Uptake and Fate of Fluorescently Labeled DNA Nanostructures in Cellular Environments: A Cautionary Tale.

2019

[Image: see text] Fluorescent dye labeling of DNA oligonucleotides and nanostructures is one of the most used techniques to track their fate and cellular localization inside cells. Here, we report that intracellular fluorescence, and even FRET signals, cannot be correlated with the cellular uptake of intact DNA structures. Live cell imaging revealed high colocalization of cyanine-labeled DNA oligos and nanostructures with phosphorylated small-molecule cyanine dyes, one of the degradation products from these DNA compounds. Nuclease degradation of the strands outside and inside the cell results in a misleading intracellular fluorescent signal. The signal is saturated by the fluorescence of th…

Disorder-induced single-mode transmission.

2017

Localized states trap waves propagating in a disordered potential and play a crucial role in Anderson localization, which is the absence of diffusion due to disorder. Some localized states are barely coupled with neighbours because of differences in wavelength or small spatial overlap, thus preventing energy leakage to the surroundings. This is the same degree of isolation found in the homogeneous core of a single-mode optical fibre. Here we show that localized states of a disordered optical fibre are single mode: the transmission channels possess a high degree of resilience to perturbation and invariance with respect to the launch conditions. Our experimental approach allows identification…

The Increase in Maternal Expression ofaxin1andaxin2Contribute to the Zebrafish MutantIchabodVentralized Phenotype

2015

β-catenin is a central effector of the Wnt pathway and one of the players in Ca+-dependent cell-cell adhesion. While many wnts are present and expressed in vertebrates, only one β-catenin exists in the majority of the organisms. One intriguing exception is zebrafish that carries two genes for β-catenin. The maternal recessive mutation ichabod presents very low levels of β-catenin2 that in turn affects dorsal axis formation, suggesting that β-catenin1 is incapable to compensate for β-catenin2 loss and raising the question of whether these two β-catenins may have differential roles during early axis specification. Here we identify a specific antibody that can discriminate selectively for β-ca…

Genome-wide association analysis on normal hearing function identifies PCDH20 and SLC28A3 as candidates for hearing function and loss.

2015

Hearing loss and individual differences in normal hearing both have a substantial genetic basis. Although many new genes contributing to deafness have been identified, very little is known about genes/variants modulating the normal range of hearing ability. To fill this gap, we performed a two-stage meta-analysis on hearing thresholds (tested at 0.25, 0.5, 1, 2, 4, 8 kHz) and on pure-tone averages (low-, medium-and high-frequency thresholds grouped) in several isolated populations from Italy and Central Asia (total N = 2636). Here, we detected two genome-wide significant loci close to PCDH20 and SLC28A3 (top hits: rs78043697, P = 4.71E-10 and rs7032430, P = 2.39E-09, respectively). For both…

The glocalizing duality of contemporary sport

2010

AbstractThis paper assesses the current debate, in the sphere of the sociology of sport, on globalization and sport, arguing that the advance of world sport now, at the beginning of the twenty-first century, is better understood in terms of the phenomenon of glocalization, that is, as a process by which global demands and forces conform or adapt to local conditions (Robertson), contributing in this way to the production of ‘township’ and ‘community’ and to the emergence of national identities. The 1992 Barcelona Olympics, the 1993 European Football Championship and the 2002 Korea-Japan World Football Cup are presented as examples of the dual and paradoxical character of contemporary sport i…

Binge administration of 3,4-methylenedioxymethamphetamine ("ecstasy") impairs the survival of neural precursors in adult rat dentate gyrus.

2006

3,4-Methylenedioxymethamphetamine (MDMA) is a potent stimulant and hallucinogenic drug whose ability to regulate neurogenesis in the adult has not been previously investigated. We used 5'-bromo-2-deoxyuridine (BrdU) and Ki-67 as mitotic markers, and doublecortin (DCX) as a marker of immature neurons, to study proliferation, survival and maturation of adult-generated cells in the dentate gyrus (DG) of the hippocampus following binge administration of MDMA (8 injections of 5 mg/kg at 6 h intervals). The results showed that MDMA treatment did not affect cytogenesis in the DG, but significantly decreased the survival rate of cells incorporated after 2 weeks to the granular layer of the DG by ca…

Synthetic individual binaural audio delivery by pinna image processing

2014

Purpose – The purpose of this paper is to present a system for customized binaural audio delivery based on the extraction of relevant features from a 2-D representation of the listener’s pinna. Design/methodology/approach – The most significant pinna contours are extracted by means of multi-flash imaging, and they provide values for the parameters of a structural head-related transfer function (HRTF) model. The HRTF model spatializes a given sound file according to the listener’s head orientation, tracked by sensor-equipped headphones, with respect to the virtual sound source. Findings – A preliminary localization test shows that the model is able to statically render the elevation of a vi…

Spatial identification of the overionized plasma in W49B

2010

Recent Suzaku X-ray observations of the ejecta-dominated supernova remnant W49B have shown that in the global spectrum there is a clear indication for the presence of overionized plasma whose physical origin is still under debate. In order to ascertain the physical origin of such a rapidly cooling plasma, we focus on the study of its spatial localization within the X-ray emitting ejecta. We confirm the presence of a saw-edged excess (interpreted as a strong radiative recombination continuum) in the global spectrum above 8 keV, emerging above the ionization-equilibrium model. We produce a hardness ratio map to determine where the plasma is overionized and we perform a spectral analysis of th…

Instanton Counting, Quantum Geometry and Algebra

2020

The aim of this memoir for "Habilitation \`a Diriger des Recherches" is to present quantum geometric and algebraic aspects of supersymmetric gauge theory, which emerge from non-perturbative nature of the vacuum structure induced by instantons. We start with a brief summary of the equivariant localization of the instanton moduli space, and show how to obtain the instanton partition function and its generalization to quiver gauge theory and supergroup gauge theory in three ways: the equivariant index formula, the contour integral formula, and the combinatorial formula. We then explore the geometric description of $\mathcal{N} = 2$ gauge theory based on Seiberg-Witten geometry together with it…
