The Synapse-Like Interaction between Chloroplast, dictyosome, and Other Cell Compartments during Increased Ethylene Production in Leaves of Rye (Seca…

2000

Rye (Secale cereale L.) plants were treated with an ethylene releaser ethephon (2-chloroethylphosphonic acid) in concentration of 4×10−2 M. We studied electron microscopically, if and how chloroplasts interact with well-documented sites of ethylene production/binding, i.e., with endoplasmic reticulum, dictyosomes, mitochondria, plasma membrane, and tonoplast. During the sharp increase of ethylene synthesis in mesophyll cells of rye leaves, the direct local continguity of chloroplast envelope or envelope protrusions with the above mentioned cell compartments was typical. Moreover, a large number and diversity of versatile chloroplast-dictyosome associations were conspicuous, in which both th…

Un’indagine sperimentale sul ruolo della temperatura am- biente nel processo di riscaldamento/raffreddamento per ir- raggiamento

2012

The process of heating / cooling of an object in vacuum can be studied by the known Stefan's law. Many studies have investigated this phenomenon without inquire the role of the environment and of its temperature. In this paper the author proposes an experimental method through which to study the above process, by monitoring the ambient temperature on the assumption that this can vary with an exponential law. Under this hypothesis, in defined conditions, the temporal evolution of cooling of the object is similar to the case where the con- vection and / or the conduction are the principale phenomenon of energy exchange.

Going over the notes of the Jesuit Father Ignazio de Vio on the Festino di Santa Rosalia in 1693, one can linger over the numerous artistic personalities and workers from Palermo who were engaged on that occasion in embellishing the Cassaro with their creations, from silverware to precious drapes, gold, gems, stuccoes and ephemeral devices.

Effect of different cryopreservation protocols on the metaphase II spindle in human oocytes.

2008

The aim of this study was to evaluate the impact of different cryopreservation protocols on the repolymerization of metaphase (M)II spindles in human oocytes. Fresh aspirated donor oocytes were cryopreserved 3–4 h after retrieval using four different protocols: slow freezing using 1.5 mol/l 1,2-propanediol (PROH) + 0.2 mol/l sucrose (n = 36); 1.5 mol/l PROH + 0.3 mol/l sucrose (n = 34); 1.5 mol/l PROH + 0.3 mol/l sucrose with Na + depleted–choline replaced media (n = 27), and vitrification by the Cryotip method (n = 23). The control group comprised 34 fresh oocytes. Three hours after thawing, surviving and control oocytes were fixed for meiotic spindle/chromatin assessment. Survival rates w…

An interface protection system based on an embedded metrology system platform

2021

Abstract The aim of this work is to present an interface protection system (IPS) for Distributed Generators (DG) and Energy Storage Systems (ESS). The new prototype of IPS guarantees standard protection requirements, in terms of both voltage and frequency measurement accuracies and trip times. Moreover, it has the additional functionalities of implementing a communication link between the Distribution System Operator (DSO) and the DG and ESS Inverter. The new IPS is based on a smart meter platform with an integrated power line communication modem. Moreover, it has also an integrated metrology section. Experimental tests will show how this last feature allows a significant reduction of the m…

Molecular Mechanisms Controlling the Self-Assembly Process of Polyelectrolyte Multilayers

1998

The distance dependent interaction between polyelectrolyte-covered mica surfaces in aqueous solution was investigated with the surface forces apparatus. We find the following:  (i) The surface charge changes sign, when an oppositely charged polyelectrolyte from a concentrated polyelectrolyte solution is adsorbed. (ii) Tails and loops of the adsorbed polyions dangle into the bulk phase, inducing a small steric force. If polycations and poyanions are adsorbed on top of each other, a strong short range attractive force is seen due to ion-pair formation after crossing a large repulsive electrostatic/steric barrier. (iii) Obviously, after polyelectrolyte adsorption, there are still nonoccupied b…

4-4-20 anti-fluorescyl IgG Fab' recognition of membrane bound hapten: direct evidence for the role of protein and interfacial structure.

1995

The surface forces apparatus was used to identify the molecular forces that control the interactions of monoclonal 4-4-20 antifluorescyl IgG Fab' fragments with fluorescein-presenting supported planar bilayers. At long range, the electrostatic force between oriented Fab' and fluorescein monolayers was controlled by the composition of the protein exterior surrounding the antigen-combining site rather than by the overall protein charge. The measured positive electrostatic potential of the Fab' monolayer at pH > pI(Fab') was consistent with the structure of the exposed Fab' surface in which a ring of positive charge at the mouth of the antigen-combining site dominates the local electrostatic s…

Molecular mechanisms determining the strength of receptor-mediated intermembrane adhesion

1995

The strength of receptor-mediated cell adhesion is directly controlled by the mechanism of cohesive failure between the cell surface and underlying substrate. Unbinding can occur either at the locus of the specific bond or within the bilayer, which results in tearing the hydrophobic anchors from the membrane interior. In this work, the surface force apparatus has been used to investigate the relationship between the receptor-ligand bond affinities and the dominant mechanism of receptor-coupled membrane detachment. The receptors and ligands used in this study were membrane-bound streptavidin and biotin analogs, respectively, with solution affinities ranging over 10 orders of magnitude. With …

Specific Protein Binding to Functionalized Interfaces

1992

We report on the characterization of specific binding reactions between streptavidin and biotinylated model membrane surfaces. Self-assembly techniques as well as the Langmuir-Blodgett-Kuhn method were employed to prepare reactive, functionalized surfaces on various solid supports in contact with the aqueous protein solution. Plasmon surface polaritons optical measurements as well as atomic force microscopy and studies with the surface forces apparatus give rather detailed information as to the streptavidin monolayer formation, the kinetics of this process (either binding site- or diffusion limited), the selectivity of the reaction at laterally heterogeneous membranes, and the involved inte…

Ligand-receptor interactions directly measured with the surface forces apparatus

1991

Ligand-receptor interactions give rise to very strong bonds due to perfect geometrical fit. Using the Surface Forces Apparatus we have studied the interactions between membrane-bound biotin ligands and streptavidin receptors. We find an unusually strong short-range binding force associated with equally specific molecular rearrangements-both qualitatively and quantitatively unlike anything previously measured.
