FACS-based protocol to assess cytotoxicity and clonogenic potential of colorectal cancer stem cells using a Wnt/β-catenin signaling pathway reporter

Summary Cancer stem cells (CSCs) play a key role in tumor initiation and progression. A real-time tool to evaluate the activation of CSC-specific signaling pathways is crucial for the study of this cancer cell subset. Here, we present a protocol to monitor, in vitro, the activation of Wnt/β-catenin signaling pathway, which is considered a functional biomarker for colorectal CSCs (CR-CSCs). This flow-cytometry-based protocol allows it to isolate CR-CSCs and to evaluate their cytotoxicity upon anti-tumor treatments. In this paper, the strategy of Intermediate Isolation System (IIS) is proposed for the vertical extension of an aggregate of masonry buildings located in Pozzuoli, south Italy. The effectiveness of IIS working as a nonconventional Tuned Mass Damper (TMD) for the existing construction is assessed in terms of reduction of seismic demand and damage in the masonry structure undergoing inelastic deformation under seismic input. The present work reports the isolation of a new serovar of Vibrio vulnificus pathogenic for eels cultured in freshwater farms. This serovar, named Serovar A, produced two outbreaks of low morbidity and mortality in one Spanish eel farm in September 2000 and March 2001. The vibriosis affected healthy eels, weighting between 10 and 100 g, that had been vaccinated against the serovar E of the species (formerly biotype 2) at least 6 months before. Some of the diseased eels showed symptoms different to those reported for the classical vibriosis due to serovar E. In order to obtain an automated system of railway management, it is necessary to automatically detect, isolate and identify all switches in a point cloud which represents the railway. To realize this automated system of detection, a set of pre-processing steps is applied. The system begins by detecting and isolating tracks through application of a mask on each section of the point cloud. Then, it does a denoising through mathematical morphology and a compression in replacing a group of points by their centroid. Finally, it closes tracks holes through extrapolation. Atoll-like structures of the endemic Mediterranean seagrass Posidonia oceanica were encountered in the innermost area of the Stagnone di Marsala, a semi-enclosed coastal lagoon along the western coasts of Sicily.
