AbstractDuring early postnatal life, speed up of signal propagation through many central and peripheral neurons has been associated with an increase in axon diameter or/and myelination. Especially in unmyelinated axons postnatal adjustments of axonal membrane conductances is potentially a third mechanism but solid evidence is lacking. Here, we show that axonal action potential (AP) conduction velocity in theDrosophilagiant fiber (GF) interneuron, which is required for fast long-distance signal conduction through the escape circuit, is increased by 80% during the first day of adult life. Genetic manipulations indicate that this postnatal increase in AP conduction velocity in the unmyelinated…

Detecting Protein Aggregation on Cells Surface: Concanavalin A Oligomers Formation

2009

A number of neurodegenerative diseases involve protein aggregation and amyloid formation. Recently evidence has emerged indicating small-transient prefibrillar oligomers as the primary pathogenic agents. Noteworthy, strict analogies exist between the behaviour of cells in culture treated with misfolded non-pathogenic proteins and in pathologic conditions, this instance together with the observation that the oligomers and fibrils are characterised by common structural features suggest that common mechanisms for cytotoxicity could exists and have to be perused in common interactions involved in aggregation. We here report an experimental study on ConcanavalinA (ConA) aggregation and its effect…

Conjugated linoleic acid isomers in mitochondria

2002

The beneficial effects exerted by low amounts of conjugated linoleic acids (C222222237) suggest that CLA are maximally conserved and raise the question about their mitochondrial oxidizability. Cis-9,trans-11-C18:2 (CLA1) and trans-10,cis-12-C18:2 (CLA2) were compared to cis-9,cis-12-C18:2 (linoleic acid; LA) and cis-9-C16:1 (palmitoleic acid; PA), as substrates for total fatty acid (FA) oxidation and for the enzymatic steps required for the entry of FA into rat liver mitochondria. Oxygen consumption rate was lowest when CLA1 was used as a substrate with that on CLA2 being intermediate between it and the respiration on LA and PA. The order of the radiolabeled FA oxidation rate was PA >> LA >…

Fatty acid composition of french butters, with special emphasis on conjugated linoleic acid (CLA) isomers

2005

Conjugated linoleic acids (CLA) are fatty acids primarily found in dairy products. These fatty acids may have potential biological properties interesting for human health. Update of French food composition data banks was needed to determine CLA levels in French dairy products and to evaluate CLA intake by the French population. As part of this investigation, we analyzed 54 butters from local producers at different times of the year. The average CLA levels in butter were 0.45 g CLA/100 g butter in winter, 0.58 in spring, and 0.80 in summer. Regional variations of CLA levels in butters were observed from different producing areas in France. Butters from hill and mountain regions, and butters …

Association of liver steatosis with lipid oversecretion and hypotriglyceridaemia in C57BL/6j mice fed trans-10, cis-12-linoleic acid

2003

AbstractConjugated linoleic acids (CLA) have recently been recognized to reduce body fat and plasma lipids in some animals. This study demonstrated that the steatosis accompanying the fat loss induced by trans-10,cis-12-C18:2 (CLA2) and not cis-9,trans-11-C18:2 (CLA1) isomer in C57BL/6j mice was not due to an alteration of the liver lipoprotein production that was even increased. The 3-fold decrease in plasma triacylglycerol contents and the induction of mRNA expression of low-density lipoprotein receptors concomitantly observed in CLA2-fed mice suggested an increase in the lipoprotein clearance at the level of the liver itself. CLA1 feeding produced similar but attenuated effects on trigly…

Geometry of conjugated double bonds of CLA isomers in a commercial mixture and in their hepatic 20∶4 metabolites

1999

Rats were fed a fat-free diet for 2 wk. After this period, while maintaining the animals on the same diet, the rats were given intragastrically 180 mg per day of a mixture of conjugated linoleic acids (CLA) as triacylglycerols. Gas chromatography mass spectrometry (GC-MS) analyses of this mixture, as well as hydrazine reduction and GC-MS and GC-Fourier transform infrared analyses of the resulting monoenes, revealed the presence of two major isomers, the 9c,11t- and the 10t,12c-18:2 accompanied by smaller amounts of the 8t, 10c and the 11c, 13t-18:2 isomers. Minor quantities of cis,cis and trans, trans conjugated isomers also were detected. The total fatty acid methyl esters from the liver li…

An Interactive Framework for Offline Data-Driven Multiobjective Optimization

2020

We propose a framework for solving offline data-driven multiobjective optimization problems in an interactive manner. No new data becomes available when solving offline problems. We fit surrogate models to the data to enable optimization, which introduces uncertainty. The framework incorporates preference information from a decision maker in two aspects to direct the solution process. Firstly, the decision maker can guide the optimization by providing preferences for objectives. Secondly, the framework features a novel technique for the decision maker to also express preferences related to maximum acceptable uncertainty in the solutions as preferred ranges of uncertainty. In this way, the d…

A Note on Laws of Motion for Aggregate Distributions

2020

I derive the law of motion for the aggregate distribution directly from the laws of motion for the individuals’ states. By relying on concepts from measure theory, the derivation is concise and intuitive. I address random shocks both at the micro level and at the macro level. Micro-level shocks completely cancel at the aggregate level provided that a law of large numbers applies. Therefore, the law of motion for the aggregate distribution is a deterministic process in the absence of macro-level uncertainty. If there are macro-level risks, the law of motion for the aggregate distribution exhibits a stochastic component additionally. I illustrate the formalism in a model of wealth accumulatio…

Surrogate Measures of Safety at Roundabouts in AIMSUN and VISSIM Environment

2018

This paper addresses issues on road safety analysis through microscopic traffic simulation models. The Surrogate Safety Assessment Model (SSAM) was applied to read vehicle trajectory files generated by two micro-simulators and then calculate surrogate measures of safety. Since safety assessment of any road entity can provide different results based on the micro-simulator which is used, the main objective of this research was to estimate the safety performance of three roundabout layouts and compare the conflict events simulated by AIMSUN and VISSIM. The two micro-simulators were used to build the calibrated models of the roundabouts, each fitting the corresponding empirical capacity functio…

pH Feedback Lifecycles Programmed by Enzymatic Logic Gates Using Common Foods as Fuels

2021

Abstract Artificial temporal signaling systems, which mimic living out‐of‐equilibrium conditions, have made large progress. However, systems programmed by enzymatic reaction networks in multicomponent and unknown environments, and using biocompatible components remain a challenge. Herein, we demonstrate an approach to program temporal pH signals by enzymatic logic gates. They are realized by an enzymatic disaccharide‐to‐monosaccharide‐to‐sugar acid reaction cascade catalyzed by two metabolic chains: invertase‐glucose oxidase and β‐galactosidase‐glucose oxidase, respectively. Lifetimes of the transient pH signal can be programmed from less than 15 min to more than 1 day. We study enzymatic k…
