Paxillin is a potential participant in the direct intracellular force transmission which is considered as the foundation of cells sensing and responding to extracellular environment. However, the detection of tension across paxillin has not been achieved due to lacking microsized tools. Herein, a paxillin tension sensor (PaxTs) based on Fluorescence Resonance Energy Transfer (FRET) technique was constructed. PaxTs can be expressed and assembled to FA sites spontaneously to visualize the tension across paxillin with FRET efficiency of ~62.4% in living cells. Human Topoisomerase I interaction with DNA on flexible nylon substrate for malaria diagnosis
The study of interaction between human Topoisomerase I (hTopo I) and the appropriate sequences of DNA is shown, with the aim to realize a biosensor for malaria diagnosis. Topoisomerases alter the topological state of DNA by catalysing the breaking and rejoining of DNA strands1. The ligase activity is fundamental for our biosensing strategy, revealing the pathology. hTopo I was chosen as a model system for the Plasmodium analogous. International audience; Nowadays, monitoring of volatile organic compounds (VOCs) is very important in various domains. In this work, we aimed to develop sensitive olfactory biosensors using odorant binding proteins (OBPs) as sensing materials. Three rat OBP3 derivatives with customized binding properties were designed and immobilized on the same chip for the detection of VOCs in solution by surface plasmon resonance imaging (SPRi). We demonstrated that the proteins kept their binding properties after the immobilization under optimized conditions.
