Effect of changes in the cellular energy state on glucose transport activity in Brevibacterium flavum
2000
Abstract The effect of changes in cellular energy state on 6[14C]glucose uptake and activity of the phosphoenolpyruvate:glucose phosphotransferase system (PTS) in Brevibacterium flavum RC 115 cells was investigated. Energy generation in cells was varied by adding an inhibitor of cellular respiration (potassium cyanide) and an uncoupler of oxidative phosphorylation (pentachlorophenol) to the cell culture as well as by changing thiamine concentration in the bacterial growth medium. The results showed that glucose uptake in ‘respiring’ cells was inversely correlated with bacterial respiration activity: uptake declined with excess ATP generation and increased with lower ATP synthesis caused by …
Comprehensive analysis of expression, subcellular localization, and cognate pairing of SNARE proteins in oligodendrocytes
2009
Oligodendrocytes form the central nervous system myelin sheath by spiral wrapping of their plasma membrane around axons, necessitating a high rate of exocytic membrane addition to the growing myelin membrane. Membrane fusion is mediated by soluble N-ethylmaleimide-sensitive factor attachment protein receptor proteins (SNAREs), which act by specific pairing of vesicle (R)- and target (Q)-SNAREs. To characterize oligodendroglial SNAREs and their trafficking pathways, we performed a detailed expression analysis of SNAREs in differentiating cultured oligodendrocytes and myelin and determined their subcellular localization. Expression of the plasma membrane Q-SNAREs syntaxin 3, syntaxin 4, SNAP2…

: Evolution of the examination at the end of college in sub-Saharan Africa
2004
The evolution of schooling in the developing systems in sub-Saharan Africa has brought a sizeable new population into the colleges. Unfortunately, the scarcity of resources and the need to pursue development of basic education prevents maintaining this population in the lycees. Thus the examination at the end of college becomes the focus of very stressful management of different currents and finds itself divided about aims which are not easy to reconcile. Original solutions can be found, as is the case in Mauretania, while the combination of better-known solutions may allow this examination to deal with these contradictions.
Experimental System Identification and Black Box Modeling of Hydraulic Directional Control Valve
2015
Directional control valves play a large role in most hydraulic systems. When modeling the hydraulic systems, it is important that both the steady state and dynamic characteristics of the valves are modeled correctly to reproduce the dynamic characteristics of the entire system. In this paper, a proportional valve (Brevini HPV 41) is investigated to identify its dynamic and steady state characteristics. The steady state characteristics are identified by experimental flow curves. The dynamics are determined through frequency response analysis and identified using several transfer functions. The paper also presents a simulation model of the valve describing both steady state and dynamic charac…
Propiedades psicométricas y estructura factorial del BRCS en una muestra de personas mayores españolas
2015
Successful resilience is a variable often related to an optimal ag-ing process. However, literature is rather limited when dealing with assess-ment instruments for the elderly in the Spanish language. The objective of this work is to validate the Brief Resilient Coping Scale (Sinclair & Wallston, 2004), a four item likert scale, in the Spanish elderly. For this propose, the scale was administrated to a sample of 991 elderly Spanish participants, and the data set analysed in terms of exploratory and confirm-atory factor analysis, internal consistency and criterion validity. The scale demonstrated good psychometric properties. Furthermore, the homogenei-ty indices were higher than the recentl…
Investigating bacterial populations in styrene-degrading biofilters by 16S rDNA tag pyrosequencing
2014
Microbial biofilms are essential components in the elimination of pollutants within biofilters, yet still little is known regarding the complex relationships between microbial community structure and biodegradation function within these engineered ecosystems. To further explore this relationship, 16S rDNA tag pyrosequencing was applied to samples taken at four time points from a styrene-degrading biofilter undergoing variable operating conditions. Changes in microbial structure were observed between different stages of biofilter operation, and the level of styrene concentration was revealed to be a critical factor affecting these changes. Bacterial genera Azoarcus and Pseudomonas were among…
Polyphasic study of wine Lactobacillus strains: taxonomic implications
2005
One hundred and seventy-eight lactobacilli isolated from wine were characterized by a polyphasic approach. Strains were phenotypically identified at genus and species level by classical tests including the analysis of cell morphology, homo/heterofermentative character, sugar fermentation patterns, growth at different temperatures and the optical nature of the isomer of lactic acid produced from glucose. Molecular techniques such as random amplification of polymorphic DNA (RAPD), amplified 16S rDNA restriction analysis (16S-ARDRA), PFGE-RFLP and ribotyping were used to characterize strains, and their potential for identification and/or typing was evaluated. The information obtained with thes…
Molecular analysis of the dominant lactic acid bacteria of chickpea liquid starters and doughs and propagation of chickpea sourdoughs with selected W…
2020
Abstract Fermented chickpea liquid is used as a leavening agent in chickpea bread production. In the present study, traditional chickpea liquid starter and dough samples were collected from bakeries in Turkey and microbiologically investigated. Culture-independent analysis for microbiota diversity, performed by MiSeq Illumina, identified Clostridium perfringens as major group in all samples, while Weissella spp. Dominated LAB community. A culture-dependent methodology was applied and 141 isolates were confirmed to be members of the LAB group based on 16s rRNA gene sequence analysis. In particular, 11 different LAB species were identified confirming the high frequency of isolation of weissel…
16S-ARDRA, a tool for identification of lactic acid bacteria isolated from grape must and wine.
2003
Lactic acid bacteria (LAB) are found in a great variety of habitats, including grape must and wines. There is a close relationship between the species of LAB which develop during fermentation and the eventual quality of the wine. For these reasons analytical techniques allowing fast and reliable identification of wine LAB are needed. In this work a simple and accurate protocol for identifying species of LAB isolated from grape must and wine is presented. This protocol is based on the amplification, directly from colony, of 16S rDNA and later digestion with one of the following restriction enzymes BfaI, MseI and AluI. A sequential use of the three enzymes is proposed to simplify LAB wine ide…
