An investigation of the material, chromatographic, thermodynamic, and kinetic properties of thermally treated (i.e., annealed) polymeric stationary phases imprinted with l-phenylalanine anilide (l-PA) was carried out. The imprinting procedure of the solid phase used in this study was the same as for the untreated imprinted stationary phase studied previously. However, after polymerization, these new stationary phases were treated at elevated temperatures (50, 120, 140, and 160 °C) for 24 h. The treatment at 120 and 140 °C led to a larger decrease in the retention of l-PA than that of d-PA. The polymer treated at 160 °C could no longer resolve the d,l-PA racemate. Inactivation and Regeneration Kinetics of Horseradish Peroxidase Heated at High Temperatures.

The inactivation kinetics of horseradish peroxidase (HRP) heated in capillary tubes in the range 110 to 135°C was studied. Its regeneration kinetics when stored at 4 and 25°C was also considered. As the severity of the treatment increased, the absolute value of the regeneration decreased. The storage temperature of the enzyme did not affect the percentage of maximum activity regenerable, although when this temperature was raised from 4 to 25°C the speed of regeneration increased. Kinetics of HRP inactivation determined after heating and after regeneration were compared. Thermal inactivation at high temperatures and regeneration of green asparagus peroxidase

A spectrophotometric method was developed for determining the peroxidase activity of green asparagus in small samples. The optimum conditions for the analysis in the cuvette were 45 mM of H2O2 36 mM of guaiacol, and pH 7. The method can be used to determine enzyme activity at up to two decimal reductions. A study was performed of the regeneration and inactivation kinetics of the enzyme when heated between 90 and 125°C. Regenerated asparagus peroxidase reached its maximum activity after being stored 6 days at 25°C. The regenerated enzyme followed first-order inactivation kinetics, showing an Ea = 13.62 kcal/mol and k100°C = 2.07 min-1.

Isolation and characterization of a chlorogenic acid esterase from Aspergillus niger.

Abstract The isolation and characterization of a specific chlorogenic acid esterase is described. The enzyme activity is measured by determination of the hydrolysis product caffeic acid. The enzyme had been concentrated by means of ultrafiltration and column-chromatography. The pH- and temperature optimum were 6.5 and 45 °C respectively. Divalent cations were not required for the enzyme activity. As other esterases, this enzyme is inhibited by di-isopropyl-phosphorofluoridate. The Km-value is 0.70 mᴍ chlorogenic acid, the molecular weight 240000. The described enzyme is specific for chlorogenic acid. Deoxyribonucleases in Herpes simplex Virus Type 1 and 2 Infected Primary Rabbit Kidney Cells

Abstract In primary rabbit kidney cells infected with herpes simplex virus four different neutral deoxyribonuclease activities can be detected by means of the deoxyribonuclease assay in DNA-containing polyacrylamide gels following their separation by discelectrophoresis. The method is suitable to follow independently the change in each activity of the different enzymes using only about 5 × 105 cells for each assay during the time-course of infection. Under these conditions one enzyme activity is constant, two disappear while the activity of a fourth one present only in infected cells, increases.

A continuous reaction system to investigate dispersion polymerization of vinyl monomers in supercritical carbon dioxide

A laboratory-scale continuous reaction system using a stirred tank reac- tor was assembled in our laboratory to study the dispersion polymerization of vinyl monomers in supercritical carbon dioxide (scCO2). The apparatus was equipped with a suitable downstream separation section to collect solid particles entrained in the effluent stream from the reactor, whose monomer concentration could be measured online with a gas chromatograph. The impact of Pleurotus ostreatus on organic matter transformation processes.

This study showed the effect of Pleurotus ostreatus on the external organic matter (OM) transformation processes. The kinetics of these changes were determined. The experiment was conducted as 42-day pot experiment done in four combinations (with four replications). The four combinations of different substrates were: (1) organic, (2) composted hay, (3) organic soil + composted hay, (4) organic soil + composted hay + lead. The study results showed that P. ostreatus had the ability to transform external OM as well as metal–organic complexes. Thermodynamically induced shear degradation

The shear degradation of polymers in semiconcentrated solutions is viewed as a function of solvent quality. It is shown that the deterioration of the thermodynamic quality of the solvent leads to a marked flow resistance due to an increasing number of contacts between the chains. This may be probed by viscosimetric measurements as well as by degradation experiments (“thermodynamically induced shear degradation”). For a detailed discussion of this phenomenon the experimental setup for the mechanochemical experiment is outlined first, followed by a survey of the kinetics of chain scission. Iodine and mercury resonance lamps for kinetics experiments and their spectra in the far ultraviolet

Electrodeless iodine and mercury iodide radio-frequency discharge lamps have been made to provide intense sources of resonance radiation of iodine and mercury in the 183-253 nm region. Production technology and emission spectra in the ultraviolet and vacuum ultraviolet regions are described. Both lamps (iodine and mercury iodide) are designed and used for monitoring the iodine 2P1/2 (206.163 nm) and 2P3/2 (183.038 nm) level population kinetics after flash photolysis of I2 or other precursor molecules in laboratory experiments. The technique is suitable for the investigation of the gas phase reaction kinetics following the photolysis of I2 in the presence of O3.

Summary: Several techniques based on different principles have been proposed to measure lipid hydroperoxides. Enzymatic methods are sensitive and can be quite specific but they are subject to interference by inhibitors and not all are stoichiometric. The present work proposes some modifications of the Heath & Tappel (Anal Biochem 1976; 7:184—91) enzymatic method of determination of lipid hydroperoxides in order to standardize and automate it and to meet the analytical criteria required for a biological assay.
