Salix tarraconensis Pau ex Font Quer, an endemic willow species from northeast Spain, was micropropagated with nodal segments. Shoot multiplication was obtained with different cytokinins, either on Murashige and Skoog medium or woody plant medium. Best results for shoot formation were obtained on Murashige and Skoog medium containing 4.9 μM of 6-γ-γ-dimethylallylaminopurine. Shoots showed strong apical dominance, and some cultures displayed apical necrosis. Benzyladenine gave the worst results; shoots displayed very slow growth, deformed leaves, and hyperhydrity. Evaluation of some growth regulator effects on encapsulated in vitro-derived microcuttings of three Italian Ficus carica L. genotypes.

In this study, the encapsulation technology based on the calcium alginate coating was applied to some Ficus carica L. genotypes. Uninodal microcuttings (3-4 mm long), excised from in vitro proliferating shoots of three Italian cultivars (‘Bifera’, ‘Palazzo’ and ‘Catalanisca’), were employed. The influence of three different plant growth regulators (PGRs): 6-benzylaminopurine (BAP), meta-topolin (MT) and zeatine (ZEA), added to the artificial endosperm, were evaluated. Morphogenesis in Root Tip Meristem Cultures of Digitalis obscura L.

Summary The morphogenic capacity of Digitalis obscura L. root tip meristems cultured in vitro has been studied, noting hormonal factors inducing differentiation and development of shoots as well as those promoting somatic embryogenesis. Caulogenesis was obtained with different auxin/cytokinin combinations but a high rate of shoot regeneration was induced by 0.1 or 0.5 ppm IAA and 1.0 ppm BA. Different stages of somatic embryo development were observed in media supplemented with auxins or high auxin/cytokinin ratios, although no complete plant regeneration was achieved. This is the first report known of shoot regeneration from root tips of Digitalis obscura L.

Potential use of new diphenylurea derivatives in micropropagation of Capparis spinosa L.

A protocol for in vitro multiplication of caper (Capparis spinosa L. subsp. rupestris) from nodal segments collected from mature plants was developed. For shoot multiplication, one auxin (indol-3-butyric acid, IBA) and cytokinins of two different classes were used: the N6-substituted adenine derivatives 6-benzylamino purine (BAP), and the two synthetic phenylurea derivatives N-phenyl-N′-benzothiazol-6-ylurea (PBU) and N-phenyl-N′-(1,2,3-thidiazol-5-yl) urea (thidiazuron, TDZ). In vitro plant regeneration of caper (Capparis spinosa L.) from floral explants and genetic stability of regenerants

A new technique to regenerate caper plants (Capparis spinosa L. subsp. rupestris) starting from flower explant is reported. In vitro plant regeneration was attempted using stigma, anthers and unfertilized ovules of unopened flowers collected in the field. Plant regeneration was achieved from unfertilized ovules on MS medium supplemented with 88 mM sucrose and 13 lM 6-benzyladenine (BA). Explants taken from the leaves of yams (Dioscorea bulbifera L.) at different stages of development were cultured in vitro on a checkerboard using various combinations and/or concentrations of auxin (2,4-d) and cytokinin (6-BAP). An addition of cytokinin to the culture media was not essential for callus induction from explants derived from young leaves in the very early stages of expansion. In vitro morphogenesis of grapevine (Vitis vinifera L.) inflorescence primordia, cvs Pinot Noir and Chardonnay

Hormonal regulatory processes controlling in vitro morphogenesis of grapevine inflorescence primordia, excised from latent buds from November to April, were investigated. Cytokinin (benzylaminopurine, BAP), gibberellin (GA3) and auxin (3-indolebutyric acid, IBA) were used at 1 mg/L. BAP induced branch development and flower calyx differentiation. This study was conducted to micropropagate 'Brown Turkey' fig, one of the most popular cultivars in California, USA for fresh and dried uses. Nodal segments cut lengthwise or not were cultured on Woody Plant Medium containing 0.3, 1.0 or 3.0 μM of N6 benzyladenine (BA) or kinetin in a 3 (concentrations) × 2 (cytokinins) × 2 (explant types) factorial experiment. 60% of explants became contaminated with various microbes when the standard protocol of washing shoots for 3 h under running tap water.
