Enhancing Sensitivity of Microflow-Based Bottom-Up Proteomics through Postcolumn Solvent Addition.

2019

The introduction of more sensitive mass spectrometers allows researchers to adapt front-end liquid chromatography (LC) to individual needs for the analysis of complex proteomes. Where absolute sensitivity is not paramount, it is advantageous to switch from a highly sensitive nanoflow-LC setup, the de facto standard platform in mass-spectrometry (MS)-based discovery proteomics, to a more robust, high-throughput-compatible microflow or conventional-flow setup. To enhance the microflow-LC-MS electrospray process of complex proteomic samples, we tested the effects of different solvents, including 2-propanol, methanol, and acetonitrile, pure or as mixture with dimethyl sulfoxide, which were adde…

Hybrid QconCAT-Based Targeted Absolute and Data-Independent Acquisition-Based Label-Free Quantification Enables In-Depth Proteomic Characterization o…

2021

Wheat amylase/trypsin inhibitors (ATIs) have gained significant relevance as inducers of intestinal and extra-intestinal inflammation. In this study, we present a novel hybrid data-independent acquisition (DIA) liquid chromatography-mass spectrometry (LC-MS) approach, combining QconCAT technology with short microflow LC gradients and DIA and apply the method toward the quantitative proteome analysis of ATI extracts. The presented method is fast, robust, and reproducible and provides precise QconCAT-based absolute quantification of major ATI proteins while simultaneously quantifying the proteome by label-free quantification (LFQ). We analyzed extracts of 60 varieties of common wheat grown in…

Register affects language comprehension: ERP evidence from article omission in newspaper headlines

2011

Abstract Language processing involving syntax-discourse interface operations has been claimed to be particularly resource-consuming. In production, this additional complexity is claimed to be the source of article omission in the speech of young children and certain language-impaired speakers. In comprehension, article omission in some “special registers” (e.g., newspaper headlines) has been attributed to the trade-off between spending more processing resources and increasing processing speed. We investigated the comprehension of noun phrases (NPs) with and without articles (e.g., (a) policeman arrests (a) monk) when readers were or were not aware of reading headlines by recording electroph…

Ocean acidification as a driver of community simplification via the collapse of higher-order and rise of lower-order consumers

2017

AbstractIncreasing oceanic uptake of CO2 is predicted to drive ecological change as both a resource (i.e. CO2 enrichment on primary producers) and stressor (i.e. lower pH on consumers). We use the natural ecological complexity of a CO2 vent (i.e. a seagrass system) to assess the potential validity of conceptual models developed from laboratory and mesocosm research. Our observations suggest that the stressor-effect of CO2 enrichment combined with its resource-effect drives simplified food web structure of lower trophic diversity and shorter length. The transfer of CO2 enrichment from plants to herbivores through consumption (apparent resource-effect) was not compensated by predation, becaus…

Response of seagrasses and marine biofilms to natural acidification at CO2 vents

Settore ICAR/21 - UrbanisticaIn a framework in which the crisis is globalized but unequal and in the absence of a welfare system capable of responding to the urgencies of the most marginalized contexts and social groups a response to the new social and individual needs has been offered by networks of solidarity and from bottom-up mobilization practices that play a role of territorial agency often independently or in the absence of institutions. In the Palermo context a commitment made by a plurality of cultural subjects already active in the area gathered in a public assembly (Palermo si cura) during the period of the first lockdown gave rise to mutual aid actions to respond to the economic emergency and social and offer a proposal to cure the fragility of the city based not on charity but on the claim of a political dimension of collective action. Starting from the analysis of initiatives such as "Un sorriso per Ballarò" and "Abbiamo un (bi)sogno" the contribution analyzes the role the incidence and the fragility of these experiences in providing answers - often filling gaps - to a general interest and how these processes of value creation can hopefully contribute to the construction of local government policies capable of addressing emerging issues.

TOWARDS THE DECARBONIZATION OF THE EU BUILDING STOCK: AN INTEGRATED BUILDING STOCK RENOVATION MODELING APPROACH

2020

In Europe, the residential building stock is responsible for about 40% of the energy demand and about 36% of the CO2 emission at global level. Considering that almost 70% of the existing building stock will still be used in 2050, a long-term vision is needed to align with future challenges to avoid having significant increases in carbon emissions. The European policymakers have a long-recognized potential energy saving associated with the renovation of the existing building stocks, as demonstrated by the directives on the Energy performance of buildings (EPBD), introducing also the need to developed an integrated buildings design approach with environmental analysis. The purpose of this the…

2011

Introduzione.
