Reply: 18F-FDG PET for Routine Posttreatment Surveillance in Oral and Oropharyngeal Squamous Cell Carcinoma

2010

1164 Objectives 68Ga-BPAMD (BPAMD = (4-{[bis-(phosphonomethyl))carbamoyl]methyl}-7,10-bis(carboxy methyl)-1,4,7,10-tetraazacyclododec-1-yl)acetic acid) was used in a first human in vivo study for diagnosis of osteoblastic bone metastases. The DOTA-based bisphosphonate ligand BPAMD may also be suitable for complexation with therapeutic radionuclides such as 177Lu. The same ligand thus may be used for diagnosis, dosimetry calculation, therapy and therapy control via PET/CT. Methods 177Lu was provided in 0.04 M HCl via the 176Yb(n,γ)177Yb→177Lu production process. Labelling was studied using different amounts of ligand in acetate buffer at pH 4-5 and different temperatures. For quality control…

Survivin is regulated by interleukin-4 in colon cancer stem cells

2010

Colorectal cancer has provided an important model to test the stem cell hypothesis of cancer origin, which implies that cancer arises as a result of genetic aberrations in stem cells leading to deregulation of the proliferation/differentiation balance. We and others have demonstrated that, similarly to other solid tumors, colon carcinogenesis and progression are dictated by highly apoptosis-resistant stem-like cells. Our data have suggested that protection from apoptosis is achieved by autocrine production of interleukin-4 (IL-4) through up-regulation of anti-apoptotic mediators. In this study, we extend our analysis to another apoptosis inhibitor widely expressed in tumors, namely survivin…

Demonstration of dihydro-orotate dehydrogenase inNeurospora crassa hyphae with a cytochemical procedure

1971

Dihydro-orotate dehydrogenase activity was demonstrated in the hyphae of the fungusNeurospora crassa by a cytochemical technique. The activity was slightly stronger in the hyphal tips thus demonstrating the more intense metabolic activity in these parts of the mycelium. Several control reactions showed that the staining reaction was specific.

Filipin labelling and intramembrane particles on the membranes of early and later autophagic vacuoles in Ehrlich ascites cells

1987

Cholesterol and intramembrane particle distribution on autophagic vacuole membranes was studied in Ehrlich ascites cells using filipin labelling and freeze-fracture electron microscopy. Unsaturated fatty acids were stained using imidazole-buffered osmium tetroxide. Autophagocytosis was induced with vinblastine, and early autophagic vacuoles were accumulated by lowering the ATP level in the cells with iodoacetate. Filipin labelling was observed in the limiting membranes of later, apparently hydrolase-containing autophagic vacuoles, whereas the most newly-formed, doublemembrane limited vacuoles were not labelled. The limiting membranes of late, residual body-type vacuoles either showed patchy…

Imaging standardisation in metastatic colorectal cancer: a joint EORTC-ESOI-ESGAR expert consensus recommendation

2022

Background: Treatment monitoring in metastatic colorectal cancer (mCRC) relies on imaging to evaluate the tumor burden. Response Evaluation Criteria in Solid Tumors (RECIST) provide a framework on reporting and interpretation of imaging findings yet offer no guidance on a standardized imaging protocol tailored to mCRC patients. Imaging protocol heterogeneity remains a challenge for the reproducibility of conventional imaging endpoints and is an obstacle for research on novel imaging endpoints. Patients and methods: Acknowledging the recently highlighted potential of radiomics and artificial intelligence (AI) tools as decision support for patient care in mCRC, a multidisciplinary, internatio…

Beneficial Read-Through of aUSH1CNonsense Mutation by Designed Aminoglycoside NB30 in the Retina

2010

PURPOSE. The human Usher syndrome (USH) is the most frequent cause of inherited combined deaf-blindness. USH is clinically and genetically heterogeneous, assigned to three clinical types. The most severe type is USH1, characterized by profound inner ear defects and retinitis pigmentosa. Thus far, no effective treatment for the ophthalmic component of USH exists. The p.R31X nonsense mutation in USH1C leads to a disease causing premature termination of gene translation. Here, we investigated the capability of the novel synthetic aminoglycoside NB30 for the translational read-through of the USH1C-p.R31X nonsense mutation as a retinal therapy option. METHODS. Read-through of p.R31X by three com…
