Layered composite based on halloysite and natural polymers: a carrier for the pH controlled release of drugs

2019

We have prepared new biohybrid materials based on halloysite nanotubes and natural polymers (alginate and chitosan) for the controlled and sustained release of bioactive species. A functional nanoarchitecture has been designed allowing us to generate a layered tablet with a chitosan/halloysite nanocomposite film sandwiched between two alginate layers. The assembly of the raw components and the final structure of the hybrid tablet have been highlighted by the morphological and wettability properties of the prepared materials. Since the biohybrid has been designed as a smart carrier, halloysite nanotubes have been first loaded with a model drug (sodium diclofenac). The effect of the tablet th…

Intestinal absorption enhancement via the paracellular route by fatty acids, chitosans and others: a target for drug delivery.

2005

Peroral delivery of hydrophilic drugs is one of the greatest challenges in biopharmaceutical research. Hydrophilic drugs usually present low bioavailability after oral administration. One of the causes of this low bioavailability is their poor intestinal permeation through the paracellular pathway. This pathway is actually restricted by the presence of tight junctions at the apical side of the enterocytes. In the last few years, great interest has been focused on the structure and cellular regulation of tight junctions, materializing in more in-depth knowledge of this intestinal barrier. Simultaneously, and on the basis of this understanding, continuous efforts are being made to develop age…

Spray Freeze Dried Lyospheres® for Nasal Administration of Insulin

2021

Pharmacologically active macromolecules, such as peptides, are still a major challenge in terms of designing a delivery system for their transport across absorption barriers and at the same time provide sufficiently high long-term stability. Spray freeze dried (SFD) lyospheres® are proposed here as an alternative for the preparation of fast dissolving porous particles for nasal administration of insulin. Insulin solutions containing mannitol and polyvinylpyrrolidone complemented with permeation enhancing excipients (sodium taurocholate or cyclodextrins) were sprayed into a cooled spray tower, followed by vacuum freeze drying. Final porous particles were highly spherical and mean diameters r…

In Vitro and In Silico Studies of Two 1,4-Naphthoquinones and Their Topical Formulation in Bigels.

2021

Background: 1,4-Naphthoquinones (1,4-NQs) are secondary plant metabolites with numerous biological activities. 1,4-NQs display low water solubility and poor bioavailability. Bigels are a new technology with great potential, which are designated as drug delivery systems. Biphasic bigels consisting of solid and liquid components represent suitable formulations improving diffusion and bioavailability of NQs into the skin. Objective: We evaluated the in silico and in vitro activity of 5,8-dihydroxy-1,4-naphthoquinone (M1) and 2,3-dichloro-5,8-dihydroxy-1,4-naphthoquinone (M2) on elastase and assessed their cytotoxicity towards COLO38 melanoma cells. The 1,4-NQs were loaded into bigels for topi…

Clay-biosurfactant materials as functional drug delivery systems: Slowing down effect in the in vitro release of cinnamic acid

2017

Abstract The main objectives of the present paper were the preparation and characterization of new surfactant-modified clays and the evaluation of their potential applicability as drug delivery systems for the oral administration of the cinnamic acid (CA) drug. The organoclays (OC) were prepared by loading different amounts of the biocompatible nonionic polyoxyethylene sorbitan monolaurate surfactant (Tween20) onto K10 montmorillonite (Mt) clay and characterized through the construction of the adsorption isotherms by means of the spectrophotometric method. The performance of the prepared material was verified by gathering the adsorption isotherms of the cinnamic acid onto the Mt/Tween20 org…

Multicomponent solid dispersion as a formulation strategy to improve drug permeation: A case study on the anti-colorectal cancer irinotecan

2019

Abstract Multicomponent solid dispersions (MSD)s are frequently proposed as efficient drug delivery systems to improve drug solubility and bioavailability. In this study, the effects of specific excipients, such as mannitol, inulin, poly(methyl methacrylate-co-methacrylic)acid (PMMA) and cellulose acetate phthalate (CAP) have been tested to potentially improve irinotecan (IRN) permeation in the intestinal tract with the intention to protect the drug from the gastric environment. MSDs were formulated as microparticles by Spray-Drying technique. Raw materials and microparticles have been characterized by FTIR analysis to determine hydrogen bonding. SEM images were recorded to investigate morp…

Supercritical Fluid Adsorption of Domperidone on Silica Aerogel

2013

Silica aerogel (SA) was loaded with domperidone to demonstrate the potentiality of adsorption processes based on the usage of supercritical carbon dioxide to treat poorly water-soluble drugs, forming new kinds of drug delivery systems. The effects of pressure, temperature and solution concentration on loaded SA were studied. Adsorption isotherms were measured at 35℃ and 45℃ and fitted with Langmuir model. Release kinetics of the adsorbed drug were also evaluated by in vitro dissolution tests. Results showed that domperidone can be uniformly dispersed into the aerogel and that the release rate of domperidone from the composite, constituted by drug and silica aerogel, is much faster than that…

Fostering Collaborative Governance in Chronic Disease Management Programs: A Dynamic Performance Management Approach

2020

Chronic diseases are the leading cause of disability and mortality in the world and represent a global health emergency due to the increase in frequency and complexity that has been occurring in recent years. The outcomes related to chronic care needs depend on the joint effort of a multi-provider, multi-disciplinary, and multi-professional service network, which operates along a clinical pathway. However, all the different players involved in the provision of services may have different interests and goals derived from their organizational structure and their role in the overall health system. This context of fragmented governance makes performance management of care services problematic. …

Towards Next Generation System Architecture for Emergency Services

2011

European Union has decided that all emergencies can be reported to authorities by European citizens by calling 112 or sending a text message to 112. Distributing warnings and alerts of authorities to citizens currently happens through national TV and radio channels, but telecom networks are also used now in some countries for this purpose. During the last ten years there have been attempts to develop new system architectures for various emergency service provision phases. Some of them are already deployed in special cases, like in ambulance services. Multimedia emergency alert messages can be delivered to citizens over fixed and mobile telecom networks and commercial systems are already in …

Global trends in a fragile context : public–nonpublic collaboration, service delivery and social innovation

2020

PurposeThis study aims to enhance the understanding of the nature of collaboration between public and nonpublic actors in delivering social services and achieving social innovation in a fragile context, with an emphasis on the role of civil society organisations (CSOs). The paper focuses on Bosnia and Herzegovina, a Southeastern European country which has faced a turbulent post-conflict transition and experienced challenges in its social welfare policy and practice.Design/methodology/approachThis study uses institutional theory, particularly new institutionalism and institutional networking, as a lens through which to understand public and nonpublic collaboration and social innovation withi…
