Effects of Glyburide-Cyclosporin A Interaction on Interleukin-2 Production in Rats1

The effects of simultaneous administrations of Cyclosporin A (CsA) and Glyburide on the immune system of rats has been evaluated in terms of Interleukin-2 (IL-2) production by Concanavalin A (ConA) stimulated splenocytes and exogenous IL-2 binding capacity. The inhibitory effect of Cyclosporin A on IL-2 production of lymphoid cells is well known. Spleen cells from rats receiving CsA had reduced levels of IL-2 when compared to untreated controls or rats receiving Glyburide only. Barnidipine block of L-type Ca2+ channel currents in rat ventricular cardiomyocytes

The effects of barnidipine and nifedipine on L-type Ca2+ current (ICa(L)) were investigated in ventricular cardiomyocytes from rats. Both barnidipine and nifedipine reduced ICa(L) in a concentration and voltage dependent manner; the EC50 were 80 and 130 nM at a holding potential of −80 mV, respectively, and 18 and 6 nM at −40 mV, respectively. Both drugs induced a leftward shift of the steady-state inactivation curve of ICa(L). Effect of tormentic acid on insulin secretion in isolated rat islets of langerhans

Tonnentic acid was isolated as the hypoglycemic agent of Poterium ancistroides Desf. The present study reports effects of this compound on isolated islets of Langerhans. We demonstrate that tormentic acid in the presence of 1.66 mM glucose initiates insulin secretion by isolated rat islets of Langerhans in a dose-dependent fashion at concentrations ranging from 0.05 to 0.5 mM. However, the compound has no effect on the insulin-releasing capacity of 16.6 mM glucose. Similar results were obtained with the sulfonylurea glibenclamide (0.01 mM) used as a reference. These results suggest that the hypoglycemic effect of tormentic acid is due to a diect effect of the compound in vitro.

Background Voltage-gated sodium (Nav) channels have traditionally been considered a trademark of excitable cells. However, recent studies have shown the presence of Nav channels in several non-excitable cells, such as astrocytes and macrophages, demonstrating that the roles of these channels are more diverse than was previously thought. Despite the earlier discoveries, the presence of Nav channel-mediated currents in the cells of retinal pigment epithelium (RPE) has been dismissed as a cell culture artifact. Physostigmine and Neuromuscular Transmission

Single channel studies carried out in cultured rat myoballs and cultured hippocampal neurons, and ion flux studies performed on Torpedo electrocyte membrane vesicles, showed that physostigmine (Phy), a well-established acetylcholinesterase inhibitor, interacts directly with nicotinic acetylcholine receptors (nAChR). Low concentrations (0.1 microM) of Phy activate the receptor integral channel, whereas higher concentrations blocked the channel in its opened state. The economically important activity of metal processing can tend to contribute to the degradation of the environment. Smelting is an important source of contaminants, dispersing large quantities of potentially toxic elements (PTE) and coproducts into the environment. Soils in the vicinity of smelters frequently contain high concentrations of PTE.
