A kinetic study on the regeneration ofCandida albicansprotoplasts in the presence of cell wall synthesis inhibitors

Aculeacin A and papulacandin B block cell wall regeneration in Candida albicans protoplasts at an intermediate step in which the protoplasts have not yet synthesized the rigid structure of the cell wall and are therefore still osmotically sensitive. In the presence of the antibiotics, total synthesis of glucan is not significantly lowered with respect to control cells, although most of it appears either in the culture medium or in the regenerating wall as alkali-soluble glucan. Thus, it is proposed that echinocandins (such as aculeacin A) and papulacandins may not inhibit glucan synthesis per se but instead inhibit its incorporation into the supramolecular organization of the cell wall.

Spherical polymeric microparticles have been prepared by a reverse phase suspension polymerization technique. The starting polymer was alpha,beta-poly(N-2-hydroxyethyl)-DL-aspartamide (PHEA), partially derivatized with glycidylmethacrylate (GMA). PHEA-GMA copolymer (PHG) was crosslinked in the presence of N,N'-dimethylacrylamide (DMAA) or N,N'-ethylenebisacrylamide (EBA). 5-fluorouracil was incorporated into PHG-DMAA or PHG-EBA beads both during and after the crosslinking process. Swelling studies revealed a high affinity toward aqueous medium, influenced by the presence of 5-fluorouracil. Effect of nonprotein thiols on protein synthesis in isolated rat hepatocytes.

The ability of nonprotein thiols to modulate rates of protein synthesis was investigated in isolated rat hepatocytes. Addition of cysteine stimulates protein labelling by [14C]Leucine. Glutathione depletion, induced by in vivo administration of L-buthionine sulfoximine and diethylmaleate, did not alter the effect of cysteine, although it decreased the rate of protein synthesis by 32%. The effect of cysteine on protein synthesis does not seem to be related to a perturbation of the redox state of the NAD+/NADH system or to changes in the rate of gluconeogenic pathway. Determination of antimony in drinking waters by an inexpensive, reproducible hydride generator for atomic spectroscopy.

A method for determining antimony in drinking waters is described. In order to prevent a substantial error caused by the different oxidation states of antimony, Sb(V) is reduced to Sb(III) with potassium iodide-ascorbic acid. Covalent hydride is generated with a home made device by adding NaBH4. The hydride is then atomized in a flame-heated silica tube and atomic absorption is measured spectrophotometrically. The optimal conditions for this determination are discussed and interference effects are described. To preserve bioactive compounds while inactivating Bacillus subtilis spores in powdered Spirulina microalgae (Arthrospira platensis) with a non-thermal atmospheric plasma is the challenge presented in this paper. Hypochlorous acid (HOCl) is the most powerful oxidant produced by human neutrophils and contributes to the damage caused by these inflammatory cells. It is produced from H2O2 and chloride by the heme enzyme myeloperoxidase (MPO). Based on findings that betalains provide antioxidant and anti-inflammatory effects, we performed the present kinetic study on the interaction between the betalains, betanin and indicaxanthin, with the redox intermediates, compound I and compound II of MPO, and its major cytotoxic product HOCl. Reaction kinetics of betanin and its aglycone betanidin towards peroxyl radicals generated from the azo-initiated oxidation of methyl linoleate in methanol, and of a heterogeneous aqueous/soybean phosphatidylcholine liposomal system, were studied by monitoring formation of linoleic acid hydroperoxides and consumption of the pigments. Betanin was a weak retarder in methanol, and an effective chain breaking antioxidant in the liposomal model, indicating that kinetic solvent effects and partition in lipid bilayers may affect its activity. An insufficient and heterogeneously distributed nutritive blood flow leads to an inadequate and nonuniform supply of O2 and substrates in many solid tumors (Vaupel, 1977, 1979, 1982). This deterioration of the supply conditions, which already occurs in very early growth stages and which is superimposed by a deterioration of diffusive transport during advanced growth stages, is paralleled by a decrease in the therapeutic efficacy of various cancer treatment modalities such as irradiation and chemotherapy with antiproliferative drugs.
