Comparison of the Phytotoxkit microbiotest and chemical variables for toxicity evaluation of sediments
2006
The main objective of the research was to evaluate the suitability of the Phytotoxkit microbiotest as a tool for hazard assessment of sediments. The concentrations of oil derivatives, polycyclic aromatic hydrocarbons (PAHs), and heavy metals (Cd, Cr, Cu, Hg, Ni, Pb, and Zn) were determined in sediment samples collected from the urban canal in Opole (Poland), in order to obtain a general insight of the level of sediment contamination. Phytotoxicity of sediments was estimated on the basis of seed germination and root elongation measurements, combined into an overall germination index (Gl). The results revealed spatial and vertical differentiation in sediment contamination. A good correlation …
Expression of different acid proteinases in germinating barley seed

Seed germination and reproductive features of Lysimachia minoricensis (Primulaceae), a wild-extinct plant.
2002
Lysimachia minoricensis is one of the few Mediterranean endemic plants (Minorca, Balearic islands) that has gone extinct in the wild but which persists as extant germplasm or cultivated plants in several botanical gardens. Reproductive features (seed set, number of seeds per capsule, seed weight) and germination responses to constant temperatures, sea water and dry-heat pre-treatments were investigated to determine the extent to which they may have influenced the extinction of the species. Seed set in Lysimachia is not dependent on pollinators, suggesting a functional selfer breeding system. Most plants produced a large mean number of fruits (23.2) and seeds (466), and the mean production o…
Seed cryopreservation of Fraxinus angustifolia Vahl
2016
Fraxinus angustifolia is used for afforestation and for production of both valuable timber and manna , a substance with pharmacological applications. The aim of this research was to establish the optimized condition for cryopreservation of F. angustifolia seeds, in order to protect the germplasm of this species. Germination percentage and mean time of germination of non-treated seeds (control) and liquid nitrogen-treated seeds were established in water or in 10 -6 M gibberellic acid (GA 3 ). The seeds could be cryopreserved with 3% of moisture content (MC) and germinated easily in water (70.0 ± 5.0%), while seeds cryopreserved with 6% MC showed a physiological dormancy. This state could be …
Variations in seed germination behavior ofPhleum hirsutumsubsp.ambiguumand possible applications in semi-natural grassland restoration
2015
Phleum hirsutum Honck. subsp. ambiguum (Ten.) Tzvelev is an important component of the vegetation of some semi-natural grassland habitats of central and southern Italy and especially of those referring to the 6210* habitat (Directive 92/43/EEC). Seeds of this species are required for the execution of restoration works concerning this habitat. Therefore, the recovery and the study of autochthonous germplasm are needed. This study focused on the intra-specific variations in seed mass and size and in seed germination behavior in this species since the knowledge of these aspects can be very useful in the planning of restoration projects. The present research evidenced significant differences in…
Application of Toxkit microbiotests for toxicity assessment in soil and compost
2004
The potential of Toxkit microbiotests to detect and analyze pollution in agricultural soil and the quality of compost was studied. The toxicity tests used included seed germination biotests using cress salad (Lepidum sativum L.), tomato (Lycopersicum esculentum L.), and cucumber (Cucumis sativus L.), and the Toxkit microbiotests included those with microalgae (Selenastrum capricornutum), protozoa (Tetrahymena thermophila), crustaceans (Daphnia magna, Thamnocephalus platyurus, and Heterocypris incongruens), and rotifers (Brachionus calyciflorus). Experiments on compost were undertaken in a modified solid-state fermentation system (SSF) and under field conditions (in a windrow). To promote th…
The effects of woodchip- and straw-derived biochars on the persistence of the herbicide 4-chloro-2-methylphenoxyacetic acid (MCPA) in soils
2014
Sorption and degradation are the primary processes controlling the efficacy and runoff contamination risk of agrochemicals. This study assessed the influence of two biochars, made from woodchips and straw at a pyrolysis temperature of 725°C and applied to a loamy sand and a sandy soil in the concentration of 5.3 g 100 g(-1) sandy soil and 4.1 g 100 g(-1) loamy sand soil, or 53 t ha(-1) for both soil types, on degradation of the herbicide 4-chloro-2-methylphenoxyacetic acid (MCPA). Soils were spiked with 50 mg MCPA kg(-1) soil. In the sandy soil, significantly more MCPA remained after 100 days if amended with straw-derived biochar in comparison to wood-derived biochar. Both biochars types si…
Herbicidal activity of Mediterranean essential oils and their effects on soil bioindicators.
2020
RCS1, a gene involved in controlling cell size inSaccharomyces cerevisiae
1991
Cloning and sequencing of RCS1, Saccharomyces cerevisiae gene whose product seems to be involved in timing the budding event of the cell cycle, is described. A haploid strain in which the 3'-terminal region of the chromosomal copy of the gene has been disrupted produces cells that are, on average, twice the size of cells of the parental strain. The critical size for budding in the mutant is similarly increased, and the disruption mutation is dominant in a diploid heterozygous for the RCS1 gene. Spores from this diploid have a reduced ability to germinate, the effect being more pronounced in the spores carrying the disrupted copy of RCS1. However, disrupted cells recover from alpha-factor tr…
Preventive and post-infection control ofBotrytis cinereain tomato plants by hexanoic acid
2008
The antifungal activity of hexanoic acid on the phytopathogen Botrytis cinerea was studied. This chemical inhibited both spore germination and mycelial growth in vitro in a concentration- and pH-dependent manner, and stopped spore germination at a very early stage, preventing germ-tube development. The minimum fungicidal concentration (MFC) for in vitro spore germination was 16 m m . Hexanoic acid also inhibited in vitro mycelial growth of germinated spores at an MFC of 12 m m . Studies performed to characterize the mechanisms underlying the antimicrobial effect of hexanoic acid showed that it alters fungal membrane permeability. In addition, hexanoic acid treatment increased the levels of …
