Discrepancies between archaeological and 14C-based chronologies: problems and possible solutions

2018

14C dating of bone collagen is believed to produce the most reliable absolute dates for the Central European Early Neolithic, as the selection of bones in anatomical context minimises taphonomic problems. In contrast, a comparison of three newly published local or regional chronological models as well as a comparison of several series of dates from bone collagen, charcoal and cereals highlights problems probably caused by diagenetic influences, especially on collagen. Therefore, at least the checking of bone collagen 14C dates against charcoal or cereal dates from the same contexts seems to be indispensable.

X-ray investigations of the superstructure of collagen

2008

When compared with results from humid fibers, characteristic changes of the SAXS-pattern taken from dried collagen fibers have been observed. The average lateral width of the various reflexions increases with the order of reflexion whereby a fluctuation is superimposed, the type of which cannot easily be related to the reflexion order. Moreover, each of the reflexions is obviously laterally split into three components. An explanation of these characteristics can be given with the aid of a distinct model, assuming that the collagen fibrils have definitely varying diameters along their axis, whereby paracrystalline distortions appear, represented by nematic fluctuations. Additionally, there i…

Disruption of apical-basal polarity of human embryonic stem cells enhances hematoendothelial differentiation

2007

Abstract During murine development, the formation of tight junctions and acquisition of polarity are associated with allocation of the blastomeres on the outer surface of the embryo to the trophoblast lineage, whereas the absence of polarization directs cells to the inner cell mass. Here, we report the results of ultrastructural analyses that suggest a similar link between polarization and cell fate in human embryos. In contrast, the five human embryonic stem cell (hESC) lines displayed apical-basal, epithelial-type polarity with electron-dense tight junctions, apical microvilli, and asymmetric distribution of organelles. Consistent with these findings, molecules that are components of tigh…

Isolation of fibroblasts for coating of meshes for reconstructive surgery: differences between mesh types.

2009

Aims: An extensive colonization of surgical meshes with autologous fibroblasts may reduce complications. Therefore, we aimed to establish a technique that allows isolation and propagation of fibroblasts from vaginal biopsies. Using these cells we tested the applicability of several clinically applied meshes for fibroblast coating. Materials & methods: Fibroblasts were isolated from vaginal tissue after digestion with collagenase. Characterization was performed by immunostaining for cytokeratin 5, 6 and 14, smooth muscle actin and vimentin. A semiquantitative technique was applied to determine the degree of mesh coating 5 h and 5 weeks after seeding of fibroblasts. Seven meshes of diffe…

Calpains promote α2β1 integrin turnover in nonrecycling integrin pathway

2012

A novel virus- and integrin clustering–specific pathway diverts integrin from its normal endo/exocytic traffic to a nonrecycling degradative endosomal route. Clustering of α2β1 integrin causes redistribution of the integrin to perinuclear endosomes, leading to enhanced integrin turnover promoted by calpains.

Extracorporeal Shock Waves Increase Markers of Cellular Proliferation in Bronchial Epithelium and in Primary Bronchial Fibroblasts of COPD Patients

2020

Chronic obstructive pulmonary disease (COPD) is due to structural changes and narrowing of small airways and parenchymal destruction (loss of the alveolar attachment as a result of pulmonary emphysema), which all lead to airflow limitation. Extracorporeal shock waves (ESW) increase cell proliferation and differentiation of connective tissue fibroblasts. To date no studies are available on ESW treatment of human bronchial fibroblasts and epithelial cells from COPD and control subjects. We obtained primary bronchial fibroblasts from bronchial biopsies of 3 patients with mild/moderate COPD and 3 control smokers with normal lung function. 16HBE cells were also studied. Cells were treated with a…

Integrins α5β1, αvβ1, and αvβ6 Collaborate in Squamous Carcinoma Cell Spreading and Migration on Fibronectin

2000

Abstract The expression of αvβ6 fibronectin/tenascin receptor integrin is induced in malignant transformation of oral epithelium. In this study, we demonstrate the contribution of αvβ6 as well as other fibronectin receptor integrins in squamous cell carcinoma (SCC) cell adhesion and migration. Of 11 SCC cell lines isolated from the head and neck area, 8 (73%) expressed αvβ6 integrin on the cell surface. Three cell lines were chosen for further functional experiments: 1 with relatively high, 1 with moderate, and 1 with minimal surface expression of αvβ6 integrin. In addition to αvβ6, all 3 cell lines expressed α5β1 and αvβ1 fibronectin receptor integrins. Function-blocking experiments with i…

Galvanic deposition of Hydroxyapatite/Chitosan/Collagen coatings on 304 stainless steel

2021

The galvanic deposition method was used to deposit Hydroxyapatite/Chitosan/Collagen coatings on 304 stainless steel. Galvanic deposition is an alternative and valid way to fabricate bio-coatings with high biocompatibility and good anticorrosion properties. Physical-chemical characterizations were carried out to investigate chemical composition and morphology of the samples. Coatings consist of a mixture of calcium phosphate (Brushite and Hydroxyapatite) with chitosan and collagen. Corrosion tests were performed in the simulated body fluid (SBF) after different aging times. Results show that, in comparison with bare 304 stainless steel, coating shifts corrosion potential to anodic values and…
