The hormone-induced depletion of cellular Ca stores provides a signal for the Ca2+ influx into electrically non-excitable cells; however, the underlying molecular mechanisms remain elusive. Therefore, we analyzed bradykinin-activated Ca2+ influx into human foreskin fibroblast cells, HF-15, by fura-2 and 45Ca labeling to discriminate between Ca2+ influx into the fura-sensitive compartment and Ca uptake into fura-insensitive Ca stores. Bradykinin-activated CaZt influx into the fura-sensitive compartment was blocked by inhibitors of NO synthases. We have studied the ligand-induced phosphorylation/dephosphorylation of the bradykinin B2 receptor endogenously expressed in human HF-15 fibroblasts. An antiserum (AS346) to a synthetic peptide (CRS36), derived from the extreme carboxyl terminus of the human B2 receptor, precipitated the receptor from solubilized membranes of HF-15 cells that had been labeled with [32P]orthophosphate. A low basal level of B2 receptor phosphorylation was found in the absence of a ligand. Stimulation of the cells with the B2 receptor agonists bradykinin, [Lys0,Hyp3]bradykinin, kallidin, and T-kinin resulted in a rapid and efficient phosphorylation of the receptor. To obtain data of the bradykinin B2 receptor's agonist binding site, we used a combined approach of affinity labeling and "immunoidentification" of receptor fragments generated by cyanogen bromide cleavage. Domain-specific antibodies to the various extracellular receptor domains were applied to detect receptor fragments with covalently attached [125I-Tyr8]bradykinin. As a cross-linker we used the homobifunctional reagent disuccinimidyl tartarate (DST), which reacts preferentially with primary amines. Premeditated automaticity : the role of explicit cognition in the development of obsessive smoking

- Abstract The purpose of the present study was to examine the role of explicit cognitions in relation to the development of obsessive smoking passion. Data derives from a longitudinal internet survey conducted among 939 daily smokers over a period of four months. Mental representations were the strongest predictor of obsessive smoking passion (ß=.47, p<.001) followed by habit strength (ß=.25, p<.001) and number of cigarettes smoked (ß=.09, p<.01). Altogether the variables accounted for 48% of the variance in obsessive smoking passion. Visual impairment and blindness are often caused by retinal ischemia-reperfusion (I/R) injury. We aimed to characterize a new model of I/R in pigs, in which the intraocular pathways were not manipulated by invasive methods on the ocular system. After 12 min of ischemia followed by 20 h of reperfusion, reactivity of retinal arterioles was measured in vitro by video microscopy. "Prosjektets varighet: 1/1-2004 - 31/12-2004" Målgruppe for surveyundersøkelsen var samtlige kvinner hjemmehørende i Aust-Agder som fødte fra 1/1-04 tom 31/12-2004. Det ble det utlevert 851 spørreskjemaer. 734 svar innkom, dvs. en svarprosent på 86,3 %. 241 kvinner (32,8 %) bekreftet at de røkte ved svangerskapets start. Undersøkelsen viste at hvorvidt kvinnene definerte seg som røykere i stor grad var avhengig av hvordan spørsmål om røykevaner ble stilt. Alle grader av røyking inkludert angav 200 (27,2 %) av kvinnene at de røkte etter fødsel. Røykenedgang i løpet av svangerskapet i forhold til totalutvalget var altså på 5,6 %. However, some errors persist throughout the query writing process and are never corrected.
