Human noroviruses (HuNoVs) are a prominent cause of gastroenteritis, yet fundamental questions remain regarding epidemiology, diversity and immunity in sub-Saharan African children. We investigated HuNoV seroprevalence and genetic and sociodemographic risk factors in Ugandan children. We randomly screened 797 participants of a longitudinal birth cohort (EMaBS) in Entebbe, and 378 children from a cross-sectional survey of rural island communities in Lake Victoria (LaVISSWA), for antibodies against multiple HuNoV genotypes by ELISA. We used linear regression modelling to test for associations between HuNoV antibody levels and sociodemographic factors, and for associations with the known human…

Insulin Dissociates the Effects of Liver X Receptor on Lipogenesis, Endoplasmic Reticulum Stress, and Inflammation

2016

IF 4.258; International audience; Diabetes is characterized by increased lipogenesis as well as increased endoplasmic reticulum (ER) stress and inflammation. The nuclear hormone receptor liver X receptor (LXR) is induced by insulin and is a key regulator of lipid metabolism. It promotes lipogenesis and cholesterol efflux, but suppresses endoplasmic reticulum stress and inflammation. The goal of these studies was to dissect the effects of insulin on LXR action. We used antisense oligonucleotides to knock down Lxr alpha in mice with hepatocytespecific deletion of the insulin receptor and their controls. We found, surprisingly, that knock-out of the insulin receptor and knockdown of Lxr alpha …

Development of a Sensitive Detection Method for Alphaviruses and Its Use as a Virus Neutralization Assay

2021

Alphaviruses have a single-stranded, positive-sense RNA genome that contains two open reading frames encoding either the non-structural or the structural genes. Upon infection, the genomic RNA is translated into the non-structural proteins (nsPs). NsPs are required for viral RNA replication and transcription driven from the subgenomic promoter (sgP). Transfection of an RNA encoding the luciferase gene under the control of the sgP into cells enabled the detection of replication-competent chikungunya virus (CHIKV) or Mayaro virus (MAYV) with high sensitivity as a function of the induced luciferase activity. This assay principle was additionally used to analyze virus-neutralizing antibodies in…

Synthesis and cytotoxic activity of new artemisinin hybrid molecules against human leukemia cells

2017

A series of new artemisinin-derived hybrids which incorporate cholic acid moieties have been synthesized and evaluated for their antileukemic activity against sensitive CCRF-CEM and multidrug-resistant CEM/ADR5000 cells. The new hybrids 20-28 showed IC50 values in the range of 0.019µM-0.192µM against CCRF-CEM cells and between 0.345µM and 7.159µM against CEM/ADR5000 cells. Amide hybrid 25 proved the most active compound against both CCRF-CEM and CEM/ADR5000 cells with IC50 value of 0.019±0.001µM and 0.345±0.031µM, respectively. A relatively low cross resistance to hybrids 20-28 in the range of 5.7-fold to 46.1-fold was measured. CEM/ADR5000 cells showed higher resistance than CCRF-CEM to al…

Multidisciplinary oral rehabilitation in partially edentulous adult patients with malocclusion: a cross-sectional survey study

2018

Background A cross-sectional survey was conducted to gather information regarding the opinion of Brazilian specialists in both orthodontics and implantology on multidisciplinary oral rehabilitation in partially edentulous patients with malocclusion. Material and methods A total of 305 specialists participated in a telephone survey and answered an ad hoc 10-item questionnaire, including the request of total skull cone-beam tomographies (CBCT) and the use of 3D digital planning software, the best moment of treatment to place dental implants, and the integration of orthodontics in implantology. Results Most participants did not request CBCT (90.8%) or 3D digital planning software images (92.3%…

Thermostabilization of BSA in TMAO Water Mixtures by Infrared Spectroscopy

2019

Background:Trimethylamine-N-Oxide (TMAO) is a small organic molecule derived from the metabolism of L-carnitine and choline after ingestion of animal food. TMAO has many functions such as electron acceptor, an osmolyte, stabilizer of macromolecules folding. It seems that TMAO plays an important role in nature but, in humans, it is a remnant of the evolution of the osmolyte system. Objective:The present paper is addressed on the study of thermal stability of hydrated Bovine Serum Albumins (BSA) in the presence of water and TMAO water solution by means of InfraRed spectroscopy. In particular, this work has investigated the protein amide I spectral regions, which is sensitive to protein seconda…

Maternal Perfluoroalkyl Substances, Thyroid Hormones, and DIO Genes: A Spanish Cross-sectional Study

2021

Results of studies on perfluoroalkyl substances (PFASs) and thyroid hormones (THs) are heterogeneous, and the mechanisms underlying the action of PFASs to target THs have not been fully characterized. We examined the relation between first-trimester maternal PFAS and TH levels and the role played by polymorphisms in the iodothyronine deiodinase 1 (DIO1) and 2 (DIO2) genes in this association. Our sample comprised 919 pregnant Spanish women (recruitment = 2003-2008) with measurements of perfluorohexanesulfonic acid (PFHxS), perfluorooctanoic acid (PFOA), perfluorooctanesulfonic acid (PFOS), perfluorononanoic acid (PFNA), thyroid-stimulating hormone (TSH), total triiodothyronine (TT3), and fr…

Gross motor proficiency and intellectual functioning

2018

This cross-sectional study examines differences in gross motor proficiency as a function of different intellectual functioning profiles. Two motor areas have been investigated as being equally essential to gross motor functions in every-day life: locomotion and object control. It aims to compare gross motor skills endorsed by children with Down syndrome (DS), children with borderline intellectual functioning (BIF), and typically developing children (TDC). Group 1 was composed of 18 children with DS (chronological age=8.22), group 2 was composed of 18 children with BIF (chronological age=9.32), and group 3 was composed of 18 children with typical development (TD) (chronological age=9.28). A cross-sectional study of pre-service teachers’ views about disability and attitudes towards inclusive education

2016

Teachers’ attitudes towards students with disabilities and special educational needs (SEN) and their inclusion in regular education classrooms have been internationally identified as a key factor in the implementation of inclusive education. In this study, 501 participants representing a cross-section of pre-service teachers from three public colleges of education in Ghana were surveyed about their views regarding disability, level of discomfort interacting with people with disabilities and attitudes towards inclusive education. The results indicate that the pre-service teachers understood disability as an interaction between biological and environmental factors and felt comfortable interac…
