Isolation of a Putative Hydroxyacyl Enzyme Intermediate of an Epoxide Hydrolase

1994

A putative covalent, alpha-hydroxyacyl intermediate was isolated by the brief exposure of murine soluble epoxide hydrolase to its substrate. The reaction was reversed by time and blocked by competitive inhibitors. The formation of the intermediate was dependent upon the concentration of the enzyme and was increased by incubation under acidic conditions. The structure of the intermediate was supported by microchemical methods.

Sequestration of biological reactive intermediates by trapping as covalent enzyme-intermediate complex

One important class of biological reactive intermediates arising in the course of human xenobiotic metabolism are arene and alkene oxides. The major safeguard against the potential genotoxic effects of these compounds is the microsomal epoxide hydrolase (mEH). This enzyme has a broad substrate specificity but--on the first sight--seems to be inadequately suited for this protection task due to its low turnover number with most of its substrates. Exhalation of ethylene oxide by rats on exposure to ethylene.

Abstract When rats are exposed to ethylene in a closed desiccator jar chamber, the rate of metabolic elimination of the compound is influenced by pretreatment with Aroclor 1254 (metabolism increased) and diethyldithiocarbamate (metabolism inhibited). Biotransformation of ethylene leads to ethylene oxide as reactive intermediate. This is exhaled and can be quantitated in the gas phase of the system.

Coupling of Structural Additions for the Mitigation of Seismic Response in Existing Buildings

In the present paper, additions in structural steelwork are utilized for giving new life to old buildings in regions characterized by medium/high seismic hazard. Two models are here proposed, i.e.: vertical addition for masonry buildings and lateral addition for r.c. buildings. For the model of vertical addition, the connection between the masonry and steel structures is realized by means of an Intermediate Isolation System (IIS). Low-altitude outbreaks of human fascioliasis related with summer rainfall in Gilan province, Iran

Following human fascioliasis outbreaks in 1988 and 1999 in Gilan province, northern Iran, efforts are now made to shed light on the seasonal pattern of fascioliasis transmission in this endemic area, taking into account snail host populations, climatic conditions and human cases. Populations of the intermediate host snail (Lymnaea spp.) peak in May and November, while there is a fourfold increase in the rate of human fascioliasis in February compared to that of September. Transmission is likely to occur mainly in late autumn and sporadically in late spring. Observation of Photon Polarization in theb→sγTransition

This Letter presents a study of the flavor-changing neutral current radiative $B^{\pm} \to K^{\pm}\pi^{\mp}\pi^{\pm}\gamma$ decays performed using data collected in proton-proton collisions with the LHCb detector at $7$ and $8\,$TeV center-of-mass energies. In this sample, corresponding to an integrated luminosity of $3\,\text{fb}^{-1}$, nearly $14\,000$ signal events are reconstructed and selected, containing all possible intermediate resonances with a $K^{\pm}\pi^{\mp}\pi^{\pm}$ final state in the $[1.1, 1.9]\,$GeV/$c^{2}$ mass range. Intermediate Isolation System with Nonlinear Lower Structure and Isolation System

This paper reports a study on the Intermediate Isolation System (IIS) applied to existing buildings. This kind of application is particularly suitable when a vertical addition is planned for buildings in seismic zones; in such a case, an isolation system can be placed at the base of the extension to prevent the increase, or, better, to reduce the seismic demand on the existing structure. The mechanism of formamide hydrolysis in water from ab initio calculations and simulations.

The neutral hydrolysis of formamide in water is a suitable reference to quantify the efficiency of proteolytic enzymes. However, experimental data for this reaction has only very recently been obtained and the kinetic constant determined experimentally is significantly higher than that predicted by previous theoretical estimations.
