Identification and expression of Lactobacillus paracasei genes for adaptation to desiccation and rehydration

2018

AbstractLactobacillus paracaseiis able to persist in a variety of natural and technological environments despite physico-chemical perturbations, in particular alternations between desiccation and rehydration. However, the way in which it adapts to hydric fluctuations and in particular the genetic determinants involved are not clearly understood. To identify the genes involved in adaptation to desiccation, an annotated library ofL. paracaseirandom transposon mutants was screened for viability after desiccation (25% relative humidity, 25°C). Subsequently, the expression of the identified genes was measured at five stages of the dehydration-rehydration process to formulate the chronology of ge…

Hepatocellular carcinoma (HCC) vs. non-HCC: accuracy and reliability of Liver Imaging Reporting and Data System v2018

2019

PURPOSE: The Liver Imaging Reporting and Data System (LI-RADS) was created to standardize the diagnostic criteria for hepatocellular carcinoma (HCC), and has undergone multiple revisions including a recent update in 2018 (v2018). The primary aim of this study was to determine the diagnostic performance and interrater reliability (IRR) of LI-RADS v2018 for distinguishing HCC from non-HCC primary hepatic malignancy in patients ‘at-risk’ for HCC. A secondary aim was to assess the impact of changes introduced in the v2018 diagnostic algorithm. METHODS: This retrospective study combined a 10-year experience of pathologically-proven primary liver malignancies from two large liver transplant cente…

The community conserved landscape of the Borana Oromo, Ethiopia

2011

Purpose - This paper seeks to describe an attempt to assess at the local level the progress that has been internationally achieved in recognition of community and indigenous rights, and of indigenous and community conserved areas. An action-research exercise was implemented in Ethiopia with a mobile indigenous people of evaluating customary as well as government-led governance of the environment, with the objective of strengthening the capacity of the Borana-Oromo to conserve their landscape. Design/methodology/approach - This paper is based on collaborative research implemented by the authors in 2002 while SOS Sahel Ethiopia was introducing collaborative forest management, and on a 2007 ac…

Second Generation of Freescoo Solar DEC Prototypes for Residential Applications

2015

Freescoo is an innovative all-in-one compact solar Desiccant Evaporative Cooling (DEC) air conditioner concept. Results of the first prototype developed were presented at SHC Conference last year. Now a second generation of freescoo prototypes for applications in residential and small office buildings have been installed in Italy at ENEA Casaccia and at UNIPA. The thermodynamic cycle is based on the use of fixed and cooled adsorption beds and advanced evaporative cooling concepts. The adsorption bed, which is a fin and tube heat exchanger packed with silica gel grains, allows simultaneous dehumidification and cooling of the process air. The indirect evaporative cooling process, operated dow…

BackgroundThe insect cuticle covers the whole body and all appendages and has bi-directionnal selective permeability: it protects against environmental stress and pathogen infection and also helps to reduce water loss. The adult cuticle is often associated with a superficial layer of fatty acid-derived molecules such as waxes and long chain hydrocarbons that prevent rapid dehydration. The waterproofing properties of cuticular hydrocarbons (CHs) depend on their chain length and desaturation number.DrosophilaCH biosynthesis involves an enzymatic pathway including several elongase and desaturase enzymes.MethodsThe link between desiccation resistance and CH profile remains unclear, so we tested…

Potassium uptake system Trk2 is crucial for yeast cell viability during anhydrobiosis

2013

Yeasts grow at very different potassium concentrations, adapting their intracellular cation levels to changes in the external environment. Potassium homeostasis is maintained with the help of several transporters mediating the uptake and efflux of potassium with various affinities and mechanisms. In the model yeast Saccharomyces cerevisiae, two uptake systems, Trk1 and Trk2, are responsible for the accumulation of a relatively high intracellular potassium content (200-300 mM) and the efflux of surplus potassium is mediated by the Tok1 channel and active exporters Ena ATPase and Nha1 cation/proton antiporter. Using a series of deletion mutants, we studied the role of individual potassium tra…

The impact of dehydration rate on the production and cellular location of reactive oxygen species in an aquatic moss.

2012

† Background and Aims The aquatic moss Fontinalis antipyretica requires a slow rate of dehydration to survive a desiccation event. The present work examined whether differences in the dehydration rate resulted in corresponding differences in the production of reactive oxygen species (ROS) and therefore in the amount of cell damage. † Methods Intracellular ROS production by the aquatic moss was assessed with confocal laser microscopy and the ROS-specific chemical probe 2,7-dichlorodihydrofluorescein diacetate. The production of hydrogen peroxide was also quantified and its cellular location was assessed. † Key Results The rehydration of slowly dried cells was associated with lower ROS produc…
