Graph-theoretical derivation of brain structural connectivity

2020

Brain connectivity at the single neuron level can provide fundamental insights into how information is integrated and propagated within and between brain regions. However, it is almost impossible to adequately study this problem experimentally and, despite intense efforts in the field, no mathematical description has been obtained so far. Here, we present a mathematical framework based on a graph-theoretical approach that, starting from experimental data obtained from a few small subsets of neurons, can quantitatively explain and predict the corresponding full network properties. This model also changes the paradigm with which large-scale model networks can be built, from using probabilisti…

The Importance of Cerebellar Connectivity on Simulated Brain Dynamics

2020

The brain shows a complex multiscale organization that prevents a direct understanding of how structure, function and dynamics are correlated. To date, advances in neural modeling offer a unique opportunity for simulating global brain dynamics by embedding empirical data on different scales in a mathematical framework. The Virtual Brain (TVB) is an advanced data-driven model allowing to simulate brain dynamics starting from individual subjects' structural and functional connectivity obtained, for example, from magnetic resonance imaging (MRI). The use of TVB has been limited so far to cerebral connectivity but here, for the first time, we have introduced cerebellar nodes and interconnecting…

The Drosophila Larval Locomotor Circuit Provides a Model to Understand Neural Circuit Development and Function

2021

It is difficult to answer important questions in neuroscience, such as: “how do neural circuits generate behaviour?,” because research is limited by the complexity and inaccessibility of the mammalian nervous system. Invertebrate model organisms offer simpler networks that are easier to manipulate. As a result, much of what we know about the development of neural circuits is derived from work in crustaceans, nematode worms and arguably most of all, the fruit fly, Drosophila melanogaster. This review aims to demonstrate the utility of the Drosophila larval locomotor network as a model circuit, to those who do not usually use the fly in their work. This utility is explored first by discussion…

Brain-like large scale cognitive networks and dynamics

2018

A new approach to the study of the brain and its functions known as Human Connectomics has been recently established. Starting from magnetic resonance images (MRI) of brain scans, it is possible to identify the fibers that link brain areas and to build an adjacency matrix that connects these areas, thus creating the brain connectome. The topology of these networks provides a lot of information about the organizational structure of the brain (both structural and functional). Nevertheless this knowledge is rarely used to investigate the possible emerging brain dynamics linked to cognitive functions. In this work, we implement finite state models on neural networks to display the outcoming bra…

Normative vs. patient-specific brain connectivity in Deep Brain Stimulation

2020

AbstractBrain connectivity profiles seeding from deep brain stimulation (DBS) electrodes have emerged as informative tools to estimate outcome variability across DBS patients. Given the limitations of acquiring and processing patient-specific diffusion-weighted imaging data, most studies have employed normative atlases of the human connectome. To date, it remains unclear whether patient-specific connectivity information would strengthen the accuracy of such analyses. Here, we compared similarities and differences between patient-specific, disease-matched and normative structural connectivity data and retrospective estimation of clinical improvement that they may generate.Data from 33 patien…

Structural Covariance of Cortical Gyrification at Illness Onset in Treatment Resistance: A Longitudinal Study of First-Episode Psychoses

2021

AbstractTreatment resistance (TR) in patients with first-episode psychosis (FEP) is a major cause of disability and functional impairment, yet mechanisms underlying this severe disorder are poorly understood. As one view is that TR has neurodevelopmental roots, we investigated whether its emergence relates to disruptions in synchronized cortical maturation quantified using gyrification-based connectomes. Seventy patients with FEP evaluated at their first presentation to psychiatric services were followed up using clinical records for 4 years; of these, 17 (24.3%) met the definition of TR and 53 (75.7%) remained non-TR at 4 years. Structural MRI images were obtained within 5 weeks from first…

Breakdown of Thalamo-Cortical Connectivity Precedes Spike Generation in Focal Epilepsies

2017

Electroencephalography (EEG) spikes and focal epileptic seizures are generated in circumscribed cerebral networks that have been insufficiently described. For precise time and spatial domain network characterization, we applied in patients with focal epilepsy dense array 256-channel EEG recordings with causal connectivity estimation by using time-resolved partial directed coherence and 3T-magnetic resonance imaging-derived cortical and thalamus integrity reconstruction. Before spike generation, significant theta and alpha bands driven information flows alterations were noted from both temporal and frontal lobes to the thalamus and from the thalamus to the frontal lobe. Medial dorsal and ven…

Impaired Right Temporoparietal Junction–Hippocampus Connectivity in Schizophrenia and Its Relevance for Generating Representations of Other Minds

2018

Schizophrenia is associated with impaired and exaggerated Theory of Mind processes, pointing on alterations in generating a representation of another person's mind. Despite recent work on healthy subjects suggesting that a coupling between the right temporoparietal junction (rTPJ) and the hippocampus is relevant for building representations of others' intentions, the neural basis of related dysfunctions in patients with schizophrenia remains unclear. Therefore, we used structural and functional magnetic resonance imaging together with a modified prisoner's dilemma game to test the hypotheses, that patients show dysfunctional social updating on behavioral level accompanied by altered rTPJ-hi…

Resting-State Functional Connectome in Patients with Brain Tumors Before and After Surgical Resection

2020

Purpose: High-grade glioma surgery has evolved around the principal belief that a safe maximal tumor resection improves symptoms, quality of life, and survival. Mapping brain function has been recently improved by resting-state functional magnetic resonance imaging (rest-fMRI), a novel imaging technique that explores networks connectivity at “rest.” Methods: This prospective study analyzed 10 patients with high-grade glioma in whom rest-fMRI connectivity was assessed both in single-subject and in group analysis before and after surgery. Seed-based functional connectivity analysis was performed with CONN toolbox. Network identification focused on 8 major functional connectivity networks. A v…

Reliability of Magnetoencephalography and High-Density Electroencephalography Resting-State Functional Connectivity Metrics

2019

Resting-state connectivity, for example, based on magnetoencephalography (MEG) or electroencephalography (EEG), is a widely used method for characterizing brain networks and a promising imaging biomarker. However, there is no established standard as to which method, modality, and analysis variant is preferable and there is only limited knowledge on the reproducibility, an important prerequisite for clinical application. We conducted an MEG-/ high-density (hd)-EEG-study on 22 young healthy adults, who were measured twice in a scan/rescan design after 7 – 2 days. Reliability of resting-state (15 min, eyes-closed) connectivity in source space was calculated via intraclass correlation coefficie…
