In situ forming hydrogels of new amino hyaluronic acid/benzoyl-cysteine derivatives as potential scaffolds for cartilage regeneration

A new chemical strategy is described to link ethylenediamino (EDA) groups to primary hydroxyl groups of hyaluronic acid (HA) and the obtained derivatives have been characterized by 1H-NMR and 13C-NMR analyses. Such HA–EDA derivatives have been exploited to control the functionalization degree in benzoyl-cysteine (BC) groups, chosen as moieties able to allow both self-assembling in aqueous media and an oxidative crosslinking. In particular, the kinetics of oxidation of thiol groups in HA–EDA–BC derivatives has been studied in Dulbecco's Phosphate Buffer Solution (DPBS) pH 7.4 by colorimetric assays and rheological measurements. Heme oxygenase-1 (HO-1) is induced in cells by various stimuli as a defense system against oxidative stress. It is known that reactive oxygen species (ROS) participates in the initiation and progression of osteoarthritis (OA) and several antioxidant systems may protect cartilage components. HO-1 induction or CO release from CORM-2 counteracts oxidative stress and protects against proinflammatory and catabolic effects of interleukin-1β in OA chondrocytes, osteoblasts, and synoviocytes as well as in OA osteochondral explants. Characterization of oriented protein-ceramic and protein-polymer-composites for cartilage tissue engineering using synchrotron μ-CT

Abstract In this paper we report on the synthesis of three different gelatine based scaffold materials for the reconstruction of articular cartilage defects. The first scaffold design is based on an unmodified, oriented gelatine network, while the second design further comprises an attached inorganic hydroxyapatite layer and the third design includes poly(l-lactide) microspheres as a model material for future drug-release applications. Use of reversed-phase C18 Sep-Pak cartridges for the purification and concentration of sepiapterin and other pteridines

Abstract Several pteridines have been tested for their ability to bind to C 18 Sep-Pak. Riboflavin, sepiapterin, deoxysepiapterin, 6-acetyl-7,8-dihydropterin, 6-acetyldihydrohomopterin and 3′-hydroxysepiapterin were strongly retained and all but 6-acetyldihydrohomopterin quantitatively recovered upon elution with 2 ml of methanol. The effect of the concentration and volume of the sample, pH and salt concentration on the retention of sepiapterin have been studied. The procedure was very useful for the purification, desalting, solvent exchange and concentration of pteridines having high affinity for the cartridge. Post-Processing via Cation Exchange Cartridges: Versatile Options

New 68Ge/68Ga radionuclide generators provide the positron emitter 68Ga (T½ = 67.7 min) as an easily available and relatively inexpensive source of a PET nuclide for labeling of interesting targeting vectors. However, currently available “ionic” 68Ge/68Ga radionuclide generators are not necessarily optimized for the routine synthesis of 68Ga-labeled radiopharmaceuticals in a clinical environment. Post-processing of 68Ge/68Ga generators using cation exchange resins provides chemically and radiochemically pure 68Ga with 97±2% within less than 4 min, with 68Ge almost completely removed, and ready for online labeling. Changes in collagen cross-linking of articular cartilage are revealed by spectral reflectance imaging

Excessive cross-linking of collagen during aging may contribute to degeneration of articular cartilage. Traditionally, the amount of cross-links is derived by using destructive high-performance liquid chromatography (HPLC). However, a sensitive, non-destructive method could help to evaluate tissue integrity, including cross-links. We increased collagen cross-linking in bovine articular cartilage by controlled threose incubation.
