Zespół słabości - wytyczne diagnostyki, terapii i prewencji

Frailty syndrome is a state of decreased homeostatic capacity against stress. It is an independent risk factor for subsequent mortality, institutionalization, multimorbidity, falls, incontinence, and immobility. Frailty people have a significantly reduced quality of life, as well as being the largest users of health care services. Proven direct link between the increase in the number of people with the frailty syndrome and rising costs in the health and social care in spite of clearly progressive aging process and potential reversibility of frailty, justifies the use of targeted interventions aimed at early diagnosis of frailty and to identify risk factors. General practitioners play a key…
Prostaglandin E2 and prostaglandin F2 alpha as endometrial receptivity biomarkers in successful embryo implantation
2014
Failure in the adhesion of the human blastocyst to the endometrium has been described as an important cause of infertility. Establishing the period of the so-called window of implantation and understanding the molecular mechanisms associated with embryo implantation has clinical and scientific implications. While over the last decades histological evaluation has been used to determine the phase of the menstrual cycle of the endometrium, the poor information obtained has made the case of using new technologies to identify specific markers, understand and characterize the receptive stage. This doctoral thesis investigates the existence, function and clinical impact of two specific lipids, the…
Metabolomics: An Emerging Approach to Understand Pathogenesis and to Assess Diagnosis and Response to Treatment in Spondyloarthritis
2022
Spondyloarthritis (SpA) is a group of rheumatic diseases whose pathogenesis relies on a complex interplay between genetic and environmental factors. Over the last several years, the importance of the alteration of the gut microbiota, known as dysbiosis, and the interaction of bacterial products with host immunity have been highlighted as intriguing key players in SpA development. The recent advent of the so called “-omics” sciences, that include metabolomics, opened the way to a new approach to SpA through a deeper characterisation of the pathogenetic mechanisms behind the disease. In addition, metabolomics can reveal potential new biomarkers to diagnose and monitor SpA patients. The aim of…
Rationale and design of the DARWIN-T2D (DApagliflozin Real World evIdeNce in Type 2 Diabetes): A multicenter retrospective nationwide Italian study a…
2017
Background Randomized controlled trials (RCTs) in the field of diabetes have limitations inherent to the fact that design, setting, and patient characteristics may be poorly transferrable to clinical practice. Thus, evidence from studies using routinely accumulated clinical data are increasingly valued. Aims We herein describe rationale and design of the DARWIN-T2D (DApagliflozin Real World evIdeNce in Type 2 Diabetes), a multicenter retrospective nationwide study conducted at 50 specialist outpatient clinics in Italy and promoted by the Italian Diabetes Society. Data synthesis The primary objective of the study is to describe the baseline clinical characteristics (particularly HbA1c) of pa…
Resistance Training Induces Antiatherogenic Effects on Metabolomic Pathways
2019
Introduction: Arising evidence suggests that resistance training has the potential to induce beneficial modulation of biomarker profile. To date, however, only immediate responses to resistance training have been investigated using high-throughput metabolomics whereas the effects of chronic resistance training on biomarker profile have not been studied in detail. Methods: A total of 86 recreationally active healthy men without previous systematic resistance training background were allocated into i) a resistance training (RT) group (n=68, age 33 ± 7 years, body mass index (BMI) 28 ± 3 kg/m2 ) and ii) a non-RT group (n=18, age 31 ± 4 years, BMI 27 ± 3 kg/m2 ). Blood samples were collected at…
Characterization of the metabolic profile and identification of potential therapeutic targets in advanced prostate cancer patients
2023
Photoinduced lethal and sublethal toxicity of retene, a polycyclic aromatic hydrocarbon derived from resin acid, to coregonid larvae
2004
A comparative investigation on the acute phototoxicity of retene to vendace (Coregonus albula) and whitefish (C. lavaretus), both having pelagial larvae in spring, was conducted. To test the concept of early warning of sublethal biomarkers in relation to lethality to posthatch stages, we examined the effects of ultraviolet-B (UV-B) and retene on the levels of cytochrome P4501A (CYP1A) and heat shock protein 70 (HSP70) by exposing the animals to elevated levels of these factors for 48 and 72 h, respectively. Whereas UV-B and retene on their own were not lethal, simultaneous retene and UV-B exposure caused very high mortality to both species. The median lethal concentration (LC50; i.e., the c…
Novel VAMPIRE algorithms for quantitative analysis of the retinal vasculature
2013
This paper summarizes three recent, novel algorithms developed within VAMPIRE, namely optic disc and macula detection, arteryvein classification, and enhancement of binary vessel masks, and their performance assessment. VAMPIRE is an international collaboration growing a suite of software tools to allow efficient quantification of morphological properties of the retinal vasculature in large collections of fundus camera images. VAMPIRE measurements are currently mostly used in biomarker research, i.e., investigating associations between the morphology of the retinal vasculature and a number of clinical and cognitive conditions.

Cholesterol homeostasis and retinal stress in anterior ischemic optic neuropathy
2016
Purpose: Anterior ischemic optic neuropathy (AION) is characterized by acute stress of retinal ganglion cells (RGC). Cholesterol-24S-hydroxylase (CYP46A1) is specifically expressed in RGC where it may exert neuroprotective properties by preventing from cholesterol accumulation via catalyzing the formation of 24S-hydroxycholesterol. Previous findings hypothesize the housekeeping importance of CYP46A1 in the maintenance of cholesterol in the retina. In order to better understand the role of CYP46A1 in the retina, we aimed to define whether CYP46A1 expression and 24S-hydroxycholesterol levels were modified during RGC stress in AION.Methods: Ninety nine male Sprague Dawley rats were randomized …
