All-atom simulations disentangle the functional dynamics underlying gene maturation in the intron lariat spliceosome

The spliceosome (SPL) is a majestic macromolecular machinery composed of five small nuclear RNAs and hundreds of proteins. SPL removes noncoding introns from precursor messenger RNAs (pre-mRNAs) and ligates coding exons, giving rise to functional mRNAs. Building on the first SPL structure solved at near–atomic-level resolution, here we elucidate the functional dynamics of the intron lariat spliceosome (ILS) complex through multi-microsecond-long molecular-dynamics simulations of ∼1,000,000 atoms models. Bioequivalence of oral products and the biopharmaceutics classification system: science, regulation, and public policy.

The demonstration of bioequivalence (BE) is an essential requirement for ensuring that patients receive a product that performs as indicated by the label. The BE standard for a particular product is set by its innovator, and this standard must subsequently be matched by generic drug products. The Biopharmaceutics Classification System (BCS) sets a scientific basis for an improved BE standard for immediate-release solid oral dosage forms. In this paper, we discuss BE and the BCS, as well as the issues that are currently relevant to BE as a pharmaceutical product standard.

Modulational stability brought by cubic–quartic interactions of the nearest-neighbor in FK model subjected in a parametrized on-site potential

Abstract This work extends to higher-order interactions the results of Ref. Nguetcho (2021), in which we discussed only on modulational instability in one-dimensional chain made of atoms, harmonically coupled to their nearest neighbors and subjected to an external on-site potential. Here we investigate the competition between cubic-quartic nonlinearities interactions of the nearest-neighbor and substrate’s deformability, and mainly discuss its impact on the modulational instability of the system. Defining relations of the noncommutative trace algebra of two 3×3 matrices

The noncommutative (or mixed) trace algebra $T_{nd}$ is generated by $d$ generic $n\times n$ matrices and by the algebra $C_{nd}$ generated by all traces of products of generic matrices, $n,d\geq 2$. It is known that over a field of characteristic 0 this algebra is a finitely generated free module over a polynomial subalgebra $S$ of the center $C_{nd}$. For $n=3$ and $d=2$ we have found explicitly such a subalgebra $S$ and a set of free generators of the $S$-module $T_{32}$. We give also a set of defining relations of $T_{32}$ as an algebra and a Groebner basis of the corresponding ideal. If a set in this class satisfies the strong separation condition and projects to a line segment for sufficiently many directions, then for each generic point there exists a rotation $\mathcal O$ such that all tangent sets at that point are either of the form $\mathcal O((\mathbb R \times C) \cap B(0,1))$, where $C$ is a closed porous set, or of the form $\mathcal O((\ell \times \{ 0 \}) \cap B(0,1))$, where $\ell$ is an interval.
