The designer cytokine hyper-interleukin-6 is a potent activator of STAT3-dependent gene transcription in vivo and in vitro.

1999

Interleukin-6 (IL-6) triggers pivotal pathways in vivo. The designer protein hyper-IL-6 (H-IL-6) fuses the soluble IL-6 receptor (sIL-6R) through an intermediate linker with IL-6. The intracellular pathways that are triggered by H-IL-6 are not defined yet. Therefore, we studied the molecular mechanisms leading to H-IL-6-dependent gene activation. H-IL-6 stimulates haptoglobin mRNA expression in HepG2 cells, which is transcriptionally mediated as assessed by run-off experiments. The increase in haptoglobin gene transcription correlates with higher nuclear translocation of tyrosine-phosphorylated STAT3 and its DNA binding. As H-IL-6 stimulates STAT3-dependent gene transcription, we compared t…

Lifetime predictions of non-ionic and ionic biopolymers: kinetic studies by non-isothermal thermogravimetric analysis

2021

AbstractIn this paper, films based on sustainable polymers with variable charge have been investigated by non-isothermal thermogravimetry in order to predict their lifetime, which is a key parameter for their potential use in numerous technological and biomedical applications. Specifically, chitosan has been selected as positively charged biopolymer, while alginate has been chosen as negatively charged biopolymer. Among non-ionic polymers, methylcellulose has been investigated. Thermogravimetric measurements at variable heating rates (5, 10, 15 and 20 °C min−1) have been performed for all the polymers to study their degradation kinetics by using isoconversional procedures combined with ‘Mas…

Low Temperature H2S Removal with Metal-Doped Nanostructure ZnO Sorbents: Study of the Origin of Enhanced Reactivity in Cu-Containing Materials

2011

Sulfidation of pure and metal-doped ZnO nanostructure sorbents (M0.03Zn0.97O, M = Fe, Co, Ni, Cu) was studied in order to clarify the effect of metal on the transformation kinetics at 200−350 °C. The solids were prepared by coprecipitation from metal nitrate solution followed by calcination at 400 °C. Reaction with H2S was studied by thermal gravimetric analysis (TGA) using a gas mixture containing 0.2 vol % H2S in equimolar H2−N2. It was found that at 350 °C the TGA sulfidation profiles of all studied samples are similar, with the interface reaction being the main rate-determining step. After lowering the temperature to 250 °C the transformation of Cu0.03Zn0.97O continues to be controlled …

Experimental tests on Li-ceramic breeders for the helium cooled pebble bed (HCPB) blanket design

2003

The Helium Cooled Pebble Bed (HCPB) Test Blanket Module (TBM) to be tested in ITER (International Thermonuclear Experimental Reactor) Reactor foresees the utilization of Lithiate ceramics as Tritium breeder in form of pebble beds. Since 1998, ENEA has launched many experimental activities for the evaluation of the breeder thermomechanics and the interaction between the pebble beds and the prismatic steel containment walls. Main objectives of these activities are the measurement of the pebble bed effective thermal conductivity, the wall heat transfer coefficient, the pressure loads and deformations on the lateral walls and their dependency from the mechanical constraints. The paper presents …

Computer simulation of ferroelectric property changes in PLZT ceramics under neutron irradiation

2001

The response of ferroelectric materials to high energy irradiation is of great interest because of their possible application in radiation environments such as thermonuclear reactors. In the present work a physical model for the defect evolution in PLZT ceramics under neutron irradiation and annealing is proposed. The influence of the defect system on the ferroelectric properties of these materials has been investigated. Satisfactory agreement between the theoretical estimated oxygen defect concentration after irradiation and annealing and the experimentally determined polarization has been obtained.

Prospektive Machbarkeitsstudie zum Vergleich von Röntgenübersichtsaufnahme und Thorax-MRT in Atemanhaltetechnik am offenen Niederfeldgerät

2002

PURPOSE MR investigations using a breath-hold sequence at an open low-field MR had to be compared to chest X-rays in patients with a wide spectrum of cardio-thoracic pathologies. MATERIAL AND METHODS 114 patients and three volunteers who actually received a chest X-ray due to different indications underwent triplanar breath-hold (17 - 20 s) True-FISP sequence using a 0.2 T low-field MR (Siemens Magnetom Open, TR/TE/alpha: 7.3/3.5/80 degrees, SD: 10 mm, Pixel: 2.81 x 1.41 mm) a mean of 5.1 (+/- 8.2) days later. RESULTS Signal-to-noise ratio as basics for pattern recognition was 3.2 in nodule, 5.0 in infiltration, and 12.0 in effusion, and therefore True-FISP is usable for the detection of th…

JNK phosphorylation relieves HDAC3-dependent suppression of the transcriptional activity of c-Jun

2003

The AP-1 transcription factor c-Jun is a prototypical nuclear effector of the JNK signal transduction pathway. The integrity of JNK phosphorylation sites at serines 63/73 and at threonines 91/93 in c-Jun is essential for signal-dependent target gene activation. We show that c-Jun phosphorylation mediates dissociation of an inhibitory complex, which is associated with histone deacetylase 3 (HDAC3). The subsequent events that ultimately cause increased mRNA synthesis are independent of c-Jun phosphorylation and its interaction with JNK. These findings provide an 'activation by de-repression' model as an explanation for the stimulatory function of JNK on c-Jun.

Critical behavior of a tumor growth model: directed percolation with a mean-field flavor.

2012

We examine the critical behaviour of a lattice model of tumor growth where supplied nutrients are correlated with the distribution of tumor cells. Our results support the previous report (Ferreira et al., Phys. Rev. E 85, 010901 (2012)), which suggested that the critical behaviour of the model differs from the expected Directed Percolation (DP) universality class. Surprisingly, only some of the critical exponents (beta, alpha, nu_perp, and z) take non-DP values while some others (beta', nu_||, and spreading-dynamics exponents Theta, delta, z') remain very close to their DP counterparts. The obtained exponents satisfy the scaling relations beta=alpha*nu_||, beta'=delta*nu_||, and the general…

Fluctuation Methods To Study Protein Aggregation in Live Cells: Concanavalin A Oligomers Formation

2011

Prefibrillar oligomers of proteins are suspected to be the primary pathogenic agents in several neurodegenerative diseases. A key approach for elucidating the pathogenic mechanisms is to probe the existence of oligomers directly in living cells. In this work, we were able to monitor the process of aggregation of Concanavalin A in live cells. We used number and brightness analysis, two-color cross number and brightness analysis, and Raster image correlation spectroscopy to obtain the number of molecules, aggregation state, and diffusion coefficient as a function of time and cell location. We observed that binding of Concanavalin A to the membrane and the formation of small aggregates paralle…
