%), the orthodontic resin without AgNPs was used as control. Polymerized resin discs (n=80) were submitted to Agar diffusion test on Petri dishes inoculated with Streptococcus mutans and Lactobacillus acidophilus. In addition, resin discs of each group (n=40) were placed in 96-well plates with bacterial suspensions to evaluate the colony-for…
Effect of an orthodontic resin modified with silver-nanoparticles on enamel color change
2021
Background: To evaluate the effect of an orthodontic resin modified with various concentrations of silver-nanoparticles (AgNPs) on enamel color. Material and Methods: Twenty lower premolars were collected and divided into four groups (n = 5) according to the concentration of AgNPs (0.05%, 0.1%, 0.5%, and 1% wt/wt). Additionally, a group bonded with a conventional resin was used as control (n=5). Previous to the bracket bonding, enamel color was measuring with a spectrophotometer. Specimens were stored for 6 months in deionized water. Brackets were debonded and color changes of enamel were calculated with the CIEDE 2000 formula (ΔE00). One-way ANOVA and Tukey test were used to assess the col…
Pga13 in Candida albicans is localized in the cell wall and influences cell surface properties, morphogenesis and virulence.
2011
The fungal cell wall is an essential organelle required for maintaining cell integrity and also plays an important role in the primary interactions between pathogenic fungi and their hosts. PGA13 encodes a GPI protein in the human pathogen Candida albicans, which is highly up-regulated during cell wall regeneration in protoplasts. The Pga13 protein contains a unique tandem repeat, which is present five times and is characterized by conserved spacing between the four cysteine residues. Furthermore, the mature protein contains 38% serine and threonine residues, and therefore probably is a highly glycosylated cell wall protein. Consistent with this, a chimeric Pga13-V5 protein could be localiz…
Identification of a mannoprotein present in the inner layer of the cell wall of Saccharomyces cerevisiae.
1997
Cell wall extracts from the double-mutant mnn1 mnn9 strain were used as the immunogen to obtain a monoclonal antibody (MAb), SAC A6, that recognizes a specific mannoprotein--which we have named Icwp--in the walls of cells of Saccharomyces cerevisiae. Icwp runs as a polydisperse band of over 180 kDa in sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis of Zymolyase extracts of cell walls, although an analysis of the secretory pattern of the mannoprotein shows that at the level of secretory vesicles, it behaves like a discrete band of 140 kDa. Immunofluorescence analysis with the MAb showed that Icwp lies at the inner layer of the cell wall, being accessible to the antibody on…
The Antioxidant Potential of White Wines Relies on the Chemistry of Sulfur-Containing Compounds: An Optimized DPPH Assay
2019
The DPPH (2,2-Diphenyl-1-picrylhydrazyl) assay is an easy and efficient method commonly used to determine the antioxidant capacity of many food matrices and beverages. In contrast with red wines, white wines are poorer in antioxidant polyphenolics, and the more hydrophilic sulfur-containing compounds in them may contribute significantly to their antioxidant capacity. The modification of the classical DPPH method, with a methanol-buffer and the measure of EC20 (quantity of sample needed to decrease the initial DPPH concentration by 20%) has shown that sulfur-containing compounds such as cysteine (0.037 ±
