Benzo(a)pyrene is metabolized to two major groups of mutagenically reactive metabolites: Monofunctional epoxides and dihydrodiol-epoxides. Various monooxygenase forms catalyze the various pathways at very different rates. In metabolic situations where the contribution by dihydrodiol-epoxides is small, epoxide hydratase represents a very efficient protective system. However, in situations where the mutagenic effect is predominately due to dihydrodiol-epoxide, the effect of epoxide hydratase is complicated and weak. Dihydrodiol Dehydrogenase: Substrate Specificity, Inducibility and Tissue Distribution

The present study shows that: Dihydrodiol dehydrogenase activity is present in the 100,000 g supernatant fraction of extrahepatic tissues. Dihydrodiol dehydrogenase is able to oxidize the hydroxy group and to reduce the keto group of a number of xenobiotics including quinones derived from polycyclic aromatic hydrocarbons. Dihydrodiol dehydrogenase was not inducible by various substances including hormones, polycyclic aromatic hydrocarbons, substrates of the enzyme and potent inducers of monooxygenases, epoxide hydrolase and glutathione S-transferases. Only in the case of thyroxine was a weak induction with a high dose of the hormone observed.

Inhibition of Plant Lactate Dehydrogenase Isoenzymes by Benzoic Acid and Cinnamic Acid Derivatives

Summary Several phenolic compounds such as derivatives of benzoic and cinnamic acid were investigated with respect to their inhibitory effect on potato tuber lactate dehydrogenase isoenzymes. Ki values were determined and it was found that they were in the range of 10−2 M for several benzoic acid derivatives while they were in the range of 10−5 M for several hydroxylated cinnamic acid derivatives such as 3,4-dihydroxy cinnamic acid, p(m)nitrocinnamic acid, and 7,8-dihydroxy-4-methyl coumarin as well as kynurenic acid. Publisher Summary This chapter describes both electrophoretic and catalytic studies on lactate dehydrogenase (LDH) in the developing brain. It has been found that the enzyme exhibits at birth a composition that gradually changes to attain the final pattern observed in the adult. The neurochemical findings on LDH isozymes parallel the rapid growth in the first days after birth. Isozyme E, which is not present in detectable amounts 2 days after birth, represents 15% of the enzyme on the 22nd day. Cancer stem cells (CSC) have a central role in driving tumor growth. Since metabolism is becoming an important diagnostic and therapeutic target, characterization of CSC line energetic properties is an emerging need. Embryonic and adult stem cells, compared to differentiated cells, exhibit a reduced mitochondrial activity and a stronger dependence on aerobic glycolysis. The reactions of [Fe2(CO)6(μ-sdt)] (1) (sdt = SCH2SCH2S) with phosphine ligands have been investigated. Treatment of 1 with dppm (bis(diphenylphosphino)methane) or dcpm (bis(dicyclohexylphosphino)methane) affords the diphosphine-bridged products [Fe2(CO)4(μ-sdt)(μ-dppm)] (2) and [Fe2(CO)4(μ-sdt)(μ-dcpm)] (3), respectively. The complex [Fe2(CO)4(μ-sdt)(κ2-dppv)] (4) with a chelating diphosphine was obtained by reacting 1 with dppv (cis-1,2-bis(diphenylphosphino)ethene).
