Clostridium difficile IStron CdISt1: Discovery of a Variant Encoding Two Complete Transposase-Like Proteins

ABSTRACT Screening a Clostridium difficile strain collection for the chimeric element Cd ISt1 , we identified two additional variants, designated Cd ISt1 -0 and Cd ISt1 -III. In in vitro assays, we could prove the self-splicing ribozyme activity of these variants. Structural comparison of all known Cd ISt1 variants led us to define four types of IStrons that we designated Cd ISt1 -0 through Cd ISt1 -III. Since Cd ISt1 -0 encodes two complete transposase-like proteins (TlpA and TlpB), we suggest that it represents the original genetic element, hypothesized before to have originated by fusion of a group I intron and an insertion sequence element.

Aims: This report describes an investigation into the genetic profiles of 38 Candida albicans and 19 Candida glabrata strains collected from a dental hospital of Monastir (Tunisia) and the Laboratory of Parasitology, Farhat Hached Hospital of Sousse (Tunisia), using two typing methods: random amplified polymorphic DNA (RAPD) and contour-clamped homogenous electric fields (CHEF). Methods and Results: The two methods (RAPD and CHEF electrophoresis) were able to identify clonal-related isolates from different patients. Collective and directed cell movements are crucial for diverse developmental processes in the animal kingdom, but they are also involved in wound repair and disease. During these processes groups of cells are oriented within the tissue plane, which is referred to as planar cell polarity (PCP). SUMMARYChromosome number n = 9 and 2n = 18 for Clavelina lepadiformis (Ascidiacea, Aplousobranchiata) from the Gulf of Palermo have been determined. Silver staining analysis of testicular cells reveals that within-individual variability in NOR banding patterns is present. Using the C-banding procedure, a very impressive heterochromatin amount seems to characterize the chromosome set of this species.

Summary. We compared two phenotypic methods, colony morphotyping on Sabouraud-tripheniltetrazolium agar (STTZ) and serotyping, with two genotypic methods, karyotyping and Random Amplified Polymorphic DNA bands obtained by PCR amplification (RAPD-PCR), for strain delineation in 33 Candida clinical isolates and two C. albicans strains from culture collections. Analysis of isolates on STTZ showed 11 different morphotypes. The research aimed at using geospatial platforms to collect and map discursive, visual and oral testimonies, stories and narratives of individuals. It means, exploring the various testimonies of people related to quarantine, physical distancing, resistances and experiences, and practices that occur in the daily spaces of their homes and in local communities.

The pattern of morphological and mtDNA cytochrome b diversity of three calanoid copepod species belonging to the diaptomid genus Hemidiaptomus has been investigated with the aim of checking the reliability of the morphological characters currently used for species identification, and the possible presence of cryptic taxa.
