Acetylcholine may play a role in cell activation and airway inflammation. Background: Insulin receptor substrate 1 (IRS-1) is a signaling molecule that exerts a key role in mediating cross talk between estrogen receptor a (ERa) and insulin-like growth factor 1 (IGF-1) in breast cancer cells. Previously, we demonstrated that a fraction of IRS-1 binds ERa, translocates to the nucleus, and modulates ERa-dependent transcription at estrogen response elements (ERE). Hormonal and nutritional control of L‐carnitine uptake in myoblastic C2C12 cells

L-Carnitine plays an important role in skeletal muscle bioenergetics, and its bioavailability and thus its import may be crucial for muscle function. We studied the effect of thyroid hormone, insulin, and iron overload, hormones and nutrients known to alter muscle metabolism, on L-carnitine import into C2C12 cells. We report here L-carnitine uptake is increased by thyroid hormones and decreased by iron. Insulin was found to be ineffective in altering the L-carnitine uptake.

Abstract Aims Acetylcholine (ACh) is synthesized by Choline Acetyl-Transferase (ChAT) that exerts its physiological effects in airway epithelial cells via muscarinic receptor (MR) activation. We evaluate the effect of ACh stimulation on human bronchial epithelial cells (16-HBE) and test whether cigarette smoke extract (CSE) can modify the basal cellular response to ACh affecting the non-neuronal cholinergic system signalling. The role of insulin-like growth factor II in the malignant transformation of rat liver oval cells

Oval cells are small nonparenchymal epithelial cells that first appear in the periportal areas of the liver and thereafter invade the whole parenchyma when mice or rats are exposed to a variety of chemical carcinogens. In the present study we have analyzed the expression of insulin-like growth factor II (IGF II) in the recently established oval cell line OC/CDE 22 and its malignantly transformed counterpart (the M22 cells) and the biological consequences of the constitutive expression of IGF II in oval cells. A rat colon cancer cell line (CC531) was implanted into the livers of 85 WAG rats and in animals surviving the implantation the liver tumour was subsequently treated with sham laparotomy (n = 15), resection (n = 20), cryotherapy (n = 19) or laser therapy (n = 20). Local recurrence or persistence of the tumour in animals surviving the procedure was observed in 15 of 15 following sham laparotomy, in 0 of 15 following resection, in 3 of 17 following cryotherapy and in 10 of 14 following laser therapy. However, peritoneal disease and lung metastases were more frequent and extensive following resection as compared to cryotherapy or laser therapy.

Lysine triggers apoptosis through a NADPH oxidase-dependent mechanism in human renal tubular cells

Progressive chronic kidney disease (CKD) is common in lysinuric protein intolerance (LPI), a primary inherited aminoaciduria characterized by massive Lysine excretion in urine. However, by which mechanisms Lysine may cause kidney damage to tubule cells is still not understood. This study determined whether Lysine overloading of human proximal tubular cells (HK-2) in culture enhances apoptotic cell loss and its associated mechanisms.
