ChemInform Abstract: Photochemical Transformations of Some Neoclerodane and Labdane Diterpene Ketones.
2010
Some neoclerodane (2–4) and labdane (5 and 6) diterpene ketones having a β- or γ-hydroxy function, an α-epoxy group, or an α-olefinic double bond have been irradiated at λ = 313 nm in dry solvents (benzene or methanol). Fruticolone (2) yielded the naturally occurring 5,6-seco-neoclerodan-6,1α-olide derivative fruticolide (1), while hispanolone (5) gave the δ-lactone 8, both transformations involving a Norrish type I photoreaction. The α,β-unsaturated ketone derivative 4 was transformed into 7 by a stereoselective intramolecular [2+2] cycloaddition reaction involving its furanic 13(16)-double bond. Finally, the α-epoxy ketone 6 was rearranged to the 10(98)abeo-labda-7,9-dione derivative 10. …
Catalytic activity of rare-earth-supported catalysts in Friedel–Crafts acylations
1999
Abstract Friedel–Crafts acylations are catalysed by rare-earth-supported catalysts. The preparation, characterization and performance of these solid catalysts in a test acylation reaction and in a variety of syntheses of aromatic ketones are reported. In contrast to the reactions using AlCl3, the experimental conditions are non-polluting and the final work-up does not require any aqueous treatment.
Stereoselective Total Synthesis and Absolute Configuration of the Natural Decanolides (−)-Microcarpalide and (+)-Lethaloxin. Identity of (+)-Lethalox…
2005
[reaction: see text] Convergent, stereoselective syntheses of the pharmacologically active, naturally occurring lactones (-)-microcarpalide and (+)-lethaloxin have been achieved from the commercially available, chiral reagents (R)-glycidol, (S,S)-tartaric acid, and d-ribose as the starting materials. These syntheses have further served to establish the hitherto unknown absolute configuration of (+)-lethaloxin and to show its identity with (+)-pinolidoxin.
The synthesis and effect of fluorinated chalcone derivatives on nitric oxide production
2002
Abstract Dimethoxy- and trimethoxychalcone derivatives, with various patterns of fluorination, were synthesized and evaluated for their influence on nitric oxide production. Some of them, chalcones 1 , 5 , 7 , 10 , 11 and 17 , inhibited NO production with an IC 50 in the submicromolar range; 17 is especially noteworthy because of its potency (IC 50 30 nM). These effects were not the consequence of a direct inhibitory action on enzyme activity but the inhibition of enzyme expression.
Ketogenic Diet Decreases Alcohol Intake in Adult Male Mice
2021
The classic ketogenic diet is a diet high in fat, low in carbohydrates, and well-adjusted proteins. The reduction in glucose levels induces changes in the body’s metabolism, since the main energy source happens to be ketone bodies. Recent studies have suggested that nutritional interventions may modulate drug addiction. The present work aimed to study the potential effects of a classic ketogenic diet in modulating alcohol consumption and its rewarding effects. Two groups of adult male mice were employed in this study, one exposed to a standard diet (SD, n = 15) and the other to a ketogenic diet (KD, n = 16). When a ketotic state was stable for 7 days, animals were exposed to the oral self-a…
Hibernation impact on the catalytic activities of the mitochondrial D-3-hydroxybutyrate dehydrogenase in liver and brain tissues of jerboa (Jaculus o…
2003
Abstract Background Jerboa (Jaculus orientalis) is a deep hibernating rodent native to subdesert highlands. During hibernation, a high level of ketone bodies i.e. acetoacetate (AcAc) and D-3-hydroxybutyrate (BOH) are produced in liver, which are used in brain as energetic fuel. These compounds are bioconverted by mitochondrial D-3-hydroxybutyrate dehydrogenase (BDH) E.C. 1.1.1.30. Here we report, the function and the expression of BDH in terms of catalytic activities, kinetic parameters, levels of protein and mRNA in both tissues i.e brain and liver, in relation to the hibernating process. Results We found that: 1/ In euthemic jerboa the specific activity in liver is 2.4- and 6.4- fold high…
Conditions for the Enhancing Effect of Protease Inhibitors on the Concanavalin A Induced Thymidine Response of Murine Lymphocytes
1983
Incorporation of [3H]-thymidine – [3H]-TdR – into concanavalin A (Con A) stimulated murine splenocytes and thymocytes was found to be enhanced by addition of certain concentrations of phenyl-methyl-sulfonylfluoride (PMSF), di-isopropylfluorophosphate (DFP), N-α-tosyl-<i>L</i>-lysyl-<i>L</i>-chloromethylketone (TLCK), and soybean trypsin inhibitor (SBTI). No enhancement could be observed when mononuclear cells of the peripheral blood were used, and a medium enhancement when thymocytes were applied. Furthermore, no enhancing effect of the protease inhibitors (PI) on the Con A response of murine splenocytes could be observed…
Possible Reduction of Cardiac Risk after Supplementation with Epigallocatechin Gallate and Increase of Ketone Bodies in the Blood in Patients with Mu…
2020
Multiple sclerosis (MS) is a neurodegenerative disease that causes anthropometric changes characterised by functional disability, increase in fat mass, and decrease in lean mass. All these variables are related to a greater cardiac risk. The polyphenol epigallocatechin gallate (EGCG) and an increase in ketone bodies in the blood have been shown to have beneficial effects on anthropometric and biochemical variables related to cardiovascular activity. The aim of this study was to analyse the impact of the intervention with EGCG and ketone bodies on cardiac risk in MS patients. A population of 51 MS patients were randomly assigned to a control group and an intervention group (daily dose of 800…
Effects of small interfering RNAs targeting fascin on human esophageal squamous cell carcinoma cell lines
2010
Abstract Background Fascin induces membrane protrusions and cell motility. Fascin overexpression was associated with poor prognosis, and its downregulation reduces cell motility and invasiveness in esophageal squamous cell carcinoma (ESCC). Using a stable knockdown cell line, we revealed the effect of fascin on cell growth, cell adhesion and tumor formation. Methods We examined whether fascin is a potential target in ESCC using in vitro and in vivo studies utilizing a specific siRNA. We established a stable transfectant with downregulated fascin from KYSE170 cell line. Results The fascin downregulated cell lines showed a slower growth pattern by 40.3% (p In vivo, the tumor size was signific…
Dynamics of ketone body metabolism in diabetic rats.
1972
Steady state blood levels of ketone bodies during infusions of acetoacetate at various rates have been compared in healthy and diabetic rats. The characteristics of the metabolic elimination of ketone bodies from the blood are completely changed in diabetic rats. Whereas steady state levels of ketone bodies increase linearly with the infusion rate in healthy rats, this increase is exponential in diabetic animals. This difference, which is due to an impaired metabolic elimination, becomes evident only above a dosage of 50 μmoles acetoacetate per kg per min. Chronic treatment with insulin for 4–6 days, but not acute insulin injection, restores the capacity of diabetic rats to metabolize keton…
