To evaluate CD36 expression with quantum dots 605 (QDs 605) on untreated and 7-ketocholesterol (7KC)-treated monocytic U937 cells by flow cytometry (FCM) and confocal and multiphoton laser scanning microscopy (CLSM).Cells were analyzed by CLSM, following flow cytometric quantification of CD36 expression and 7KC uptake. Image sequences were obtained by spectral analysis in monophoton and multiphoton CLSM and analyzed by the factor analysis of medical image sequences (FAMIS) algorithm to differentiate emission spectra. In CLSM analysis, cell deposits were screened in ultraviolet excitation modes to optimize the possibilities of QDs 605 and have the benefit of nuclei counterstaining by DAPI.FC…

CD39 is highly involved in mediating the suppression activity of tumor-infiltrating CD8+ T regulatory lymphocytes

CD39 is an ectoenzyme, present on different immune cell subsets, which mediates immunosuppressive functions catalyzing ATP degradation. It is not known whether CD39 is expressed and implicated in the activity of CD8+ regulatory T lymphocytes (Treg). In this study, CD39 expression and function was analyzed in both CD8+ and CD4+CD25hi Treg from the peripheral blood of healthy donors as well as from tumor specimens. CD39 was found expressed by both CD8+ (from the majority of healthy donors and tumor patients) and CD4+CD25 hi Treg, and CD39 expression correlated with suppression activity mediated by CD8+ Treg. Multispectral and autofluorescence RGB imaging for skin cancer diagnostics

This paper presents the results of statistical clinical data, combining two diagnostic methods. A combination of two skin imaging methods – diffuse reflectance and autofluorescence – has been applied for skin cancer diagnostics. Autofluorescence (AF) and multispectral diffuse reflectance images were acquired by custom made prototype with 405 nm, 526 nm, 663 nm and 964 nm LEDs and RGB CMOS camera. Parameter p’ was calculated from diffuse reflectance images under green, red and infrared illumination, AF intensity (I’) was calculated from AF images exited at 405nm wavelength. Conformationally regulated fluorescent sensors. Study of the selectivity in Zn 2+ versus Cd 2+ sensing

Abstract The Zn 2+ and Cd 2+ complexing properties of four ligands containing a 4,4′-substituted biphenyl moiety are described. Ligands 1 and 3 , containing only one 1-aza-18-crown-6 cavity, lead to selective complexation of Cd 2+ versus Zn 2+ . Ligand 4 , with two crown cavities linked to a tetramethylbenzidine unit, is able to form 1:1 complexes with Zn 2+ and Cd 2+ , showing a higher complexing constant with Zn 2+ than with Cd 2+ , probably due to enthalpic factors. Glutamatergic projection from the nucleus incertus to the septohippocampal system

Abstract Recent findings support a relevant role of the nucleus incertus in the control of the hippocampal activity through the modulation of theta rhythm. Previous studies from our group have shown that this nucleus is a critical relay between reticularis pontis oralis and the medial septum/diagonal band, regarded as the main activator and the pacemaker of the hippocampal oscillations, respectively. Calcium/calmodulin-dependent protein kinases in the carotid body: an immunohistochemical study

We determined the presence of Ca(2+)/calmodulin-dependent protein kinases (CaMKs), a family of multifunctional proteins engaged in Ca(2+)-linked signaling, in carotid body chemoreceptor cells which are critical for the hypoxia-sensing. Carotid bodies were dissected from anesthetized normoxic adult Wistar rats and were double stained for individual CaMKs and for tyrosine hydroxylase (TH), a marker of chemoreceptor cells. Immunofluorescence was examined by confocal laser scanning microscopy. We found that CaMKI and CaMKII were expressed in chemoreceptor cells, but their distribution and intensity varied.
