Abstract We have investigated the binding of a new dansylcadaverine derivative of substance P (DNC-SP) with negatively charged small unilamellar vesicles composed of a mixture of phosphatidylcholine (PC) and either phosphatidylglycerol (PG) or phosphatidylserine (PS) using fluorescence spectroscopic techniques. The changes in fluorescence properties were used to obtain association isotherms at variable membrane negative charges and at different ionic strengths. Cholinergic-Adrenergic Presynaptic Interactions on the Heart in the Millisecond Range as Studied With the Pulse-to-Pulse Method

A method has been devised to study the interactions of cholinergic and adrenergic nerves in the rabbit atria, a tissue known to be densely innervated by both vagus and sympathetic nerve fibres. The atria were isolated and perfused via the ascending branches of the coronary arteries. The acetylcholine (ACh) stores were labelled with [14C]-choline and the noradrenaline (NA) stores with [3H]-NA. The radioactive compounds were separated on columns and determined by scintillation spectrometry.

δ 1‐OPIOID receptor‐mediated controlofacetylcholine (ACh) release in human neocortex slices

In slices of human neocortex, prelabelled with [3H]-choline, the release of [3H]-acetylcholine reflects the evoked release of endogenous acetylcholine which was elicited by the same electrical stimulation paradigm. [3H]-Acetylcholine release was depressed by the delta-opioid receptor agonist D-Pen2-D-Pen5-enkephalin. When the nerve endings were depolarized by elevating extracellular potassium the evoked [3H]-acetylcholine release was similarly depressed by D-Pen2-D-Pen5-enkephalin in the absence, but not in the presence, of tetrodotoxin which blocks action potential propagation. 1. The effect of histamine and histamine H1- and H2-receptor agonists on isolated myometrium strips of premenopausal women has been examined. The effect of acetylcholine was also determined. 2. Histamine, 2-pyridylethylamine, 4-methylhistamine and acetylcholine, but not dimaprit, produced a concentration-related contractile response in human isolated myometrial strips. Impact of insulin on microvascular blood flow and endothelial cell function in the postprandial state in patients with Type 1 diabetes.

The aim of the present study was to investigate postprandial microvascular blood flow following a standardized test meal in nondiabetic subjects and in patients with Type 1 diabetes after regular insulin or insulin lispro. It is expected that microvascular blood flow might be affected by blood glucose, blood insulin and C-peptide levels. In our investigation skin microvascular blood flow (LDF) was measured using laser doppler fluxometry at skin temperatures of 37 degrees C and 44 degrees C during a 75 g oral glucose load (OGT) or water in ten healthy volunteers (6 male, 4 female, age: 28.1+/-4.0) who had fasted overnight.
