From Small Peptides to Large Proteins against Alzheimer'sDisease.

Alzheimer’s disease (AD) is the most common neurodegenerative disorder in the elderly. The two cardinal neuropathological hallmarks of AD are the senile plaques, which are extracellular deposits mainly constituted by beta-amyloids, and neurofibrillary tangles formed by abnormally phosphorylated Tau (p-Tau) located in the cytoplasm of neurons. Although the research has made relevant progress in the management of the disease, the treatment is still lacking. Only symptomatic medications exist for the disease, and, in the meantime, laboratories worldwide are investigating disease-modifying treatments for AD. Abstract Background The acoustic levitation technique is a useful sample handling method for small solid and liquids samples, suspended in air by means of an ultrasonic field. This method was previously used at synchrotron sources for studying pharmaceutical liquids and protein solutions using x-ray diffraction and small angle x-ray scattering (SAXS). Methods In this work we combined for the first time this containerless method with small angle neutron scattering (SANS) and synchrotron radiation circular dichroism (SRCD) to study the structural behavior of proteins in solutions during the water evaporation. Low angle neutron scattering of ferritin studied by contrast variation.

Ferritins containing different amounts of iron have been studied by neutron small angle scattering in H 2 O/D 2 O mixtures. Apoferritin has also been studied in solutions of small organic molecules. There is a larger contrast variation in the solutions of small organic molecules than in the corresponding H 2 O/D 2 O experiments from which it may be calculated that I g of apoferritin contains 0·3 g of H 2 O. The solvent composition at zero contrast varies regularly with the iron content of the protein: there is evidence for polydispersity in the iron content of the ferritin used. Abstract Three dithiocarbamatonickel(II) complexes, [Ni(bzbudtc)2] (1), [Ni(bzbudtc)(PPh3)(NCS)] (2) and [Ni(bzbudtc)(PPh3)(CN)] (3) (where, bzbudtc = the N-benzyl-N-butyldithiocarbamato anion and PPh3 = triphenylphosphine), were prepared. All three complexes were analyzed by UV–Vis, IR and NMR (1H, 13C and 31P) spectra. The 13C NMR spectra of complexes 1–3 show the most crucial thioureide signal at around 200 ppm. A significant deshielding observed for the 31P signals in 2 and 3 reveals the effective bonding of phosphorus to the metal center. The crystal packing is characterized by N—H...N hydrogen bonds, which result in a two-dimensional network parallel to theac-plane.

In the title compound, C25H18NO, a stable phenanthridinic nitroxide, the ring containing the nitroxide function assumes a twist-boat conformation and the dihedral angle formed by adjacent benzene rings is 21.78 (5)°. The phenyl substituents at position 2 are approximately orthogonal to each other, forming a dihedral angle of 81.04 (4)°. The crystal structure is stabilized by an intramolecular C—H...O hydrogen bond and by C—H...π interactions.

International audience; The Mn(II) complex {[Mn(atpa)(H2O)2]·H2O}n (1),with the dicarboxylate ligand 2-aminoterephthalic acid (H2atpa), has been synthesized and crystallographically, spectroscopically and magnetically characterized. Complex 1 shows a very unusual 1κ2N,O coordination mode of the aminoterephthalate dianion with the Mn(II) ion. One of the carboxylate groups shows a syn-anti-μ2-η1:η1 binding mode to form a 2D square grid. The magnetic properties of this compound can be very well reproduced with a regular S = 5/2 chain model with a very weak antiferromagnetic coupling constant of J = −0.2 cm−1 through the single syn-anti carboxylate bridges.
