The most commonly used drugs are analgesics, mainlyacetaminophen (paracetamol, APAP) and, to a lesser extent, ketoprofen (KPF). International audience; Myotonic dystrophy type 1 (DM1) is caused by an unstable expanded CTG repeat located within the DMPK gene 3'UTR. The nature, severity and age at onset of DM1 symptoms are very variable in patients. Different forms of the disease are described, among which the congenital form (CDM) is the most severe. Molecular mechanisms of DM1 are well characterized for the adult form and involve accumulation of mutant DMPK RNA forming foci in the nucleus. Investigations into the distribution of subchondral bone density in the human elbow have suggested that the geometry of the trochlear notch deviates from a perfect fit with the trochlea, and that the load is transmitted ventrally and dorsally rather than through the centre of the humero-ulnar joint. We therefore decided to make a quantitative assessment of the degree of incongruity between the two components in 15 human specimens (age distribution 60 to 93 years) with different types of joint surface. Type V collagen counteracts osteo-differentiation of human mesenchymal stem cells

In search of novel gene signatures for osteo-differentiation of mesenchymal stem cells (MSCs), we submitted cDNA preparations from undifferentiated and differentiating MSCs to differential display- and semiquantitative-PCR and found down-regulation of COL5A1 in osteo-induced cultures at days 21 and 28, when the mineralized matrix accumulates. We also cultured osteo-differentiating MSCs onto type V collagen substrates and found a decrease in the accumulation of extracellular calcium compared to those grown in uncoated flasks. To our knowledge, this is first evidence that type V collagen might represent a stromal component that impairs osteogenesis.
The effects of post-translational processing on dystroglycan synthesis and trafficking1

Dystroglycan is a component of the dystrophin glycoprotein complex that is cleaved into two polypeptides by an unidentified protease. To determine the role of post-translational processing on dystroglycan synthesis and trafficking we expressed the dystroglycan precursor and mutants thereof in a heterologous system. A point mutant in the processing site, S655A, prevented proteolytic cleavage but had no effect upon the surface localisation of dystroglycan. Mutation of two N-linked glycosylation sites that flank the cleavage site inhibited proteolytic processing of the precursor. En esta tesis doctoral se muestra la conversión directa de fibroblastos embrionarios de ratón a células con morfología y fenotipo similares al hepatocito mediante la expresión retroviral de seis factores de transcripción hepáticos, Hhex, Gata4, Foxa2, HNF4α HNF1α y HNF6. Los fibroblastos infectados mantenidos durante 3-4 semanas en cultivo adquieren una morfología epitelial y expresan genes típicamente hepáticos como Albumina, Cyp7a1, Tat o Slc10a1 a niveles próximos al hepatocito. Durante la manipulación de los fibroblastos mediante la expresión de los factores de transcripción ya mencionados, observamos que el factor de transcripción Gata4 es capaz de bloquear la reprogramación a células …
