Non-structural proteins P17 and P33 are involved in the assembly of the internal membrane-containing virus PRD1.

2015

AbstractBacteriophage PRD1, which has been studied intensively at the structural and functional levels, still has some gene products with unknown functions and certain aspects of the PRD1 assembly process have remained unsolved. In this study, we demonstrate that the phage-encoded non-structural proteins P17 and P33, either individually or together, complement the defect in a temperature-sensitive GroES mutant of Escherichia coli for host growth and PRD1 propagation. Confocal microscopy of fluorescent fusion proteins revealed co-localisation between P33 and P17 as well as between P33 and the host chaperonin GroEL. A fluorescence recovery after photobleaching assay demonstrated that the diff…

The Slow Adoption Rate of Software Robotics in Accounting and Payroll Services and the Role of Resistance to Change in Innovation-Decision Process

2022

Robotic process automation (RPA) has by now for years been viewed as a disruptive innovation that will have a significant impact on accounting, HR and payroll services, and yet the rate of adopting the innovation has not reached a level anticipated in past predictions. As several elements have a negative impact on the organization’s rate of adopting RPA, passive resistance to change has a significant impact in the form of constant dithering. Resistance to change can emerge at any stage of the Innovation-Decision process and fluctuate throughout the continued adoption, causing wasted investments, capabilities and resources. peerReviewed

Expression and purification of polyhistidine-tagged firefly luciferase in insect cells

2001

The coleopteran firefly, Photinus pyralis, luciferase was produced in lepidopteran Trichoplusia ni insect cells using a baculovirus expression vector. The recombinant protein was equipped with a polyhistidine affinity tag at the carboxyl terminus and purified by immobilized metal-ion affinity chromatography in combination with an expanded bed adsorption system. This approach enabled an efficient, one-step purification protocol of a genetically modified luciferase with properties similar to those of the authentic counterpart. According to light emission measurements, the final yield of highly purified protein was 23 mg l−1 of cell culture. In addition, no specific interaction of interfering …

Current state and future perspectives of biochar applications in Finland

2018

The binding of G-protein to rod outer segment phospholipids at the nitrogen–water interface

1989

In the visual process, one photoexcited rhodopsin (R*) catalyzes the activation of hundreds of G-proteins. It remains to be determined whether G-protein and R* find one another by membrane surface diffusion of these components (diffusion model) or by diffusion of G-protein through the aqueous phase (hopping model). A monolayer of each main rod outer segment (ROS) phospholipid interacting with a subphase containing G-protein, has been used to simulate the interaction of G-protein with the cytoplasmic surface of discal membranes. The possible diffusion of G-protein through the aqueous phase was then measured by observing its adsorption–desorption in the monolayer of each main ROS phospholipi…
