The SF3B1 protein, part of the SF3b complex, recognizes the intron branch point sequence of precursor messenger RNA (pre-mRNA), thus contributing to splicing fidelity. SF3B1 is frequently mutated in cancer and is the target of distinct families of splicing modulators (SMs). Among these, H3B-8800 is of particular interest, as it induces preferential lethality in cancer cells bearing the frequent and highly pathogenic K700E SF3B1 mutation. The SWI/SNF subunits BRG1 affects alternative splicing by changing RNA binding factor interactions with RNA

AbstractBRG1 and BRM are ATPase core subunits of the human SWI/SNF chromatin remodelling complexes. The function of the SWI/SNF complexes in transcriptional initiation has been well studied, while a function in alternative splicing has only been studied for a few cases for BRM-containing SWI/SNF complexes. Here, we have expressed BRG1 in C33A cells, a BRG1 and BRM-deficient cell line, and we have analysed the effects on the transcriptome by RNA sequencing. Correcting the effect of stellar spots on ARIEL transmission spectra – II. The limb-darkening effect

This paper is part of an effort to correct the transmission spectra of a transiting planet orbiting an active star. In Paper I (Cracchiolo et al. 2020) we have demonstrated a methodology to minimize the potential bias induced by unocculted star spots on the transmission spectrum, assuming a spot model parameterized by filling factor and temperature. In this work we introduce the limb darkening effect, therefore the position of the spot in the stellar disk and the impact parameter of the transiting planet now play a key role. The flexible framework of Generalized Item Response Models is adopted here for controlling the sources of heterogeneity in the data structure that make evaluations across courses not directly comparable. The topoisomerase I inhibitor topotecan (TPT) is used in the therapy of different tumors including high-grade gliomas. We previously showed that TPT-induced apoptosis depends on p53 with p53 wild-type (wt) cells being more resistant because of p53-controlled degradation of topoisomerase I. Here, we show that p53-deficient (p53(-/-)) fibroblasts undergo excessive mitochondrial apoptosis featuring H2AX phosphorylation, Bcl-x(L) decline, cytochrome c release, caspase-9/-3/-2 activation, and cleavage of Bid.
