‘Green’ Wine through a Responsible and Efficient Production: A Case Study of a Sustainable Sicilian Wine Producer

2016

Abstract Sustainability is progressively gaining importance in the winegrowing sector. Implementing this concept implies environmental soundness, social equity and economic feasibility. A proliferation of initiatives to develop the sustainable production of wine started officially in Italy since the year 2010. The objective of this paper is to illustrate the reshaping of Sicilian winegrowing according to the ‘Triple bottom line’ approach to sustainability. By analyzing the case-study of the Tasca d’Almerita firm, pilot farm of various national projects in wine-sustainability, we found that adopting sustainable operative, organizational and competitive strategies has lead to a well-managed a…

Large Blooms of Bacillales (Firmicutes) Underlie the Response to Wetting of Cyanobacterial Biocrusts at Various Stages of Maturity.

2018

ABSTRACT Biological soil crusts (biocrusts) account for a substantial portion of primary production in dryland ecosystems. They successionally mature to deliver a suite of ecosystem services, such as carbon sequestration, water retention and nutrient cycling, and climate regulation. Biocrust assemblages are extremely well adapted to survive desiccation and to rapidly take advantage of the periodic precipitation events typical of arid ecosystems. Here we focus on the wetting response of incipient cyanobacterial crusts as they mature from “light” to “dark.” We sampled a cyanobacterial biocrust chronosequence before (dry) and temporally following a controlled wetting event and used high-throug…

Thermophilic anaerobic conversion of raw microalgae: Microbial community diversity in high solids retention systems

2019

[EN] The potential of microbial communities for efficient anaerobic conversion of raw microalgae was evaluated in this work. A long-term operated thermophilic digester was fed with three different Organic Loading Rates (OLR) (0.2, 0.3 and 0.4¿g·L¿1·d¿1) reaching 32¿41% biodegradability values. The microbial community analysis revealed a remarkable presence of microorganisms that exhibit high hydrolytic capabilities such as Thermotogae (~44.5%), Firmicutes (~17.6%) and Dictyoglomi, Aminicenantes, Atribacteria and Planctomycetes (below ~5.5%) phyla. The suggested metabolic role of these phyla highlights the importance of protein hydrolysis and fermentation when only degrading microalgae. The …

The seed endosphere of Anadenanthera colubrina is inhabited by a complex microbiota, including Methylobacterium spp. and Staphylococcus spp. with pot…

2018

Background and aims: Plant seeds are emerging micro–habitats, whose importance as reservoir and vector of beneficial microbes just begins to be recognized. Here we aimed to characterize the bacterial microbiota of the Anadenanthera colubrina seed endosphere, with special focus to beneficial traits and to the colonization pattern. Methods: Cultivation–dependent (isolation from surface–sterilized seeds) and cultivation–independent (pyrosequencing of 16S rRNA gene from metagenomic seed DNA) analyses, functional tests and microscopical investigations (fluorescence in situ hybridization coupled with confocal laser scanning microscopy (FISH-CLSM) were performed. Results: We isolated several Methy…

Complete Genome Sequence of a New Firmicutes Species Isolated from Anaerobic Biomass Hydrolysis

2017

ABSTRACT A new Firmicutes isolate, strain HV4-6-A5C, was obtained from the hydrolysis stage of a mesophilic and anaerobic two-stage lab-scale leach-bed system for biomethanation of fresh grass. It is assumed that the bacterial isolate contributes to plant biomass degradation. Here, we report a draft annotated genome sequence of this organism.

Microbial community characterization during anaerobic digestion of Scenedesmus spp. under mesophilic and thermophilic conditions

2017

[EN] Microbial communities were thoroughly characterized in a mesophilic anaerobic membrane bioreactor (AnMBR) and a thermophilic continuous stirred tank reactor (CSTR), which were both treating recalcitrant microalgal biomass dominated by Scenedesmus. 16S rRNA amplicon sequencing analysis was performed when the AnMBR achieved 70% algal biodegradation and revealed high microbial diversity, probably due to the high solid retention time (SRT) of the AnMBR configuration. The bacterial community consisted of Chloroflexi (27.9%), WWE1 (19.0%) and Proteobacteria (15.4%) as the major phyla, followed by Spirochaetes (7.7%), Bacteroidetes (5.9%) and Firmicutes (3.6%). These phyla are known to exhibi…

Prokaryotic assemblages within permafrost active layer at Edmonson Point (Northern Victoria Land, Antarctica)

2018

This study was aimed at gaining insights on the prokaryotic community (in terms of both taxonomic composition and activities) inhabiting the active layer at Edmonson Point, an ice-free area on the eastern slope at the foot of Mount Melbourne (Northern Victoria Land, Antarctica). Samples were collected during the thawing period, when microbial physiological activities are restored to utilize previously frozen organic substrates. Despite the very small cell sizes (600 daltons) substrates, as indicated also by the obtained rates of enzymatic hydrolytic activities over proteolytic, glycolitic and phosphoric compounds. Taxonomical composition showed that Proteobacteria, Actinobacteria and Firmic…

Sensing of O 2 and nitrate by bacteria: alternative strategies for transcriptional regulation of nitrate respiration by O 2 and nitrate

2020

Many bacteria are able to use O2 and nitrate as alternative electron acceptors for respiration. Strategies for regulation in response to O2 and nitrate can vary considerably. In the paradigmatic system of E. coli (and γ-proteobacteria), regulation by O2 and nitrate is established by the O2 -sensor FNR and the two-component system NarX-NarL (for nitrate regulation). Expression of narGHJI is regulated by the binding of FNR and NarL to the promoter. A similar strategy by individual regulation in response to O2 and nitrate is verified in many genera by the use of various types of regulators. Otherwise, in the soil bacteria Bacillus subtilis (Firmicutes) and Streptomyces (Actinobacteria), nitrat…

2020

The rapid developments in the next-generation sequencing methods in the recent years have provided a wealth of information on the community structures and functions of endophytic bacteria. However, the assembly processes of these communities in different plant tissues are still currently poorly understood, especially in wild plants in natural settings. The aim of this study was to compare the composition of endophytic bacterial communities in leaves and roots of arcto-alpine pioneer plant Oxyria digyna, and investigate, how plant tissue (leaf or root) or plant origin affect the community assembly. To address this, we planted micropropagated O. digyna plants with low bacterial load (bait pla…

Spanish Validation of the Polymorphous Prejudice Scale in a Sample of University Students

2016

ABSTRACTThe shortened version of the Polymorphous Prejudice Scale (PPS) analyzes new manifestations of prejudice toward gay men and lesbian women. Specifically, this instrument consists of 16 items distributed in four subscales: values gay progress, positive beliefs about gay men, positive beliefs about lesbian women, and resistance to heteronormative expectations. The aim of the current study is to add new evidence about the reliability and validity of the scale. The scale is administrated to 348 heterosexual university students from Spain with a mean age of 22.62 years (SD = 7.63). Reliability and factorial validity estimates are presented. A four-factor structure is supported using confi…
