Targeting human {gamma}delta} T cells with zoledronate and interleukin-2 for immunotherapy of hormone-refractory prostate cancer.

Abstract The increasing evidence that γδ T cells have potent antitumor activity suggests their value in immunotherapy, particularly in areas of unmet need such as metastatic carcinoma. To this end, we initiated a phase I clinical trial in metastatic hormone-refractory prostate cancer to examine the feasibility and consequences of using the γδ T-cell agonist zoledronate, either alone or in combination with low-dose interleukin 2 (IL-2), to activate peripheral blood γδ cells. Integrin heterodimers sharing the common alphaV subunit are receptors for adhesion glycoproteins such as vitronectin and fibronectin. They are suggested to play an essential role in cell anchoring, differentiation, and survival. Here, we describe the construction of an expression plasmid coding for an intracellular single-chain antibody against alphaV integrin subunit. Saos-2 osteosarcoma cells transfected with this DNA construct showed an approximately 70-100% decrease in the cell surface expression of alphaVbeta3 and alphaVbeta5 integrins as shown by flow cytometry. The influence of an ion-exchange resin on the kinetics of hydration of tricalcium silicate

The addition of a finely-ground ion-exchange resin makes it possible to modify the hydration kinetics of C3S pastes. Analyses of the liquid phase in pastes and more dilute suspensions show that the resin exchanges calcium ions for sodium ions very rapidly during the early stage of hydration and therefore the concentration of silica in solution increases. The resin impacts the hydration of C3S by other mechanisms which depends on the resin quantity added. For a high resin quantity, the induction period is very short, but the longer-term hydration is enhanced compared to a reference sample without resin. Cellular and molecular mechanisms in the induction phase of contact sensitivity.

During the induction phase of contact sensitivity, hapten-specific Th1 cells are primed by epidermal Langerhans cells. These Langerhans cells present hapten on MHC class II molecules and provide costimulatory signals. This presentation discusses the induction of cytokines in Langerhans cells and keratinocytes by haptens and their regulatory effects on contact sensitivity. Haptens were painted on the skin of normal BALB/c mice and epidermal cells were prepared at various times thereafter. Langerhans cell-derived interleukin (IL)-1 beta mRNA was observed as early as 15 min after hapten paining. In order to elucidate the role of keratinocytes (KCs) in the induction of contact sensitivity, we applied various contact sensitizers [2,4-dinitrofluorobenzene (DNFB), urushiol, 3-n-pentadecylcatechol (PDC), 4-ethoxymethylene-2-phenyloxazol-5-one (oxazolone)] and tolerizing compounds [2,4-dinitrothiocyanobenzene (DNTB), 5-methyl-3-n-pentadecyl-catechol (5-Me-PDC)] onto the earskin of non-sensitized Balb/c mice. Optimal control of the inversion of two spins in Nuclear Magnetic Resonance

International audience; We investigate the optimal control of the inversion of two spin 1/2 particles in Nuclear Magnetic Resonance. The two spins, which differ by their resonance offset, are controlled by the same radio frequency magnetic field. Using the Pontryagin Maximum Principle, we compute the optimal control sequence which allows to reach the target state in a given time, while minimizing the energy of the magnetic field. A comparison with the time-optimal solution for bounded control amplitude realizing the same control in the same time is made. An experimental illustration is done using techniques of Nuclear Magnetic Resonance.

Solid rotor induction machines for use in electrically-assisted turbochargers

The stator leakage reactance has a major influence on the performance of a solid rotor induction machine. It is larger and it is more difficult to define and calculate than in a cage-rotor machine. When the higher leakage reactance is taken into account, the equivalent circuit model gives predictions of sufficient precision for design purposes. The airgap, normally chosen to be quite small, is preferably quite large by general standards. The torque varies as (V/f) 3 which makes it difficult to provide a wide range of constant power operation in a variable speed application. Slitting the rotor helps to improve the characteristic for variable-speed duties.

Abstract Background & Aims: In addition to acting as a regulator of food intake and energy expenditure, leptin can also modulate immune and inflammatory responses. The role of leptin in intestinal inflammation is the focus of the present study. Methods: Acute and chronic colitis were induced in leptin-deficient ob/ob or wild-type (WT) mice using dextran sulfate sodium (DSS) or trinitrobenzene sulfonic acid (TNBS). Design criteria of tubular linear induction motors and generators: A prototype realization and its characterization

In this paper a mathematical model of tubular linear induction machines (TLIM) with hollowed induced part is recalled. Moreover the design criteria of a TLIM with hollowed iron induced part are presented as well as the technological processes to be adopted and the choice of materials to construct the various parts. The methodologies for mechanical assembling and electric wiring are considered too. A prototype with bimetallic induced part has been designed and built. Finally some experimental results on electrical and mechanical variables, when the machines are used as motors, are shown.
