Genome-wide DNA profiling of marginal zone lymphomas identifies subtype-specific lesions with an impact on the clinical outcome

Abstract Marginal zone B-cell lymphomas (MZLs) have been divided into 3 distinct subtypes (extranodal MZLs of mucosa-associated lymphoid tissue [MALT] type, nodal MZLs, and splenic MZLs). Nevertheless, the relationship between the subtypes is still unclear. We performed a comprehensive analysis of genomic DNA copy number changes in a very large series of MZL cases with the aim of addressing this question. Splenic marginal zone lymphoma: A prognostic model for clinical use

The Integruppo Italiano Linfomi (IIL) carried out a study to assess the outcomes of splenic marginal zone lymphoma and to identify prognostic factors in 309 patients. The 5-year cause-specific survival (CSS) rate was 76%. In univariate analysis, the parameters predictive of shorter CSS were hemoglobin levels below 12 g/dL (P < .001), albumin levels below 3.5 g/dL (P = .001), International Prognostic Index (IPI) scores of 2 to 3 (P < .001), lactate dehydrogenase (LDH) levels above normal (P < .001), age older than 60 years (P = .01), platelet counts below 100 000/μL (P = .04), HbsAg-positivity (P = .01), and no splenectomy at diagnosis (P = .006). Sjogren syndrome and parotid marginal zone lymphoma: Report of a case and review of literature

Five percent of the cancers in the parotid region are non-hodgkin lymphomas. This lymphoma, involving the mucosa-associated lymphoid tissue (MALT), is called MALToma. A chronic autoimmune inflammatory process of the parotid gland, such as Sjogren's syndrome, results in an increased incidence of parotid MALToma. Biopsy of the parotid is essential for appropriate diagnosis; this can then be completed by radiotherapy, chemotherapy or therapy with monoclonal antibodies according to the stage of the disease. This study shows a clinical case of a 50-year-old patient with Sjogren's syndrome who developed a parotid MALToma.

A genome-wide association study of marginal zone lymphoma shows association to the HLA region

Marginal zone lymphoma (MZL) is the third most common subtype of B-cell non-Hodgkin lymphoma. Here we perform a two-stage GWAS of 1,281 MZL cases and 7,127 controls of European ancestry and identify two independent loci near BTNL2 (rs9461741, P=3.95 × 10−15) and HLA-B (rs2922994, P=2.43 × 10−9) in the HLA region significantly associated with MZL risk. This is the first evidence that genetic variation in the major histocompatibility complex influences MZL susceptibility.

The purpose of the present study was to estimate the standardised incidence ratio (SIR) and the absolute excess risk (AER) of SPC in patients with splenic marginal zone lymphoma (SMZL). The Assessment of Second Primary Cancers (SPCs) in a Series of Splenic Marginal Zone Lymphoma (SMZL) Patients

The purpose of this study is to estimate the risk of second primary cancer (SPC) in 129 consecutive patients with splenic marginal zone lymphoma (SMZL) diagnosed in three Italian haematological centres. The person-years method deriving as a sum of products of age- and sex- specific rates and of the corresponding time at risk was used. The SPC Standardized Incidence Ratio (SIR) was 2.03 with a 95% confidence interval: [1.05, 3.56] (p < 0.05) and the corresponding Absolute Excess Risk (AER) was 145.8 (per 10000 SMZL patients per year). Abstract Splenic marginal zone lymphoma (SMZL) is an indolent B cell malignancy whose diagnosis is based on lymphocyte morphology, immunophenotype and marrow and/or splenic histology. Unlike other lymphomas, there is not a common chromosomal translocation specific for SMZL, and genetic prognostic factors are poorly defined. To investigate the pattern of genomic aberrations in SMZL, we applied comparative genomic hybridization to BAC microarrays (array CGH) to a well characterized series of 75 SMZL specimens.
