Effect of Jasonia glutinosa on immune and oxidative status of gilthead seabream (Sparus aurata L.).

2020

Jasonia glutinosa (rock-tea, RT) has numerous biological activities. In the present work, the beneficial effects of dietary RT on gilthead seabream (Sparus aurata L.) were studied. Fish fed experimental diets containing 0 (control), 10 or 30% of RT for 15 and 30 days. Samples from skin mucus, blood, head kidney, liver and gut were obtained at 15 and 30 days. The antioxidant properties from RT were analysed such as the antioxidant capacity and phenolic content. The heat shock protein 70 level (HSP70) and the total oxidized proteins were evaluated on skin mucus as stress markers. Immune parameters, both humoral (peroxidase activity, immunoglobulin M levels and complement activity) and cellula…

Network-Wide Adaptive Burst Detection Depicts Neuronal Activity with Improved Accuracy

2017

Neuronal networks are often characterized by their spiking and bursting statistics. Previously, we introducedan adaptive burst analysis methodwhich enhances the analysis power for neuronal networks with highly varying firing dynamics. The adaptation is based on single channels analyzing each element of a network separately. Such kind of analysis was adequate for the assessment of local behavior, where the analysis focuses on the neuronal activity in the vicinity of a single electrode. However, the assessment of the whole network may be hampered, if parts of the network are analyzed using different rules. Here, we test how using multiple channels and measurement time points affect adaptive b…

NOX2ko Mice Show Largely Increased Expression of a Mutated NOX2 mRNA Encoding an Inactive NOX2 Protein

2020

Background: The superoxide-generating enzyme nicotinamide adenine dinucleotide phosphate (NADPH) oxidase (NOX2 or gp91phox, the phagocytic isoform) was reported as a major source of oxidative stress in various human diseases. Genetic deletion is widely used to study the impact of NOX2-derived reactive oxygen species (ROS) on disease development and progression in various animal models. Here, we investigate why NOX2 knockout mice show no NOX2 activity but express NOX2 mRNA and protein. Methods and Results: Oxidative burst (NOX2-dependent formation of ROS) was measured by L-012-based chemiluminescence and was largely absent in whole blood of NOX2 knockout mice. Protein expression was still de…

The Cumulative Effect of Transient Synchrony States on Motor Performance in Parkinson's Disease.

2020

Bursts of beta frequency band activity in the basal ganglia of patients with Parkinson's disease (PD) are associated with impaired motor performance. Here we test in human adults whether small variations in the timing of movement relative to beta bursts have a critical effect on movement velocity and whether the cumulative effects of multiple beta bursts, both locally and across networks, matter.

Rapid generation of hydrogen peroxide contributes to the complex cell death induction by the angucycline antibiotic landomycin E

2017

Landomycin E (LE) is an angucycline antibiotic produced by Streptomyces globisporus. Previously, we have shown a broad anticancer activity of LE which is, in contrast to the structurally related and clinically used anthracycline doxorubicin (Dx), only mildly affected by multidrug resistance-mediated drug efflux. In the present study, cellular and molecular mechanisms underlying the anticancer activity of landomycin E towards Jurkat T-cell leukemia cells were dissected focusing on the involvement of radical oxygen species (ROS). LE-induced apoptosis distinctly differed in several aspects from the one induced by Dx. Rapid generation of both extracellular and cell-derived hydrogen peroxide alr…

Post-transcriptional bursting in genes regulated by small RNA molecules

2018

Gene expression programs in living cells are highly dynamic due to spatiotemporal molecular signaling and inherent biochemical stochasticity. Here we study a mechanism based on molecule-to-molecule variability at the RNA level for the generation of bursts of protein production, which can lead to heterogeneity in a cell population. We develop a mathematical framework to show numerically and analytically that genes regulated post transcriptionally by small RNA molecules can exhibit such bursts due to different states of translation activity (on or off), mostly revealed in a regime of few molecules. We exploit this framework to compare transcriptional and post-transcriptional bursting and also…

GRB 090313 AND THE ORIGIN OF OPTICAL PEAKS IN GAMMA-RAY BURST LIGHT CURVES: IMPLICATIONS FOR LORENTZ FACTORS AND RADIO FLARES

2010

We use a sample of 19 gamma-ray bursts (GRBs) that exhibit single-peaked optical light curves to test the standard fireball model by investigating the relationship between the time of the onset of the afterglow and the temporal rising index. Our sample includes GRBs and X-ray flashes for which we derive a wide range of initial Lorentz factors (40 < Γ < 450). Using plausible model parameters, the typical frequency of the forward shock is expected to lie close to the optical band; within this low typical frequency framework, we use the optical data to constrain εe and show that values derived from the early time light-curve properties are consistent with published typical values derived from …
