SOX2 and BCL-2 Expressions in Odontogenic Keratocyst and Ameloblastoma

2020

Background The purpose of this experimental study was to compare the immunohistochemical expression of SOX2 and BCL-2 in Odontogenic Keratocyst (OKC) and Ameloblastoma (AB) specimens, and to identify a possible correlation in their expression. Material and Methods Immunohistochemical analysis was performed to evaluate SOX2 and BCL-2 expression in OKC (n = 20) and AB (n = 20). The immunoexpression was analyzed by a quantitative and qualitative scoring system. The comparison between the immunoexpression of SOX 2 and BCL-2 was assessed by the Mann-Whitney U-test. Spearman’s correlation coefficient evaluated the correlation between SOX2 and BCL-2 expressions. Results SOX2 and BCL-2 expression w…

GLANDULAR ODONTOGENIC CYST ASSOCIATED WITH AMELOBLASTOMA: CASE REPORT AND REVIEW OF THE LITERATURE

2017

Glandular odontogenic cyst (GOC) associated with ameloblastoma is an exceedingly rare histologic presentation with no known clinical significance or treatment applications. Four cases have been reported, three in the mandible and one in the maxilla. The age range is 14-65 and with male predilection. All four presented with swellings and two with pain. We add one more case to the literature of a 58-year old male presenting with an expansile multilocular radiolucency between teeth #19-23. The ameloblastomatous changes in this case are consistent with those of a unicystic ameloblastoma-mural subtype. Although the histologic changes are those of a unicystic ameloblastoma, the clinical and radio…

Odontogenic tumors in Western India (Gujarat): analysis of 209 cases

2011

Objective: Odontogenic tumors show a distinct geographic variation. In 2005 a new WHO classification was published which included odontogenic keratocyst as one of the odontogenic tumors, renaming it as a keratocystic odontogenic tumor. To our knowledge there are only few studies based on 2005 classification in Asian subcontinent. This study was done to determine the relative frequency of odontogenic tumors in Gujarat and compare it with reports from other parts of the world. Study Design: A retrospective study was designed. Necessary information was obtained from the records of the Oral Pathology Department, GDCH Ahmedabad. The histopathological diagnosis were re-evaluated according to the …

Ameoblastic carcinoma of mandible - A rare case report with review of literature

2010

Ameloblastic carcinoma is a rare malignant lesion with characteristic histologic features and behavior that dictates more aggressive surgical approach than that of a simple ameloblastoma. However, reliable evidence of its biologic activity is currently unavailable due to the scarcity of well-documented cases. It occurs primarily in the mandible in a wide range of age groups; no sex or race predilection has been noted. It may present as a cystic lesion with benign clinical features or as a large tissue mass with ulceration, significant bone resorption and tooth mobility. Because the lesion is usually found unexpectedly after an incisional biopsy or the removal of a cyst, a guide to different…

Whole exome sequencing and system biology analysis support the "two-hit" mechanism in the onset of Ameloblastoma

2021

Background Ameloblastoma is the most frequent odontogenic tumor. Various evidence has highlighted the role of somatic mutations, including recurrent mutation BRAF V600E, in the tumorigenesis of Ameloblastoma, but the intact genetic pathology remains unknown. Material and Methods We sequenced the whole exome of both tumor tissue and healthy bone tissue from four mandibular ameloblastoma patients. The identified somatic mutations were integrated into Weighted Gene Co-expression Network Analysis on publicly available expression data of odontoblast, ameloblast, and Ameloblastoma. Results We identified a total of 70 rare and severe somatic mutations. We found BRAF V600E on all four patients, sup…

High frequency of BRAF V600E mutation in Iranian population ameloblastomas

2020

Background Ameloblastoma is a common locally invasive but slow-growing neoplasm of the jaws with an odontogenic origin. Association between BRAF V600E mutation and clinicopathologic features and behavior of ameloblastoma remains controversial. This study aimed to evaluate BRAF V600E gene mutation and expression of its related proteins with clinicopathologic parameters in conventional ameloblastoma. Material and Methods 50 Formalin-fixed paraffin-embedded blocks were included in this study. Immunohistochemistry was done using rabbit monoclonal BRAF V600E mutation-specific antibody VE1. Quantitative real-time polymerase chain reaction assay was used for evaluating of BRAF V600E mutation. Resu…

Radiographic estimation of the growth rate of initially underdiagnosed ameloblastomas.

2019

Background To evaluate the specific growth rate (SGR) of ameloblastoma. Material and Methods Cases of ameloblastoma initially underdiagnosed (e.g. cases overlooked or diagnosed as reactive lesions) which had adequate radiographic documentation to evaluate their progression were retrospectively selected. Two panoramic radiographs were analyzed to determine the specific growth rate (SGR) of each tumor, defined as the logarithm of the ratio of final tumor area (when the diagnosis of ameloblastoma was made) to the initial tumor area (when the lesion was underdiagnosed), divided by the time interval between the radiographic images. The tumor area was measured using the software ImageJ. Results T…

Extreme ameloblastoma of the mandible with hypoproteinemia. A case report and review of clinicopathological features

2011

Ameloblastoma is a tumor of odontogenic epithelium. It has an intermediate malignant potential, thus positioning it in a gray zone between benign and malignant neoplasms. Invasive ameloblastomas of excessive size are rarely confronted. Although these ameloblastomas are benign, they are life threatening. Many have eventuated in death due to airway obstruction, starvation from restriction of feeding, and complications of hypoproteinemia. These tumors cause significant expansion of skin and lips [tissue expanders]. Due to its metabolic demands, a tumor of enormous size will have numerous feeding vessels. Patients with extreme ameloblastomas are usually from rural areas of developing countries …

Investigation of PTEN promoter methylation in ameloblastoma

2019

Background Phosphatase and tensin homolog (PTEN) acts as a tumor suppressor gene. Inactivation of PTEN has been reported in various types of cancers. PTEN promoter methylation possibly underlies PTEN inactivation, which results in tumorigenesis. The aim of this study was to investigate whether PTEN promoter methylation contributes to PTEN inactivation in ameloblastoma and its associated protein expression. Material and Methods In total, 20 fresh-frozen ameloblastoma samples were evaluated for PTEN promoter methylation using methylation-specific polymerase chain reaction (MS-PCR). A subset of 10 paraffin-embedded ameloblastoma samples was examined for PTEN expression through immunohistochemi…
