CENTENARIANS TRANSCRIPTOME IS UNIQUE AND REVEALS A ROLE OF BCL-XL IN SUCCESSFUL AGING

2017

Centenarians not only enjoy an extraordinary aging, but also show a compression of morbidity. We identified 1721 mRNAs differentially expressed by PMBCs from centenarians when compared with septuagenarians and young people. Sub-network analysis led us to identify Bcl-xL as an important gene up-regulated in centenarians. We found that centenarians display lower plasma cytochrome C levels, higher mitochondrial membrane potential and also less cellular damage accumulation. Immune function is significantly impaired in septuagenarians compared with young people whereas centenarians maintain it. To further ascertain the functional role of Bcl-xL in cellular aging, we found in transduced lymphocyt…

The soluble form of pan-RTK inhibitor and tumor suppressor LRIG1 mediates downregulation of AXL through direct protein–protein interaction in gliobla…

2019

Abstract Background Targeted approaches for inhibiting epidermal growth factor receptor (EGFR) and other receptor tyrosine kinases (RTKs) in glioblastoma (GBM) have led to therapeutic resistance and little clinical benefit, raising the need for the development of alternative strategies. Endogenous LRIG1 (Leucine-rich Repeats and ImmunoGlobulin-like domains protein 1) is an RTK inhibitory protein required for stem cell maintenance, and we previously demonstrated the soluble ectodomain of LRIG1 (sLRIG1) to potently inhibit GBM growth in vitro and in vivo. Methods Here, we generated a recombinant protein of the ectodomain of LRIG1 (sLRIG1) and determined its activity in various cellular GBM mo…

Depletion of Blautia Species in the Microbiota of Obese Children Relates to Intestinal Inflammation and Metabolic Phenotype Worsening

2020

Cross-sectional studies conducted with obese and control subjects have suggested associations between gut microbiota alterations and obesity, but the links with specific disease phenotypes and proofs of causality are still scarce. The present study aimed to profile the gut microbiota of lean and obese children with and without insulin resistance to characterize associations with specific obesity-related complications and understand the role played in metabolic inflammation. Through massive sequencing of 16S rRNA gene amplicons and data analysis using a novel permutation approach, we have detected decreased incidence of Blautia species, especially Blautia luti and B. wexlerae, in the gut mic…

Stable isotopes covering 96-7 ka BP from stalagmite CM (Santo Tomas Cave, Cuba)

2020

We present a new speleothem trace element and stable isotope record, which extends previous paleoclimate evidence from Cuban speleothems to the last 96 ka. Stable isotope samples were micromilled at a resolution of 0.10-0.33mm, and measured using an IRMS equipped with a Gasbench. Line scans of Element/Calcium ratios of the speleothem were measured by laser ablation ICPMS and were reduced to the resolution of the stable isotope records.

María Magdalena. Continuidad y variación en la cultura audiovisual (1897-2014)

2017

Sources of geomaterials in the Sicani Mountains during the Early Middle Ages: A case study of Contrada Castro, central western Sicily

2022

From 2017, an unknown rural settlement in Contrada Castro at Corleone (Palermo Province, western Sicily) was investigated as part of the `Harvesting Memories Project¿. The stratigraphic sequence, supported by radiocarbon dating, has demonstrated a reoccupation of a pre-Roman site during the transition between the Byzantine and Islamic periods. In particular, the main occupation occurred in the late 8th¿9th century when pottery kilns and a probable warehouse were constructed. During the 10th¿11th century, a new structure with different orientations replaced the previous buildings that had already collapsed. Specifically focusing on a perspective of the household production and its relationsh…

Correction of Long Vehicle Off Tracking by Multiple Axle Steering

2019

When planning the road vehicles for the transport of passengers or goods, engineers should take care to contrast some undesirable phenomena in the lateral dynamics, such as high-speed instability or off tracking. While the former is less troubling in the slow heavy vehicles, the latter is a serious problem due to the risk of invading the neighbouring lanes. The use of multi-wheel steering may completely cancel this drawback and, pursuing an optimized correlation among the steering angles, a functional inter-dependence based on the cubic “witch of Agnesi” is found to be very promising. Simple control systems with electromechanical or electronic components may be easily implemented for the mu…

[3]Rotaxanes and [3]pseudorotaxanes with a rigid two-bidentate chelate axle threaded through two coordinating rings

2009

New [3]rotaxanes and [3]pseudorotaxanes have been synthesised using the “gathering and threading” effect of copper(I). By using click chemistry as the “stoppering” reaction, a good yield of the [3]rotaxane was obtained, either as a dicopper complex or as a metal-free compound after demetallation. The axle contains a central rigid aromatic block incorporating two bidentate chelates, and the threaded macrocycles are 30-membered rings. A model dicopper(I) [3]pseudorotaxane whose axle was end-functionalised by triisopropylsilyl groups could be crystallised and studied by X-ray diffraction. A particularly attractive structure was obtained showing a “slanted” geometry for the two rings and the ax…

Study of the role of BCL-XL in healthy aging and frailty prevention

2022

Aging is defined as an intrinsic, progressive, deleterious and inevitable multifactorial phenomenon that occurs as a result of the gradual accumulation of damage at cellular and molecular levels. Due to the recent discovery of BCL-XL being overexpressed in peripheral blood mononuclear cells from centenarians, a new approach has emerged to investigate the BCL-XL function in healthy aging. The major goal of this thesis is to study the role of BCL-XL in frailty and healthy aging in transgenic mice overexpressing a human form of BCL-XL in T cells. For this, we used the heterozygous transgenic mice from Cg-Tg(LCKprBCL2L1)12Sjk/J strain, which express the human BCL-XL cDNA sequence in all thymocy…
