Characterization of the first extended-spectrum beta-lactamase-producing nontyphoidal Salmonella strains isolated in Tehran, Iran.

2009

The infections caused by Salmonella remain a significant public health problem throughout the world. beta-Lactams and fluoroquinolones are generally used to treat invasive Salmonella infections, but emergence and spread of antibiotic-resistant strains are being increasingly notified in many countries. In particular, detection of extended-spectrum beta-lactamases (ESBLs) in Salmonella spp. is a newly emerging threat worldwide. This study was carried out to characterize beta-lactamase-producing Salmonella strains identified in Tehran, Iran. Over the 2-year period from 2007 to 2008, 6 of 136 Salmonella isolates recovered from pediatrics patients, including three Salmonella enterica serotypes E…

New site-specific bioconjugation strategies and their application to the development of molecular imaging agents for cancer diagnostic

2018

The preparation of targeted imaging agents suitable for cancer diagnostic involves the coupling of an imaging probe to a vector, generally a protein, able to specifically target cancerous tissues. Unfortunately, this bioconjugation step is often problematic. Indeed, proteins present a large variety of reactive functions that can potentially interfere with the coupling reaction. The purpose of this work is the design of innovative selective bioconjugation strategies that can facilitate the synthesis of molecular imaging agents. Our research led to the development of a simple and efficient approach allowing the modification of a protein with a fluorophore by using a site-specific bioconjugatio…

Detoxication of carcinogenic fjord-region diol epoxides of polycyclic aromatic hydrocarbons by glutathione transferase P1-1 variants and glutathione.

1998

AbstractEpidemiological studies suggest that individuals differing in the expression of allelic variants of the human glutathione transferase (GST) Pi gene differ in susceptibility to chemical carcinogens such as polycyclic aromatic hydrocarbons (PAH). This study reports the catalytic efficiencies (kcat/Km) of two naturally occurring variants, GSTP1-1/I-105 and GSTP1-1/V-105, towards a series of fjord-region diol epoxides representing potent biologically active PAH metabolites, and two GSTP1-1 mutants with Ala105 and Trp105 in the active site. The results indicate that individuals who are homozygous for the allele encoding GSTP1-1/V-105 might be more susceptible to PAH carcinogenesis due to…

G2(MP2) molecular orbital study of the substituent effect in the H3BPH3−nFn (n=0–3) donor–acceptor complexes

2000

Abstract The complexation energies of H3BPH3−nFn (n=0–3) and the proton affinities of PH3−nFn compounds have been investigated at the G2(MP2) level of theory. G2(MP2) results show that the successive fluorine substitution on the phosphine increases the stability of H3BPH3−nFn complexes although the basicity of the PH3−nFn ligands reduces with this substitution. The NBO partitioning scheme shows that this stability was related to the hyperconjugation effect. It proves also that the shortening of the P–H and P–F bond lengths, upon complexation, is due to an increasing `s' character in these bonds.

Multifunctional platforms for cancer theranosis

2018

The objective of this thesis was to develop multifunctional tools for cancer theranosis, particularly based on nanotechnology. The work is based on the premise of monomolecular multimodal platform that aims to create the building blocks that can be used for the synthesis of functionalized nanoparticles, bioconjugates or targeted small molecular imaging probes. The initial work focused on the synthesis of the chelator based silane precursors. These novel precursors were employed for the functionalization of AGuIX nanoparticles and have also been implicated in a one pot AGuIX synthesis. Radiolabelling of these nanoparticles was performed with the aim to evaluate the stability of these nanopart…

Plasmid conjugation from Proteobacteria as evidence for the origin of xenologous genes in Cyanobacteria

2014

Comparative genomics have shown that 5% of Synechococcus elongatus PCC 7942 genes are of probable proteobacterial origin. To investigate the role of interphylum conjugation in cyanobacterial gene acquisition, we tested the ability of a set of prototype proteobacterial conjugative plasmids (RP4, pKM101, R388, R64, and F) to transfer DNA from Escherichia coli to S. elongatus. A series of BioBrick-compatible, mobilizable shuttle vectors was developed. These vectors were based on the putative origin of replication of the Synechococcus resident plasmid pANL. Not only broad-host-range plasmids, such as RP4 and R388, but also narrower-host-range plasmids, such as pKM101, all encoding MPFT-type IV …

Kondrat'eva ligation: Diels-Alder-based irreversible reaction for bioconjugation.

2014

International audience; Diversification of existing chemoselective ligations is required to efficiently access complex and well-defined biomolecular assemblies with unique and valuable properties. The development and bioconjugation applications of a novel Diels-Alder-based irreversible site-specific ligation are reported. The strategy is based on a Kondrat'eva cycloaddition between bioinert and readily functionalizable 5-alkoxyoxazoles and maleimides that readily react together under mild and easily tunable reaction conditions to afford a fully stable pyridine scaffold. The potential of this novel bioconjugation is demonstrated through the preparation of fluorescent conjugates of biomolecul…

Hepatīta B kora antigēna veidoto vīrusveidīgo daļiņu modificēšana vakcīnu prototipa izstrādei

2017

From Biocompatible to Biodegradable: Poly(Ethylene Glycol)s with Predetermined Breaking Points

2013

Poly(ethylene glycol) (PEG) is the gold standard polymer for biomedical applications. PEG is known for its biocompatibility and antifouling properties and is widely used for bioconjugation. However, like other synthetic polymers in the field, PEG is not biodegradable, limiting its use for parenteral formulations and protein conjugation to a molecular weight range with a specific upper limit (commonly 40–60 kDa) to avoid polyether accumulation in human tissue. For these biomedical applications, but also for other purposes such as cleavable hydrogels and templates for porous membranes, several routes for the insertion of in-chain biocleavable moieties, such as acetals or disulfides, into PEG …
