A microscale Kjeldahl nitrogen determination for environmental waters.

A microscale Kjeldahl system has been designed which has been homemade reduced. The digestor unit of a macroKjeldahl system and a modified glassware microdistillation unit have been used. The optimal conditions for digestion and distillation have been established for ammonium and methylamine as model compound of organic amino nitrogen. The optimised procedure has been applied to the determination of Kjeldahl nitrogen in water samples. Recoveries near 100% and good precision have been achieved. This procedure combines nitrogen miniaturized system and the use of a selective ion electrode for ammonia detection. Purification and Characterization of <I>Bacillus cereus</I> Protease Suitable for Detergent Industry

An extracellular alkaline protease from an alkalophilic bacterium, Bacillus cereus, was produced in a large amount by the method of extractive fermentation. The protease is thermostable, pH tolerant, and compatible with commercial laundry detergents. The protease purified and characterized in this study was found to be superior to endogenous protease already present in commercial laundry detergents. The enzyme was purified to homogeneity by ammonium sulfate precipitation, concentration by ultrafiltration, anion-exchange chromatography, and gel filtration. Characterization of high molecular mass material in ECF and TCF bleaching liquors by Py-GC/MS with and without TMAH methylation

Abstract High molecular weight (HMW) fractions (MW > 1000) of effluents were obtained by ultrafiltration of elemental chlorine-free (ECF) bleaching (O-D-EOP-D-ED) and totally chlorine-free (TCF) bleaching (O-Z-Q-P-Z-Q-P-P) liquors of softwood (Pinus sylvestris) pulp. The fractions were studied by analytical pyrolysis coupled to gas chromatography/mass spectrometry (Py-GC/MS) with and without TMAH (tetramethyl ammonium hydroxide) derivatization. The pyrograms showed a number of pyrolysis products mainly derived from lignin and carbohydrates. Separation of U(VI) and Th(IV) from Some Rare Earths by Thin Layer Chromatography with Di‐(2‐Ethylhexyl)‐Dithiophosphoric Acid on Silica Gel

Abstract Separation of U(VI) and Th(IV) from some rare earths by TLC has been studied using silica gel H and silica gel H impregnated with ammonium nitrate as the stationary phase. The solvent mixture ethyl‐methyl‐ketone–tetrahydrofuran (6∶3, v/v) containing di-(2‐ethylhexyl)-dithiophosphoric acid was used as the mobile phase. When silica gel H impregnated with 2.5 M NH4NO3 was used as stationary phase, the resolution was much improved and the separation of rare earths from each other was also achieved. The tetrad‐effect was observed in variation of R*m versus the atomic number of rare earths.

Improved detection of alkaloids in crude extracts applying capillary electrophoresis with field amplified sample injection

Abstract A simple and effective method for the sensitive detection of alkaloids in crude plant extracts applying capillary electrophoresis with field amplified sample injection (FASI) is described. This method was compared with normal pressure injection for the determination of alkaloids in methanolic extracts from roots of Berberis vulgaris L. (Berberidaceae) and Hydrastis canadensis L. (Ranunculaceae) using a 1:1 mixture of 200 mM ammonium acetate at pH 3.1 and methanol. The enantiomeric resolution of chiral phenoxy acid herbicides was performed by electrokinetic chromatography using a cyclodextrin as chiral pseudophase (CD-EKC). A systematic evaluation of several neutral and charged cyclodextrins was made. Among the cyclodextrins tested, (2-hydroxy)propyl beta-cyclodextrin (HP-beta-CD) was found to be the most appropriate for the enantioseparation of phenoxy acids. The interaction of native DNA with Zn(II) and Cu(II) complexes of 5-triethyl ammonium methyl salicylidene orto-phenylendiimine

The interaction of native calf thymus DNA with the Zn(II) and Cu(II) complexes of 5-triethyl ammonium methyl salicylidene orto-phenylendiimine (ZnL(2+) and CuL(2+)), in 1 mM Tris-HCl aqueous solutions at neutral pH, has been monitored as a function of the metal complex-DNA molar ratio by UV absorption spectrophotometry, circular dichroism (CD) and fluorescence spectroscopy. The results support for an intercalative interaction of both ZnL(2+) and CuL(2+) with DNA, showing CuL(2+) an affinity of approximately 10 times higher than ZnL(2+). A water-in-oil (w/o) microemulsion (ME) constituted by 15% Tris buffer, pH 8.4, in water and 85% sodium dodecyl sulfate (SDS)/n-pentanol 1:4 mixture, capable of dissolving up to 30% vegetable oils and lard, was used as background electrolyte in reverse microemulsion electrokinetic capillary chromatography (RMEEKC).
