α-Chymotrypsin-Catalyzed Reaction Confined in Block-Copolymer Vesicles

2010

Herein the reactivity of the enzyme α-chymotrypsin in the confinement of polystyrene-block-poly(acrylic acid) (PS-b-PAA) vesicles was investigated. Enzyme and substrate molecules were encapsulated in PS-b-PAA vesicles with internal diameters ranging from 26 nm to 165 nm during the formation of the vesicles. While the loading efficiencies of enzyme and substrate molecules were practically identical for vesicles of identical size, they were found to increase with decreasing vesicle size. The kinetics of the α-chymotrypsin catalyzed hydrolysis of N-succinyl-Ala-Ala-Phe-7-amido-4-methylcoumarin (AMC) was evaluated following the increase of the absorption of the product 7-amino-4-methylcoumarin …

Sumatriptan Succinate Transdermal Delivery Systems for The Treatment of Migraine

2007

We have successfully obtained sumatriptan transdermal systems with different polymer compositions: methyl cellulose (MC), polyvinyl pyrrolidone (PVP) and a polyvinyl pyrrolidone (PVP)-polyvinyl alcohol (PVA) mixture. The systems contained 1,2-propylenglycol (MC) or sorbitol as a plasticizer (PVP and PVP-PVA), methacrylate copolymer as an adhesive agent, and an occlusive liner. Azone (5%, w/w) was incorporated into all the systems as a percutaneous enhancer. Transdermal systems are thin, transparent and non-adhesive when in a dry state. The permeation of sumatriptan succinate across pig ear skin was studied using the systems prepared. The formulation with MC polymer produced a statistically …

Hygrothermal effect on moisture kinetics and mechanical properties of hemp/polypropylene composite: Experimental and numerical studies

2015

The exposition of thermoplastic composites reinforced with natural fibers to moisture and heat is likely to lower their mechanical properties and restrain their use in the design of parts. The present work is focussed on investigating the effects of hygrothermal aging on the mechanical behaviour of injected polypropylene reinforced with short hemp fibers. Moreover, the kinetic of isothermal moisture absorption has been studied for four immersion temperatures. The experimental results show that the time taken to reach saturation decreases significantly with increase in immersion temperature. These tests also reveal that moisture absorption in this type of materials does not follow Fick's law…

Comparative enhancer effects of Span20 with Tween20 and Azone on the in vitro percutaneous penetration of compounds with different lipophilicities.

2000

Sorbitan monolaurate (Span20) was used in this study to analyze the influence of the polar functional group on the effects that non-ionic surfactants have on skin permeability. Its ethoxylate derivative polysorbate 20 (Tween20) and Azone, both with the same C12 alkyl chain as Span20, were used for comparative purposes. We evaluated the relative potency of the three molecules as enhancers in the permeability of a series of compounds with lipophilicities ranging from log Poct=-0.95 to log Poct=2.33. The influence of the enhancer concentration was also studied. For this purpose the epidermis of Wistar rat was pretreated with ethanolic solutions (1 and 5%, w/v) of each enhancer. Our results ind…

Absorption-partition relationships for true homologous series of xenobiotics as a possible approach to study mechanisms of surfactants in absorption.…

1992

Abstract This paper — the latest in this series of reports — deals with the effects of the anionic surfactant sodium lauryl sulfate on the colonic absorption of acidic xenobiotics (phenylalkylcarboxylic acids), as compared with those exerted by the nonionic polysorbate 80. The effects of these surfactants are qualitatively identical but quantitatively different. Thus, the increase in polarity of the colonic absorbant membrane is greater with lauryl sulfate, whereas micellar solubilization is much higher with polysorbate. As for the rest (i.e. the elimination of the stagnant aqueous diffusion layer as a limiting step for absorption), the two surfactants behave in a similar way. The biopharma…

Levofloxacin effect on erlotinib absorption. Evaluation of the interaction in undernutrition situations through population pharmacokinetic analysis i…

2017

The main objective of this study was to develop a pharmacokinetic model in order to describe the intestinal absorption of erlotinib in rat and to quantify the interaction of levofloxacin on this process in well- and under-nourished rats. Absorption studies were performed in male Wistar rats. Concentration-time profiles in proximal and distal intestine were analysed through non-linear mixed effect modelling using the NONMEM software version 7.3. Simulations were performed in order to explore the influence of covariates on the apparent absorption rate constant. A passive absorption and an active secretion process best-described erlotinib absorption from lumen to enterocyte. The developed mode…

Laboratory tests addressed to realize customized restoration procedures of underwater archaeological ceramic finds

2013

The present contribution is part of a biennial research project funded by the Italian Ministry of Education, Universities and Research (MIUR). This study, currently in progress, deals with innovative experimental approaches applied to the chemical, mineralogical and physical transformations occurring during the prolonged permanence of archaeological ceramic finds in seawater environments as well as to restoration and conservation issues of the same underwater artifacts. The experimental approach used in this research consisted in the manufacture of ceramic test-pieces (briquettes) and their successive placing in underwater environment. This work aims at assessing how textural and compositio…

Water Dynamics at the Solid-Liquid Interface to Unveil the Textural Features of Synthetic Nanosponges.

2020

A Fast-Field-Cycling NMR investigation was carried out on a set of polyurethane cyclodextrin nanosponges, in order to gain information on their textural properties, which have been proven to be quite difficult to assess by means of ordinary porosimetric techniques. Experiments were performed on both dry and wet samples, in order to evaluate the behavior of the “non-exchangeable” C-bound 1H nuclei, as well as the one of the mobile protons belonging to the skeletal hydroxyl groups and the water molecules. The results acquired for the wet samples accounted for the molecular mobility of water molecules within the channels of the nanosponge network, leading back to the possible pore size distrib…

Blood absorption capacity of different xenograft bone substitutes. An in-vitro study

2019

Background Commercially available xenograft blocks, claim to have adequate characteristics to interact with biological media and thus permitting biological fluid absorption. The objective of this in vitro study was to compare the blood absorption capacity of four different xenograft block materials of different composition of collagen and porosity. Material and Methods Four brands of xenograft block materials were used (NuOss®, Bio-Oss®, Osteobiol® and Smartbone®). Five samples of each brand were analyzed, making a total of 20 tests. Human blood was used as the absorption liquid for the present experiment. The time period, in which the block remains in contact with the blood, was registered…

Porphyrin amino acids-amide coupling, redox and photophysical properties of bis(porphyrin) amides.

2013

New trans-AB2C meso-substituted porphyrin amino acid esters with meso-substituents of tunable electron withdrawing power (B = mesityl, 4-C6H4F, 4-C6H4CF3, C6F5) were prepared as free amines 3a-3d, as N-acetylated derivatives Ac-3a-Ac-3d and corresponding zinc(II) complexes Zn-Ac-3a-Zn-Ac-3d. Several amide-linked bis(porphyrins) with a tunable electron density at each porphyrin site were obtained from the amino porphyrin precursors by condensation reactions (4a-4d) and mono- and bis(zinc(II)) complexes Zn(2)-4d and Zn(1)Zn(2)-4d were prepared. The electronic interaction between individual porphyrin units in bis(porphyrins) 4 is probed by electrochemical experiments (CV, EPR), electronic abso…
