The study of calcium signaling in lipid gustatory cells in mice

The lipid-binding glycoprotein CD36, expressed by circumvallate papillae (CVP) of the mouse tongue, has been shown to be implicated in oro-gustatory perception of dietary lipids. We demonstrate that linoleic acid (LA) by activating sPLA2, cPLA2 and iPLA2 via CD36, produced arachidonic acid (AA) and lyso-phosphatidylcholine (Lyso-PC) which triggered Ca2+ influx in CD36-positive taste bud cells (TBC), purified from mouse CVP. LA induced the production of Ca2+ influx factor (CIF). CIF, AA and Lyso-PC exerted different actions on the opening of store-operated Ca2+ (SOC) channels, constituted of Orai proteins and regulated by STIM1, a sensor of Ca2+ depletion in the endoplasmic reticulum. Yarrowia lipolytica can grow on many hydrophobic substrates and metabolize them via the β-oxidation pathway or store them into lipid bodies. The first contact between the cell and the hydrophobic substrates is by the cell wall. In this step, cell-wall proteins (CWP), β-glucane and chitin of the cell wall could play an important role for the adhesion and uptake of hydrophobic substrates. Recently, several studies have shown an association between obesity and ECS overactivity. It is well admitted that polyunsaturated fatty acids are the most sensitive fatty acids to oxidative damage because of their high number of double bonds. This concept could not be confirmed by in vitro, animal, or human studies since unexpected or even contradictory results were obtained. The explanation of that is probably related to the methods evaluating lipid peroxidation whose relevance is not always established, but especially to movements of polyunsaturated fatty acids within lipoproteins that results in their protection. This short review aims to discuss the main results related to this topic.

International audience; Near-infrared (NIR) spectroscopy was evaluated as a rapid method for the determination of oleic, palmitic, linoleic and linolenic acids as well as omega-3, omega-6, and to predict polyunsaturated, monounsaturated and saturated fatty acids, together with triacylglycerides, diglycerides, free fatty acids and ergosterol in salmon oil.
