This chapter discusses processes in which enolates are subjected to reaction with alkylating reagents. In all the processes examined here, at least one of the two reaction components is chiral and the reaction gives rise to one or more diastereomeric compounds. In most cases, the enolate is generated through deprotonation of the corresponding carbonyl precursor with a suitable base, but in several cases, the enolate is generated differently, for example, through Michael addition of a nucleophile to a conjugated carbonyl compound. The utility of the derivatization reagento-phthaldialdehyde (OPA) in combination with the chiral thiol N-acetyl-L-cysteine (NAC) for the enantioselective analysis of primary amphetamines and structurally related substances has been investigated. The diastereomers obtained have been chromatographed on an achiral, reversed phase C18 column (125 mm×4 mm i.d., 5 μm), and detected fluorimetrically. The influence of parameters affecting the chromatographic separation as well as the derivatization reaction has been optimized, using amphetamine, norephedrine, norepinephrine and 3,4-methylenedioxyamphetamine (MDA) as model compounds. The major diastereomer formed in the Barbier-type metal-mediated allylation of D-mannose has previously been shown to adopt a perfectly linear conformation, both in solid state and in solution, resulting in the formation of hydrogen-bonded networks and subsequent aggregation from aqueous solution upon stirring. Here, a comprehensive study of the solid state structure of both the allylated D-mannose and its racemic form has been conducted. Abstract Starting from the 1′- or 2′-phenyl-substituted 1-(2′-hydroxyethyl)-6,7-dimethoxy-1,2,3,4-tetrahydroisoquinoline diastereomers 3 and 6 , 4-unsubstituted and 4-(p-nitrophenyl)- and 4-oxo-substituted 1-phenyl- and 2-phenyl-9,10-dimethoxy-2H,4H-1,6,7,11b-tetrahydro-1,3-oxazino[4,3-a]isoquinolines ( 7–12 ) were prepared. The relative configurations and the predominant conformations of the products were determined by NMR spectroscopy, by quantum chemical calculations and, for (2R∗,4S∗,11bR∗)-9,10-dimethoxy-4-(p-nitrophenyl)-2-phenyl-2H,4H-1,6,7,11b-tetrahydro-1,3-oxazino[4,3-a]isoquinoline ( 11 ), by X-ray diffraction.

Abstract Cycloaddition reactions between 2-(1-cyanoallyl)thiophen and the dienophiles diethyl azodicarboxylate, N-phenylmaleimide , methyl propiolate, and dimethyl acetylenedicarboxylate are reported. Products include simple benzo[b] thiophen carboxylates (13,19) and reduced derivatives (8,9,10,12,18), as a mixture of diastereoisomers, except in the adduct with diethyl azodicarboxylate. With dimethyl acetylenedicarboxylate a new type of tricyclic compound was also found (20).

We have investigated several 1,3-dipolar cycloadditions of a chiral nitrone prepared from L-erythrulose. While cycloadditions to carbon-carbon multiple bonds of dipolarophiles such as ethyl acrylate, ethyl propiolate, or dimethyl acetylenedicarboxylate were poorly stereoselective, reaction with acrylonitrile provided predominantly one diastereomeric adduct. Furthermore, the regioselectivity exhibited by the two structurally similar dipolarophiles ethyl acrylate and ethyl propiolate was found to be opposite. [EN] In this paper, we report the application of chiral complexes of La(III) with pyBOX ligands as Lewis acid catalysts in the conjugate addition of malonic esters to N-tosyl imines derived from ß,gamma-unsaturated alfa-keto esters to give the corresponding chiral alfa,ß-dehydroamino esters. pyBOX complexes with La(III), Yb(III), Sc(III), and In(III) triflates were assessed in this reaction but only La(III) showed good activity and enantioselectivity, while Yb(III) provided the expected product with low yield and stereoselectivity, and the Sc(III) and In(III) complexes were completely inactive. Abstract The enantiomeric and diastereomeric profiling of chiral pharmaceuticals (ephedrine, norephedrine, atenolol and venlafaxine) and illicit drugs (amphetamine, methamphetamine, 3,4-methylenedioxyamphetamine (MDA), 3,4-methylenedioxy- N -methylamphetamine (MDMA) and 3,4-methylenedioxy- N -ethylamphetamine (MDEA)) was undertaken over a period of fourteen consecutive days in three wastewater treatment plants (WWTPs) in the city of Valencia, Spain. Degradation efficiency of WWTPs was found to be compound and enantiomer dependent. Selective enantiomer enrichment was observed for several target analytes. Natural products are an excellent source of inspiration for the development of new drugs. Among them, betalains have been extensively studied for their antioxidant properties and potential application as natural food dyes. Herein, we describe the seven-step synthesis of new betalamic acid analogs without carboxy groups in the 2- and 6-position with an overall yield of ~70%. The Folin–Ciocalteu assay was used to determine the antioxidant properties of protected intermediate 21. Additionally, the five-step synthesis of betalamic acid analog 35 with three ester moieties was performed. Using NMR techniques, the stability of the obtained compounds towards oxygen was analyzed.
