Mechanism of action of sphingolipids and their metabolites in the toxicity of fumonisin B1.

Fumonisins are a group of mycotoxins produced primarily by Fusarium moniliforme. Several fumonisins have been isolated through out the years but only fumonisin B1, B2 and B3 are the ones present in naturally contaminated foods, with B1 being the most toxic between them. The structural similarity between sphinganine and fumonisin B1 suggests that the mechanism of action of this mycotoxin is mainly via disruption of sphingolipid metabolism, this is an important step in the cascade of events leading to altered cell growth, differentiation and cell injury. ABSTRACT When the fungal pathogen Gibberella moniliformis (anamorph, Fusarium verticillioides ) colonizes maize and maize-based products, it produces class B fumonisin (FB) mycotoxins, which are a significant threat to human and animal health. FB biosynthetic enzymes and accessory proteins are encoded by a set of clustered and cotranscribed genes collectively named FUM, whose molecular regulation is beginning to be unraveled by researchers. A sensitive and reliable method using pressurized liquid extraction (PLE) and liquid chromatography (LC)/electrospray ionization (ESI) tandem mass spectrometry with a triple quadrupole (QqQ) analyzer has been developed for the analysis of fumonisin B(1) (FB(1)), fumonisin B(2) (FB(2)) and fumonisin B(3) (FB(3)) in corn-based baby foods. Fusaproliferin, beauvericin and enniatins: occurrence in food – a review

The present review summarises data on the prevalence and concentration of fusaproliferin, beauvericin and enniatins in small grains, maize and processed grain-based foods. These fungal secondary metabolites possess interesting biological activities and are presently often referred to as 'emerging mycotoxins'. They have been considered as less important since they are likely not of acute toxicity. Fumonisin B1 is a mycotoxin produced by fungi of the genus Fusarium that frequently occurs on maize (Zea mays) and feeds containing it. The use of plant-based protein sources in feeds designed for aquaculture has increased due to their low costs compared with fishmeal. This trend has resulted in a global increase in feed formulations contaminated with mycotoxins, producing economical losses in aquaculture industry. Analysis of fumonisins in corn-based food by liquid chromatography with fluorescence and mass spectrometry detectors

Abstract The presented procedure involves an extraction with methanol–water, centrifugation and cleanup with immunoaffinity columns. A comparison study between fluorescence detector, mass spectrometry, and tandem mass spectrometry with a triple quadrupole (QqQ) analyzer using an electrospray ionisation interface for the determination of fumonisin B1 and B2 in corn-based products has been performed.
