Mboat7 down-regulation by hyper-insulinemia induces fat accumulation in hepatocytes.

2020

Background: Naturally occurring variation in Membrane-bound O-acyltransferase domain-containing 7 (MBOAT7), encoding for an enzyme involved in phosphatidylinositol acyl-chain remodelling, has been associated with fatty liver and hepatic disorders. Here, we examined the relationship between hepatic Mboat7 down-regulation and fat accumulation. Methods: Hepatic MBOAT7 expression was surveyed in 119 obese individuals and in experimental models. MBOAT7 was acutely silenced by antisense oligonucleotides in C57Bl/6 mice, and by CRISPR/Cas9 in HepG2 hepatocytes. Findings: In obese individuals, hepatic MBOAT7 mRNA decreased from normal liver to steatohepatitis, independently of diabetes, inflammatio…

Selection of potential probiotic lactobacilli from pig feces to be used as additives in pelleted feeding

2006

Thirty-five isolates from pig feces were identified as Lactobacillus reuteri (12 strains), Lactobacillus mucosae (7), Lactobacillus plantarum (6), Lactobacillus kitasatonis (3), Lactobacillus rossiae (2), Lactobacillus ultunensis (2), Lactobacillus crispatus (2), and Lactobacillus intestinalis (1) by partial sequence analysis of the 16S rRNA. All isolates were detected at 8-9 log CFU g(-1). Preliminarily, strains were selected based on resistance to heat treatments (ca. 70 degrees C for 10 s). The decrease in viability for some L. reuteri, L. mucosae, L. plantarum, L. kitasatonis, and L. rossiae strains was lower than 1 log cycle. Selected strains were further characterized for acid and bil…

Citrus biotechnology

2020

Agricultural crops that can better withstand the changing climatic and pathogen landscape have been produced through natural selection throughout the millennia and, in recent years, through the process of human-assisted plant breeding and selection. However, a lack of genetic diversity in many commercially cultivated crops (due to monoculture) has made them more vulnerable to biotic and abiotic stresses (Esquinas-Alcazar, 2005; Keneni et al., 2012). Citrus, belonging to the Rutaceae family, is one of the most important commercial woody fruit crops in the tropical and subtropical areas of the world. The origin of citrus is traced back to parts of tropical and subtropical Southeast Asia (Wu e…

Fruit quality of Sicilian, commercially affirmed, and newly selected apple varieties: potential resources for genetic improvement and specialty marke…

2012

Several apple varieties are constantly selected for improved quality traits and introduced for cultivation and marketing in addition to a few traditional and affirmed varieties. Recently, specific fruit quality traits like enhanced flavor, crispness and juiciness have been particularly requested by consumers and are gradually replacing more conventional characteristics like fruit size, peel color and sweetness in the breeding programs. Furthermore, some of the newly selected varieties exhibit a relatively low level of plasticity, posing serious problems of adaptation when cultivation is proposed in a wide range of environments. In this most recent perspective, some of the old varieties or l…

Chi dice che i batteri fanno male? I CRISPR ed il loro potenziale genetico. In Le scienze naturali nella scuola

2019

In definitiva è molto p…

Investigating CRISPR-CAS13b as a tool for the RNA editing of CFTR mRNA with premature stop codon

2020

Background and Rationale Some CF patients are compound heterozygous or homozygous for nonsense mutations in the CFTR gene. Mutant CFTR gene coding for transcripts with premature termination codons (PTCs) is responsible for truncated CFTR protein and for a severe form of the disease. In a precision medicine framework the “REPAIRv2” (RNA Editing for Programmable A to I Replacement v2) tool, developed in the laboratory of Dr. Feng Zhang (USA), seems a good alternative to restore the full-length CFTR protein by editing its mRNA containing PTCs. This new approach is based on the possibility of targeting a deaminase enzyme (huADAR2) to a specific Adenosine, to be edited to Inosine (G analogue), o…

Giovanni Maurigi, un allievo illustre di Emerico Amari. Storia e politica nella Sicilia risorgimentale

2011

Giovanni Maurigi (1823-1881) was a very well known Sicilian jurist. Student of philosopher Emerico Amari, his maestro's intellectual stimulus led him to contribute to the 1848 risings in Palermo. He was a well known laywer during the Borbonic regime, who defended patriots persecuted by the government. After Italian unification he became a judge. A liberal, his own professional career can be interpreted as an example of Sicilian elite recruiting in the new unified state.
