Inter- and intraspecific chromosome pattern variation in the yeast genusKluyveromyces

1998

The analysis of the electrophoretic chromosome patterns of the species of the genus Kluyveromyces, reveals a high polymorphism in size, number and intensity of bands. DiVerent sets of electrophoresis running conditions were used to establish species-specific patterns and also to detect intraspecific variation. According to their karyotypes, the species of this genus can be divided into two major groups. The first group includes the species K. africanus, K. bacillisporus, K. delphensis, K. lodderae, K. phaY, K. polysporus and K. yarrowii, composing the so-called ‘Saccharomyces cerevisiae-like’ group, because their karyotypes resemble that of the species S. cerevisiae. The second group compri…

An assessment of genetic relationships within the genus Digitalis based on PCR-generated RAPD markers

2000

RAPD markers were used to study inter-specific variation among six species of the genus Digitalis: D. obscura, D. lanata, D. grandiflora, D. purpurea, D. thapsi and D. dubia, and the hybrid D. excelsior (D. purpurea×D. grandiflora). A total of 91 highly reproducible bands amplified with four arbitrarily chosen decamer primers were obtained. Homology of the co-emigrating RAPD markers was tested by blot hybridisation and sequencing of selected bands. The application of a range of statistical approaches for RAPD data analysis, including distance and parsimony methods, family clustering and the analysis of molecular variance (AMOVA), indicated that these molecular markers were taxonomically inf…

Multiple methods for the identification of polymorphic simple sequence repeats (SSRs) in sorghum [Sorghum bicolor (L.) Moench]

1995

Simple sequence repeats (SSRs), also known as microsatellites, are highly variable DNA sequences that can be used as markers for the genetic analysis of plants. Three approaches were followed for the development of PCR primers for the amplification of DNA fragments containing SSRs from sorghum [Sorghum bicolor (L.) Moench]: a search for sorghum SSRs in public DNA databases; the use of SSR-specific primers developed in the Poaceae species maize (Zea mays L.) and seashore paspalum grass (Paspalum vaginatum Swartz); and the screening of sorghum genomic libraries by hybridization with SSR oligonucleotides. A total of 49 sorghum SSR-specific PCR primer pairs (two designed from GenBank SSR-contai…

The Association between a MAOB Variable Number Tandem Repeat Polymorphism and Cocaine and Opiate Addictions in Polyconsumers

2021

Genetic analysis of the association between alcohol, cocaine, and opiate addiction and variable number tandem repeat (VNTR) polymorphisms in monoamine oxidase B (MAOB) and serotonergic 5-hydroxytryptamine (serotonin) receptor 1B and 2C (HTR1B 21 and HTR2C) pathway genes was performed in a sample of 302 polyconsumers. Our genetic association analysis revealed a significant association between a 184 base pair (bp) VNTR polymorphism in the MAOB gene and addiction to cocaine and opiates. This work highlights new genetic marker associations in cocaine and opiate polyconsumer addictions. These data help to clarify and quantify the complex role of genetics in addictive disorders, as well as their …

A hierarchical analysis of genetic structure and variability in patchily distributed coexisting Chiastocheta species (Diptera:Anthomyiidae)

1996

The pattern of genetic variation in four coexisting fly species of the genus Chiastocheta was studied by allozyme electrophoresis. The fly species are confined to patches of one plant, Trollius europaeus, and thus experience very similar habitat fluctuations. Collection sites were chosen in a hierarchical fashion and F-statistics were estimated at three levels: intraregion, inter-region and total population. Population characteristic genetic parameters were compared within and among species and were related to the hierarchical level. The species were used as replicate experiments for inference of habitat history, and the hierarchical levels were used as inference for specific gene flow patt…

Genetic analysis of IDDM: The GAW5 multiplex family dataset

1989

In a collaborative effort by 12 centers from Europe and North America, data were assembled from 94 multiplex families with insulin-dependent diabetes mellitus (IDDM) for analysis of genetic and other factors of possible etiological importance. The dataset contains information on the following genetic markers: HLA-DR beta and -DQ beta restriction fragment length polymorphisms (RFLPs), three RFLPs detected with two probes that map 5' to the insulin gene, the serologically defined HLA loci, and the immunoglobulin allotypes. Data also were included for auto-antibodies to insulin and pancreatic islet cells as possible indicators of pathogenesis and for antibodies to certain viruses that have bee…

Genetic variability in the endemic Leucojum valentinum

2009

The genetic variability of Leucojum valentinum Pau (Amaryllidaceae), a vulnerable endemic species restricted to a small area in the region of Valencia (Eastern Spain), has been studied using random amplified polymorphic DNA (RAPD) markers. A total of 197 individuals from eleven populations were studied using 13 RAPD primers. Our results show high variability for the species, low differentiation among populations and uncorrelated levels of genetic variability and population size. Four groups in which three populations (SAG, PUG and COL) are separated from all the others were found, but without connection to geographical location.

Microsatellite marker-based genetic variability in Spanish rice cultivars and landraces

2010

The genetic relationships among japonica rice cultivars and landraces were studied. Most of them are of Spanish origin, and were developed and cultivated for varying time periods over more than a century. To uncover genetic diversity within each cultivar, we analysed 30 plants per cultivar or accession using 10 fluorescently labelled primer pairs for SSR markers. Six cultivars were included in the study, with accessions from four different regions of Spain of the traditional cultivar Bomba. A total of 37 alleles were detected with a mean of 3.7 alleles per locus. Polymorphism information content (PIC) ranged from 0 to 0.78 with an average of 0.51 per locus. Genetic diversity for cvs. Albufe…

Evaluation of morphological and genetic diversity of loquat accessions grown in sicily

2015

In this study 20 accessions of loquat (Eriobotrya japonica Lindl.) that originated from Sicily and other countries were characterized phenotypically and genotyped using seven microsatellite markers. The microsatellites clearly distinguished 14 accessions; three pairs of local accessions were not differentiated, and three accessions from abroad can be considered bud sport mutations of the main Spanish cultivar ‘Algerie’. The mean alleles per locus was four, ranging from two with the marker EJ075 and five with EJ012 and EJ271; the mean Polymorphic Information Content was 0.53; the average expected and observed heterozygosities resulted 0.62 and 0.59, respectively. These values revealed a …
