Abstract Background Low concentration of plasma pyridoxal-5-phosphate (PLP) is associated with hyperhomocysteinemia and inflammation. Most methods for the measurement of plasma PLP require large specimen volume and involve the use of toxic reagents. Methods We have developed a HPLC method for the measurement of PLP and 4-pyridoxic acid (4-PA) in plasma, which requires small specimen volume. The samples are prepared without adding any toxic reagents. Furthermore, we have examined whether intake of vitamin B 6 affects the concentration of plasma PLP and 4-PA. Abstract BACKGROUND: This is an observational study undertaken in aim to evaluate the association between metabolic syndrome (MS) and high homocysteinemia (HHcy) in relation with cardiovascular disease (CVD). METHODS: The study involved 126 subjects with angiographically documented CVD and 65 healthy subjects. MS has been diagnosed according to the ATP III criteria and plasma homocysteine concentration has been evaluated. RESULTS: In patients with CVD the prevalence of MS and HHcy is 17.4% and 25.4% respectively; MS coexists with HHcy in 67.2% of patients; analogous results can be observed among men and women. Determining sulfur-containing amino acids by capillary electrophoresis: A fast novel method for total homocyst(e)ine human plasma

A high-performance capillary electrophoresis (HPCE) method based on laser-induced fluorescence detection is presented here. It enables the determination of sulfur-containing amino acids within 15 min. Fluorescence of sulfur-containing amino acids in plasma is linear over a range of 50-150 micromol/L for L-methionine, 5-100 micromol/L for L-homocysteine, and 50-200 micromol/L for L-cysteine. For homocysteine, we were able to detect 1 fmol injected, equivalent to a plasma concentration of 10 nmol/L. Objectives This prospective study was designed to evaluate the effect of joint determination of two important contrary biomarkers—homocysteine and glutathione peroxidase (GPx)-1—on cardiovascular risk stratification.
